    Notes: Dielectrically apertured vertical-cavity surface-emitting lasers are modeled using a Green's-function-based optical solver that allows calculation of the full-vector lasing modes and their threshold conditions. The laser is separated into subsystems consisting of a planar microcavity, a dielectric aperture, and an active gain medium. The exact Green's function solution for a radiating point source in the planar microcavity is then used to construct an eigenvalue equation for the self-consistent lasing modes. The derived eigenvalue equation is numerically solved to evaluate threshold dependence on aperture and cavity design. Results show a low threshold for thin oxide apertures placed at field antinodes, as well as for tapered oxides with thin tapers placed at field nodes. © 1998 American Institute of Physics.

    Notes: Phorbol myristate acetate (PMA) is a potent inducer of T colony formation by peripheral blood lymphocytes. A mean cloning efficiency of 0.3% (0.05-0.5%) is obtained with PMA concentrations of 100-1000 ng/ml. PMA-induced T colony formation does not require the presence of monocytes and therefore differs from other mitogens in this respect. Purified T-colony-promoting activity (TCPA) (devoid of phytohaemagglutinin (PHA)) increases PMA-induced T colony numbers and induces T colony formation at low PMA doses (0.01 to 1 ng), concentrations at which no T colonies are detected in the absence of added TCPA. PMA-induced colonies are mainly composed of cells bearing Fc receptors for IgM (54%), which is not the case for colonies obtained with PHA (11 %). PMA-induced colony cells do not bind OKT3 and OKT4 monoclonal antibodies, whereas 23% are able to bind OK-T8 antibody. These results demonstrate that PMA is a potent inducer of T colony formation and may therefore serve as a useful tool for the study of T-cell differentiation.

    Notes: X-irradiated (250 rad) or normal A mice injected with syngeneic concanavalin A-induced lymphoblasts (syn-Con A blasts) developed an inflammatory response in their footpads 24 to 72 h after injection of syngeneic lipopolysaccharide induced lymphoblasts (syn-LPS blasts) into these tissues. This immunological activity was designated syngeneic delayed type hypersensitivity (syn-DTH), because T cells transferred the response to naive recipient. Analysis on Ultrogel or Sephedex G-50 columns revealed that a Con A-blast extract contains two syn-DTH-stimulating antigens: a small antigen (6000–7000) and a large antigen (apparent MW of 160,000–175,000). This conclusion held true even when protease inhibitors were included in the fractionation procedure. The approximate molecular weights of these antigens estimated by that gel filtrations were confirmed by sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). The large lymphoblast syn-DTH-stimulating antigen contains carbohydrate residues but not products of the H-2 genetic region. The small antigen does not contain sugar moieties, but it expresses affinity to anti-H-2Dd monoclonal antibody. The immune response to the small antigen but not to the large antigen is genetically restricted at body the induction and the elicitation in phases of the DTH. A strain of mice immunized with the small antigen generated syn-DTH after challenge with lymphoblasts of B10.T (6R) mice, which share the H-2Dd subregion with A mice but not the H-2K or the H-2I subregions. Fast protein liquid chromatography of the small antigen yielded a purified material which appeared as a single band after Coomassie staining of its gel electrophoresis.

    Notes: Suppressor cells obtained from spleens of normal A mice, or factor extracted from these suppressor cells, abolished the syngeneic delayed-type hypersensitivity (syn-DTH) response of X-irradiated A mice injected with trinitrophenylated spleen cells und challenged with syngeneic lymphoblasts. Some of the physical, chemical and biological properties of the suppressive factor (SF) were characterized. The SF was relatively temperature-stable and its activity was destroyed by pronase (but not with RNase or DNase). The activity of the SF was absorbed on concanavalin A and anti-I-Jk Scpharose columns, suggesting that the factor is a glycoprotein-bearing I-Jk product. The approximate molecular weight of the factor is 50,000–60,000. The SF was absorbed on plastic adherent cells (but not on non-adherent cells). Adherent cells that absorbed the SF abrogated the ability of primed T cells to transfer the syn-DTH to naive X-irradiated recipients. In contrast, SF that was presented directly to the primed T cells failed to abolish their ability to transfer DTH. These findings suggest that the adherent cells serve as mediators, transferring the SF from factor-producing cells (Lyt-1+2+3+, I-Jk+ T cells) to target cells (Lyt-1+ primed T cells).

    Notes: : Twenty-eight untrained panelists used the signal detection rating method to determine thresholds for daidzein and genistein. Five concentrations of genistein and daidzein were suspended in starch solutions. The starch solution represented the noise, and isoflavone suspensions represented the signal. Blindfolded panelists received 15 replicates of noise and signal. To determine group detection thresholds, the concentration at an R-index of 75% was calculated. The group thresholds for genistein and daidzein were 4.006 × 10−3 M and 2.921 × 10−3 M, respectively. These results indicate a much higher threshold value than reported in the literature and in soymilks. This strongly suggests that daidzein and genistein individually do not contribute to bitterness or astringency in soy food products.

    Notes: Knowledge of relationships between kernel composition and sensory attributes of sweet corn could be helpful in selection of corn genotypes for the food industry. Frozen samples of four shrunken2 sweet corn inbred lines and two commercial hybrids were assayed for dimethyl sulfide (DMS), phytoglycogen and sugar concentrations. Sensory evaluation of the same samples by a trained descriptive panel was conducted to determine intensity of attributes of the three main components of sweet corn quality (aroma, flavor and texture). The relationships between sensory attributes, overall liking and kernel characteristics were determined. Sweetness was strongly correlated with sweet corn flavor, total sugars and sucrose. Grassy flavor and aroma were logarithmically correlated with DMS. Volatiles present in the inbred lines, but not in the commercial samples, could also contribute to the grassy flavor. Sweetness, starchy flavor, crispness and tenderness were the key sensory attributes correlated with overall liking. Selection for tender genotypes with high sugar concentration would increase the eating quality of the product.
