    Notes: Abstract  The effect of salinity on growth, starvation-survival and recovery from salt stress of a Rhizobium sp. strain isolated from nodules of Acacia tortilis from a soil of Senegal was studied. Growth parameters of C-limited continuous cultures, grown in the presence and the absence of 342 mM NaCl, decreased in the saline medium and with increasing dilution rates. The survival capacity of starved cultures depended on the previous growth conditions: culturability of cells grown with salt was inversely related to growth rate, while culturability increased with increasing dilution rate for cultures grown without salt. Culturability of the cultures subjected to the double stress of starvation and salinity was reduced and a high percentage of cells entered the viable but nonculturable state. All the starved cultures were capable of regrowth when nutrients became available, thus showing that this strain can withstand long periods of nutrient deprivation in soil while maintaining the capacity for an active metabolism and a potential infectiousness toward an appropriate host.

    Notes: Summary Chemical, ultrastructural, and immunoadjuvant properties of Candida albicans (CA) and of a number of its fractions have been characterized through the analysis of the antitumor activity of soluble and insoluble cell wall components. CD2F1 mice were inoculated IP with Moloney virus-induced lymphoma LSTRA and treated with bis-1-chloroethyl-nitrosurea (BCNU) on day +5 after tumor challenge. A significant increase of the antitumor efficacy of BCNU treatment was found in mice inoculated with CA as immunoadjuvant on days −14 and +1 (−14/+1 schedule) with respect to tumor challenge. However, no significant difference in survival time was found between mice treated with BCNU alone and those treated with BCNU plus either soluble mannan or glucan-protein fractions extracted from CA and administered according to −14/+1 or −7/+1 schedules. On the other hand, mice treated with BCNU plus the insoluble glucan fraction (wall ‘ghosts’) given on days −14/+1 or even on day −7 only (i.e., without boosting after tumor challenge) survived longer than animals treated with BCNU alone. The immunoadjuvant effect of CA and of other ‘classic’ immunoadjuvants, such as BCG and Corynebacterium parvum, was completely abolished by total-body irradiation (400 R) given 5 h before the first administration of the agent on day −7 prior to tumor challenge. These results indicate that: (a) the minimal structure required for the expression of the immunoadjuvant effect of CA is the insoluble, β-glucan component of the cell wall; (b) the soluble components of CA cell wall (i.e., mannan and glucan-protein) per se do not show any detectable immunoadjuvant effect in the present animal-tumor system; they may, however, ‘modulate’ this effect, as shown by the fact that whole CA, but not the insoluble β-glucan, needs a boosting injection for the expression of its immunoadjuvant properties; (c) the immunoadjuvanticity of CA is radiosensitive.

    Notes: In previous studies, the white-rot basidiomycete Lentinus edodes, strain SC-495, was proved to be a “selective” lignin degrader and its extracellular crude preparations arising from solid-state cultures were successfully employed in biopulping experiments on annual plants. This fungus produced extracellular laccase as the predominant phenoloxidase when growing in solid-state fermentation on corn stalks. Laccase from this strain was purified and partially characterized, as an initial approach towards the study of its ligninolytic complex. Laccase was purified 69.6-fold by anion-exchange chromatography and two affinity-chromatography steps with an overall yield of 7.45%. The native enzyme exhibited a molecular mass of 74 kDa, an isoelectric point of 3.42 and a carbohydrate content of 7.5%. The absorption spectrum of laccase showed a maximum at 605 nm, typical of blue-copper oxidases. The optimum pH and temperature for the activity of laccase were 4.0-4.2 and 50°C, respectively. Kinetic experiments, performed with a wide range of phenolic compounds, showed that the reaction rate and the substrate affinity greatly varied depending on the nature of substituents and their reciprocal positions on the aromatic ring. In particular, the enzyme showed high affinity to phenolic compounds bearing methoxyl or methyl groups, but no affinity to those bearing the nitro group directly attached to the benzene ring, nor to non-phenolic lignin-related compounds, such as trans-cinnamic acid or 3,4-dimethoxycinnamic acid. The huge differences in terms of reactivity of the enzyme towards phenolic compounds suggests that a preliminary systematic screening should be advisable when using laccase in effluent treatment applications.

    Description: Global Positioning System (CGPS) data from Mt. Etna between May 2015 and September 2016 show intense inflation and a concurrent Slow Slip Event (SSE) from 11 December 2015 to 17 May 2016. In May 2016, an eruptive phase started from the summit craters, temporarily stopping the ongoing inflation. The CGPS data presented here give us the opportunity to determine 1) the source of the inflating body; 2) the strain rate parameters highlighting shear strain rate accumulating along NE Rift and S Rift; 3) the magnitude of the SSE; and 4) possible interaction between modelled sources and other flank structures through stress calculations. By analytical inversion, we find an inflating source 5.5 km under the summit (4.4 km b.s.l) and flank slip in a fragmented shallow structure accommodating displacements equivalent to a magnitude M w 6.1 earthquake. These large displacements reflect a complex mechanism of rotations indicated by the inversion of CGPS data for strain rate parameters. At the scale of the volcano, these processes can be considered precursors of seismic activity in the eastern flank of the volcano, but concentrated mainly on the northern boundary of the mobile eastern flank along the Pernicana Fault and in the area of the Timpe Fault System.
