Description: Upon wounding or pathogen invasion, leaves of sorghum [ Sorghum bicolor (L.) Moench] plants with the P gene turn purple, whereas leaves with the recessive allele turn brown or tan. This purple phenotype is determined by the production of two 3-deoxyanthocyanidins, apigeninidin and luteolinidin, which are not produced by the tan-phenotype plants. Using map-based cloning in progeny from a cross between purple Nakei-MS3B ( PP ) and tan Greenleaf ( pp ) cultivars, we isolated this gene, which was located in a 27-kb genomic region around the 58.1 Mb position on chromosome 6. Four candidate genes identified in this region were similar to the maize leucoanthocyanidin reductase gene. None of them was expressed before wounding, and only the Sb06g029550 gene was induced in both cultivars after wounding. The Sb06g029550 protein was detected in Nakei-MS3B, but only slightly in Greenleaf, in which it may be unstable because of a Cys252Tyr substitution. A recombinant Sb06g029550 protein had a specific flavanone 4-reductase activity, and converted flavanones (naringenin or eriodictyol) to flavan-4-ols (apiforol or luteoforol) in vitro . Our data indicate that the Sb06g029550 gene is involved in the 3-deoxyanthocyanidin synthesis pathway.

Notes: Abstract A new high density geophysical data set in the Parece Vela Basin north of 15°N has been obtained through surveys conducted by the Hydrographic Department of Japan. The combined analyses of the swath bathymetry, magnetic and gravity anomalies from these surveys reveal a new scenario for the genesis of this basin. The evolutionary process is as follows: rifting and crust thinning (29–26 Ma), northward propagation of east-west opening (26-23 Ma) , east-west opening together with the Shikoku Basin (23–21 Ma), and the northeast-southwest opening (20/19–15 Ma). The western part of the basin is complicated, displaying some traces of northward propagation of the spreading center. The change between early east-west opening and the final stage of northeast-southwest spreading is marked by a distinct north-south boundary in both structural and magnetic patterns. Deep and rough topography of the extinct Parece Vela Rift is due to magma starvation in the terminal phase of the spreading.

Notes: Summary The characteristic clinical features of diabetes mellitus with mitochondrial DNA (mtDNA) 3243(A-G) mutation are progressive insulin secretory defect, neurosensory deafness and maternal inheritance, referred to as maternally inherited diabetes mellitus and deafness (MIDD). A treatment for MIDD to improve insulin secretory defects and reduce deafness has not been established. The effects of coenzyme Q10 (CoQ10) treatment on insulin secretory response, hearing capacity and clinical symptoms of MIDD were investigated. 28 MIDD patients (CoQ10-DM), 7 mutant subjects with impaired glucose tolerance (IGT), and 15 mutant subjects with normal glucose tolerance (NGT) were treated daily with oral administration of 150 mg of CoQ10 for 3 years. Insulin secretory response, blood lactate after exercise, hearing capacity and other laboratory examinations were investigated every year. In the same way we evaluated 16 MIDD patients (control-DM), 5 mutant IGT and 5 mutant NGT subjects in yearly examinations. The insulin secretory response assessed by glucagon-induced C-peptide secretion and 24 h urinary C-peptide excretion after 3 years in the CoQ10-DM group was significantly higher than that in the control-DM group. CoQ10 therapy prevented progressive hearing loss and improved blood lactate after exercise in the MIDD patients. CoQ10 treatment did not affect the diabetic complications or other clinical symptoms of MIDD patients. CoQ10 treatment did not affect the insulin secretory capacity of the mutant IGT and NGT subjects. There were no side effects during therapy. This is the first report demonstrating the therapeutic usefulness of CoQ10 on MIDD. [Diabetologia (1998) 41: 584–588]

Description: We propose a new scheme based on geopotential height fields to detect cutoff lows starting in the preexisting trough stage. The intensity and scale derived from the proposed scheme will allow for a better understanding of the cutoff low life cycle. These cutoff lows often accompany mesoscale disturbances, causing adverse weather-related events, such as intense torrential rainfall and/or tornadoes. The proposed scheme quantifies the geometric features of a depression from its horizontal height profile. The height slope of a line intersecting the depression bottom and the nearest tangential point (optimal slope) locally indicates the intensity and scale of an isolated depression. The strength of the proposed scheme is that, by removing a local background height slope from a geopotential height field, the cutoff low and its preexisting trough are seamlessly detected as an identical depression. The distribution maps for the detected cutoff lows and preexisting troughs are illustrated along with their intensities, sizes, and local background flows estimated from snapshot height fields. We conducted climatological comparisons of cutoff lows to determine the utility of the proposed scheme.

Description: On the early February 2018, a cold surge struck the Japanese Islands and brought the Japan sea polar air mass convergence zone (JPCZ). The edge of JPCZ stayed at Hokuriku district of Japan, and gave a heavy snow. On the same time, a polar meso cyclone (PMC) also stayed over the Japan Sea west of Hokkaido. Despite that this simultaneous event of JPCZ and PMCs have been observed many times, their interaction remains unclear. And, since PMCs rarely move northward, such mechanisms also are yet fully understood. In this study, by examining synoptic environments, we investigate physical backgroungs of the northward movement of the polar meso cyclone mentioned above and discuss its linkage to the JPCZ. We used data of the Meso-scale Model provided by Japan Meteorological Agency. During the PMC was moving northward, convections were observed over the north side of the PMC and their latent heating could pull the low center northward. The convections could be enhanced by warm and humid air transported from an extratropical cyclone over the Pacific. While the PMC moved northward, the upper level cold air moved southward. Thus, the PMC left from the baroclinic zone, became slow under weak upper level winds, and eventually stopped moving due to the surfase monsoonal northerly wind. We would like to show the results for other simultaneous events of JPCZ and PMCs which were observed during January to February 2018.

Description: Cutoff lows (COLs) are mid-latitude cyclones appear at the upper troposphere. They contain cold air mass derived from the polar region. COLs decrease stability of the lower troposphere (e.g., Hoskins et al. 1985) and often accompany severe weathers (e.g., Mohr et al. 2020), which sometimes causes great damages to mid-latitude regions, such as Japan. Besides, the approaching routes of COLs to the Japan Islands and their seasonal cycles have not been investigated with objective method. In this study, we attempt to elucidate them by utilizing depression detection scheme of Kasuga et al. (2021; K21), which can detect COLs from their early stage (pre-existing trough stage). We have implemented a tracking scheme of COLs by simply searching overlapping of area of COLs. Merge and split of COLs are also recorded to investigate variations of the generation/dissipation of COLs. Generally, COLs approach the Japan Islands from northwest advected by the subpolar jet throughout the seasons. They also approach from southeast in JJA and SON at 200 hPa passing around the North Pacific Subtropical High. These two routes were reported in different studies; we successfully track them with a consistent criterion and confirmed that they have longer durations compared to that of a geopotential minimum detection. Tracks shows different characters at 200 and 500 hPa levels as preceding studies have noted. We have found that strong class of COLs are most frequent in MAM. We are investigating climatological backgrounds of the seasonal variations of COLs and hoping to show the results in the conference.
