    Notes: This study examines the effect of the steroid analogue, RU486, on the physiological responses of fed and chronically fasted rainbow trout to an acute handling stressor. This potent ligand of the glucocorticoid receptor was administered as a slow-release implant either alone, or in combination with cortisol. There were temporal changes in plasma cortisol concentrations following administration of cortisol implants in both fed and fasted trout. By day 14, plasma cortisol levels in fed fish were similar in all treatment groups, but in fasted fish, the effect of cortisol administration on plasma cortisol concentrations was still evident; RU486 administered with cortisol, did not affect this response. Cortisol administration also elicited a small, but significant increase in plasma GH concentrations in fed rainbow trout and in plasma glucose concentrations in fasted animals. RU486-treatment prevented these responses. Conversely, whereas RU486 alone had no effect on hepatic 5′-monodeiodinase activity, when administered with cortisol it enhanced the marked suppressive effect of cortisol evident in both fed and fasted groups, suggesting that it may exert an interactive effect with cortisol on this process. Stressor-related changes in plasma cortisol, glucose, GH and thyroid hormone concentrations were evident in both fed and fasted groups; however, there was no evidence of a suppressive effect of RU486 treatment on any of the measured plasma parameters. Although RU486 did not prevent the stressor-related changes, the post-stressor cortisol profiles in RU-treated trout were extremely erratic compared with the oil-treated controls. This implies a disturbance of the normal interactions of the components of the hypothalamus-pituitary-interrenal tissue axis.

    Notes: Rainbow trout, Oncorhynchus mykiss (Walbaum), with a mean initial weight of 196 g were fed the β-agonist ractopamine incorporated into a commercial rainbow trout diet at 0, 5, 10, 20 or 40 p.p.m. for 12 weeks. The experiment was conducted as a completely randomized design, with five treatments and four replicate tanks. At the start of the trial and every 4 weeks subsequently, fish were euthanized, and blood serum and liver samples were collected. Serum glucose concentrations were significantly higher in fish fed 40 p.p.m. ractopamine for 8 weeks, while all treated groups demonstrated significantly elevated glucose levels relative to controls at the 12 weeks sampling interval. Nonesterified fatty acid (NEFA) levels were significantly higher in fish fed 5 and 20 p.p.m. ractopamine for 4 weeks, but those fed 10 p.p.m. displayed depressed NEFA concentrations compared to controls. At 4 weeks, plasma growth hormone (GH) levels were significantly lower (P 〈 0.05) in the 5, 20 and 40 p.p.m. groups compared to controls. The results suggest that dietary administration of ractopamine to rainbow trout modestly alters the concentrations of circulating GH, glucose and NEFA.

    Notes: Physiological responses of rainbow trout, Salmo gairdneri Richardson, to experimental infection with the blood haemoflagellate Cryptobia salmositica were monitored for 7 weeks to determine whether the parasitaemia elicited a classical stress-response as evidenced by elevated plasma cortisol and glucose levels. There was a progressive anaemia, hepatomegaly, splenomegaly, and depressed plasma T3, T4, protein and glucose concentration, and lowered liver glycogen content in the parasitized fish throughout the study, which clearly indicated that the animals were under physiological duress. Moreover, these hormonal and metabolic indicators showed no indication of recovery with the decline in blood parasite numbers. However, there were no differences in plasma cortisol or glucose levels between infected and control fish throughout the study, suggesting that the parasitaemia did not activate the pituitary-interrenal axis.

    Notes: Seasonal changes in serum L-thyroxine (T4) and triiodo-L-thyronine (T3) concentrations were measured in a feral population of brown bullheads, Ictalurus nebulosus. Serum T4 was elevated in mid April concomitant with the onset of gonadal recrudescence and the increase in ambient temperatures from the winter level. Thereafter, serum T4 levels were constant throughout the late spring and summer (except for a significant lowering in levels associated with the spawning and early postspawning period), declining to a low overwintering level by mid December. Serum T3 levels increased in April, and apart from a significant decrease in July associated with the postspawning period, rose progressively until August, after which levels declined progressively to reach overwintering levels again by mid December. Between mid April and mid May serum T4/T3 ratios fell from the high values found in fish during the winter months. Apart from variations associated with the fluctuations of T4 and T3 concentrations in the spawning and early postspawning periods, serum T4/T3 ratios remained at a low level throughout the summer and early autumn months, and rose again in October to the overwintering levels. The circannual cycles are discussed in relation to the possible role of the thyroid hormone in gonadal maturation and metabolism in this species.

    Notes: The hepatosomatic index (HS1) of female sexually mature coho salmon, Oncorhynchus kisutch, from Lake Superior was significantly higher (P〈0.01) than in males, whereas that of females from Lake Erie was significantly lower (P〈0.05) than in males; there was no significant sex differences in coho salmon from Lakes Ontario or Michigan. Hepatic nuclear areas (HNA) were significantly higher (P〈0.01) in males compared with females in fish collected from Lakes Ontario and Erie, but there was no sex differences in HNA of coho salmon from Lakes Superior or Michigan. In all four lakes, hepatocyte cytoplasmic volume was significantly higher (P〈0.01) in males than in females from the same lake, and the cytoplasmic volume: nuclear volume ratio of males was corresponding significantly higher (P〈 0.05) than in females from the same lake.Two hepatocytic cell types were evident, termed' light and dark cells, based on the electron density of their cytoplasmic matrix. There were considerable intersex and interlake differences in the relative populations of light ‘and dark’ cells in coho salmon from Lakes Ontario, Michigan, Erie and Superior, and also in the hepatocytic content of lipid, glycogen, mitochondria and ribosomal and smooth endoplasmic reticulum.There were no sex differences with regard to in vitro hepatic monodeiodination of L-thyroxine (T4) to triiodo-L-thyronine (T3) in salmon from any of the lakes. However, T4 to T3 conversion was significantly lower (P〈 0.05) in liver homogenates prepared from Lake Ontario coho salmon than in comparable preparations of liver from Lake Superior, Lake Erie, or Lake Michigan salmon.

    Notes: A progressive fall in serum thyroid hormone levels was found in Lake Ontario coho salmon, Oncorhynchus kisutch, between May 1978 and January 1979 concomitant with an increase in goitre frequency and thyroid hyperplasia. Despite marked differences in overt goitre frequency in sexually mature adult salmon collected from Lakes Ontario (43.7%), Erie (93.5%), or Michigan (0%), serum thyroid hormone levels and thyroid histology were similar in fish from the three lakes. Two gonadotrop-like basophil cells (‘globular’and‘vacuolar’) were found in the pituitary, particularly in sexually maturing and mature adults and‘jacks’. Whereas in salmon from Lakes Ontario or Michigan, the globular cells were vacuolated in Lake Erie salmon they were not; this observation may be correlated with the absence of obvious secondary sexual characteristics in Lake Erie salmon. Serum free fatty acids serum, triglycerides and cholesterol concentrations in Lake Ontario salmon fell progressively from the condition in fish collected from the Lake in May 1978, to that of fish collected during their potamodrous migration in November 1978 and January 1979.

    Notes: The three dimensional morphology of the thyroid tissue in juvenile rainbow trout Oncorhynchus mykiss was determined using serial transverse and longitudinal sections of lower jaws stained with haematoxylin and eosin. The thyroid tissue comprised interconnecting tubular units not the follicles that are widely described in the literature. Some of these tubules were subdivided by branching, giving the appearance of follicles in the sections. The thyroid tissue was aggregated into two major concentrations of tissue, associated with the first and second branchial arteries, with the majority of the tissue located dorsal and lateral to the aorta. These observations in juvenile rainbow trout suggest that thyroid tissue of some species of fishes may not be follicular, as previously assumed. The physiological and evolutionary implications have yet to be determined.

    Notes: Testicular and ovarian fragments of the protogynous Pacific wrasse Haliochoeres trimaculatus were incubated in vitro with [3H]pregnenolone ([3H]P5), [3H]17-hydroxyprogesterone ([3H]17OHP4), non-radioactive (nr) 17β-oestradiol (nrE2) or nrP5 to identify the major gonadal steroidogenic pathways and steroid products in females and in the two male variants of this species, the terminal phase (TP) and initial phase (IP) males. Both testis and ovarian tissues exhibited 7 hydroxylase activity resulting in the formation of 7α-hydroxypregnenolone (7OHP5) from [3H]P5, and many HPLC peaks were identified as products of testicular (c. 29) and ovarian (c. 23) steroidogenesis, and only c. 50% of these metabolites co-eluted with authentic reference standards; only very small amounts of conjugated steroid were synthesized from any of the precursors. [3H]P5 was converted by testis mainly to 7αOHP5, and two unknown steroids, whereas [3H]17OHP4 metabolism gave rise to [3H]17,20β-dihydroxy-4-pregnen-3-one (DHP), 11-ketotestosterone (11KT), and two unknown steroids. For ovarian tissues, [3H]17OHP4 and [3H]P5 were metabolized to form E2, oestrone (E1), androstenedione (A4), 20α- and 20β-dihydroprogesterone (20αDHP and 20βDHP), 7αOHP5 (from [3H]P5) and a major unknown. The HPLC steroid profiles for testis incubations for IP and TP males were similar, however, the steroidogenic response of the testis of TP males to human chorionic gonadotrophin, in vitro (determined by hormone assay), was significantly higher than that of IP males.

    Notes: Plasma thyroid hormone levels were measured in pink salmon, Oncorhynchus gorbuscha, during their spawning migration in the Fraser River, British Columbia. The plasma levels of both l-thyroxine (T4) and triiodo-l-thyronine (T3) were significantly higher in males than in females. In both sexes the hormone levels were maintained, or increased somewhat, during the early stages of migration, but fell thereafter. In females the plasma T4 and T3 levels of salmon collected on the spawning grounds were at or below detectable levels of the assays. The changes in thyroid hormone levels are correlated with changes in plasma insulin, gonadotropin, gonadal steroid hormones, cortisol and vitellogenin levels measured in the same specimens.
