    Notizen: Gangliosides were identified as constituents of the lipid fraction from egg yolk. Approximately 0.07 mμmole of ganglioside sialic acid was recovered per gram wet weight of yolk. Thin-layer chromatographic separation revealed the presence of at least eight separate ganglioside species in this fraction. Glucose, galactose, hexosamine and sialic acid, in the molar proportions of 1.00 to 1.25 to 1.95 to 1.19, were the major carbohydrates of yolk ganglioside fractions. Fourteen fatty acids were observed in the ganglioside fraction; 16:0 (26.6%) 18:0 (9.2%), 18:l (22.9%) and 22:0 (28.3%) were predominant.

    Notizen: Summary Rat liver contains a particulate (membrane-bound) glycosyl transferase, concentrated in Golgi apparatus fractions, which catalyzes the synthesis of a trisialoganglioside from the ganglioside precursor disialohematoside (GD3). Sialic acid was not incorporated into exogenous GD1a from CMP-N-acetylneuraminic acid suggesting that GD1a is an endproduct of the monosialoganglioside pathway. Thus, the disialoganglioside pathway may be a primary source of trisialoganglioside and higher ganglioside homologs in adult rat liver.

    Notizen: Summary Milk fat globule membrane was solubilized with sodium dodecyl sulfate and mercaptoethanol and the membrane proteins were separated by SDS-polyacrylamide gel electrophoresis. The membrane preparations contained three major size classes of polypeptide of 155,000, 62,500 and 43,500 daltons. At least five glycopeptides were separated of which two stained intensely with periodic acid-Schiff reagent, but poorly with coomassie blue. Trypsin hydrolysis of whole cream and isolated milk fat globule membrane revealed major differences in the rates of protein hydrolysis. Many of the membrane proteins of whole cream resisted proteolysis compared with the same proteins in the isolated membrane. Two glycopeptides were resistant to trypsin digestion in either preparation. Treatment of whole cream with neuraminidase led to the release of at least 70% of the protein-bound sialic acid. Whole cream and isolated membrane samples were iodinated with125I in the presence of lactoperoxidase and hydrogen peroxide. The membrane proteins were significantly more accessible to lactoperoxidase-125I in isolated membrane compared with the proteins of whole cream. Polypeptides of molecular weight 43,500 and approximately 48,000 daltons were predominantly labelled in whole cream and could be eluted from the fat globules with magnesium chloride (1.5m). The results strongly suggest that the proteins of milk fat globule membrane are asymmetrically arranged in the membrane and that most of the protein-bound sialic acid is present on the external surface of milk fat globules.

    Notizen: Summary Quantitative ultrastructural analysis of mid-lactation rat mammary gland demonstrated that cytoplasmic microtubules were present in nearly all secretory epithelial cells examined. Most microtubules were oriented perpendicular to the apical membrane and were found in the apical and medial portions of the cell cytoplasm. There was no statistical difference between the number of microtubules associated with vesicles and the number that were not. Most vesicles which were in contact with microtubules were small (50 to 150 nm), appeared electron lucent and were located in a supra-Golgi complex position. Many of these vesicles were seen to be aligned along the axis of longitudinally sectioned microtubules oriented perpendicular to the apical plasma membrane. As measured by a colchicine binding assay, the total tubulin content of mammary tissue from mid-lactation rats was about 107 μg/100 mg wet weight. Approximately 19% of the total tubulin was in polymerized form. This study provides evidence that microtubules may be involved in guiding transport of small secretory vesicles to the apical regions of cells for exocytosis.

    Notizen: Summary Based on cell-free processing whereby membrane glycoproteins from one cell type were processed by enzymes located in Golgi apparatus from another cell type, J. Rothman and colleagues postulated that vesicles budding from one Golgi apparatus stack migrated to and fused with cisternal membranes of other Golgi apparatus stacks in the cell-free milieu. An extension of this hypothesis was that these same or similar vesicles were involved in the trafficking of membrane material from one cisterna to the next even in the same Golgi apparatus stack [W. G. Dunphy, J. E. Rothman: Compartmental organization of the Golgi stack. Cell 42: 13–21 (1985)]. A coated bud revealed by tannic acid-containing fixatives was the morphological entity associated with this intercompartment Golgi apparatus transfer. This report summarizes information from the author's laboratories that suggests that perhaps the majority of these coated buds, while associated with the Golgi apparatus, are not vesicles per se but rather coated ends of tubules. Golgi apparatus tubules have been postulated to permit interconnections among adjacent Golgi apparatus stacks but not to function in transport between contiguous cisternae of the same Golgi apparatus stack.

    Notizen: Summary Interaction of adjacent secretory vesicles through ball and socket figures was observed in milk secreting mammary epithelial cells preserved by quick freezing without chemical fixation. Membranes of adjacent vesicles appeared to be fused together. Such vesicle-vesicle interactions have been described in chemically fixed tissues, but it has been argued that these interactions may have been fixation artifacts. Ball and socket figures in quick-frozen cells were smaller than those in chemically fixed cells. However, their existence in cells arrested by quick freezing argues that such vesicle-vesicle interactions occur in living cells.

    Notizen: Summary Isolated fractions enriched in transition vesicles isolated by preparative free-flow electrophoresis from transition elements of endoplasmic reticulum incubated with ATP and a cytosol fraction in a cell free system, exhibited a polypeptide composition distinct from that of the original transitional endoplasmic reticulum fractions. The transition vesicles were deficient in peptides of apparent molecular mass between 200 and 270 kD, in the vicinity of 65 kD and less than 40 kD. Immunoblot analysis suggested a 140 kD protein to be concentrated in transition vesicles and low or absent from the endoplasmic reticulum fractions from which the transition vesicles were derived.
