    Notes: Summary We have studied the pharmacokinetics of flecainide after the oral administration of 100 mg to 8 patients without renal impairment and 8 patients with mild to moderate renal failure. Both groups gave comparable results with respect to the peak plasma concentrations and the time to peak. There was a significant correlation between renal flecainide clearance and endogenous creatinine clearance. The elimination half-time in the patients with impaired renal function was significantly longer (19.9, SD 9.9 h) than that in the patients with normal renal function (11.5, SD 4.2 h), but the variability in the elimination half-time in renal failure could not be explained on the basis of the available results.

    Description: Inflammation is a key process in various diseases, characterized by leukocyte recruitment to the inflammatory site. This study investigates the role of a disintegrin and a metalloproteinase (ADAM) 10 and ADAM17 for leukocyte migration in vitro and in a murine model of acute pulmonary inflammation. Inhibition experiments or RNA knockdown indicated that monocytic THP-1 cells and primary human neutrophils require ADAM10 but not ADAM17 for efficient chemokine-induced cell migration. Signaling and adhesion events that are linked to cell migration such as p38 and GTPase-family activation, F-actin polymerization, adhesion to fibronectin, and up-regulation of α 5 integrin were also dependent on ADAM10 but not ADAM17. This was confirmed with leukocytes isolated from mice lacking either ADAM10 or ADAM17 in all hematopoietic cells (vav 1 guanine nucleotide exchange factor [Vav] -Adam10 –/– or Vav- Adam17 –/– mice). In lipopolysaccharide-induced acute pulmonary inflammation, alveolar recruitment of neutrophils and monocytes was transiently increased in Vav- Adam17 –/– but steadily reduced in Vav- Adam10 –/– mice. This deficit in alveolar leukocyte recruitment was also observed in LysM- Adam10 –/– mice lacking ADAM10 in myeloid cells and correlated with protection against edema formation. Thus, with regard to leukocyte migration, leukocyte-expressed ADAM10 but not ADAM17 displays proinflammatory activities and may therefore serve as a target to limit inflammatory cell recruitment.

    Description: Digital twins are fine-grained digital replicas of physical objects and systems which have been widely applied in the engineering realm for tasks such as engine optimization and port management. Recently, digital twins have also caught the attention of marine scientists as a solution to socio-environmental problems. By bundling and providing access to observational data, models, and simulations, digital twins allow users to explore current and future ‘what-if’ scenarios, especially related to human interactions with the ocean. High-value application areas include fisheries and mariculture, marine protected areas, ocean-based tourism, ecological forecasting, nature-based solutions, marine infrastructure development, and the interactions between all of these with an ever-growing collection of data streams.In the context of the UN Decade of Ocean Science for Sustainable Development, we launched the international programme “DITTO - The Digital Twins of the Ocean“ (ditto-oceandecade.org) in May 2022. DITTO’s mission is to develop and share a common understanding of Digital Twins of the Ocean, to establish best practice examples and to advance a digital framework to empower ocean professionals to effectively create their own application-focused digital twin. This powerful framework will enable users to visualize and explore ocean knowledge and empower ocean professionals and models, forecasts, citizen scientists, policymakers, and the general public alike.A successful regional implementation of a Digital Twin depends on four aspects: A) a capable ocean observing system B) An innovative ocean modeling and prediction capability C) An interoperable, shared and easily accessible data environment. D) An interactive user interface. International cooperation will be key for success.
