    Notes: The induction of pyruvate kinase (ATP: pyruvate 2-O phosphotransferase, EC 2.7.1.40; PK) by estradiol or testosterone in the cerebral hemisphere of male and female rats of different ages was studied. There is a marked decrease in the activity of the enzyme of normal male and female rats with increasing age. Orchiectomy decreases its activity in young and old rats, but not in adult rats. Ovariectomy decreases its activity significantly in all the ages. Estradiol (50μg/100g body wt.) induces its activity in castrated male and female rats, but its effect is greater in female rats. A higher dose of estradiol (100μg/100g body wt.) has age- and sex-dependent induction. Testosterone (50 and 100μg/100g body wt.) has very little effect on its activity in castrated male and female rats of the three ages. The Km values for PEP and ADP, and the Kt values for ATP and l-phenylalanine for the partially purified enzyme of the cerebral hemisphere of normal young and old rats are the same. Preincubation of the enzyme with l-alanine reverses the l-phenylalanine induced inhibition. The concentration of l-alanine required for this effect is the same for both ages. The concentration of Mg2+ required to reverse the inhibitory effect of ATP is the same for young and old rats. Estradiol and testosterone added directly to the incubation mixture do not have any effect on the enzyme activity. These data suggest that the nature of the enzyme molecule does not change with age, but that its induction properties change with age.

    Notes: Abstract In vitro ADP-ribosylation of chromosomal protein and its modulation by spermine, 3-aminobenzamide (3-AB) and benzamide were studied by incubating the nuclei of cerebral hemisphere of 3-, 14- and 30-day old rats with 32P-NAD+. Histones get ADP-ribosylated more than the non-histone chromosomal (NHC) protein. H1 is the major target for ADP-ribosylation. Among the nucleosomal histones, H2B is ADP-ribosylated most. The other core histones also get ADP-ribosylated to a lesser extent. ADP-ribosylation of both histones and NHC proteins decreases during development. Spermine stimulates, whereas 3-AB and benzamide inhibit, 32P-ADP-ribose incorporation into histones and NHC proteins. These effects decrease with development. Mild digestion of chromatin by micrococcal nuclease (MNase), EcoRI, and AluI prior to ADP-ribosylation stimulates incorporation of 32P-ADP-ribose. The degree of stimulation decreases as development proceeds. Such alterations indicate progressive condensation of chromatin with development.

    Notes: Abstract The template-engaged RNA polymerase II was assayed in the nuclei purified from the cerebral hemisphere of 3-, 14- and 30-day old rats. Its activity is the highest at 3-day and declines as development proceeds. Lower transcription at 30-day may either be due to a decrease in the active fraction of chromatin or to a decrease in the amount of RNA polymerase II that is active towards endogenous template, or both. The activity of RNA polymerase I (active in low salt) is also maximal at 3-day and declines as development proceeds. (ADP)ribosylation of chromatin depresses RNA synthesis. This may be due to inactivation of RNA polymerase itself by protein poly(ADP)ribosylation.

    Notes: Abstract The conformation and expression of the albumin gene in the liver of young (21-) and old (85-week) rats were studied. Digestion of nuclei with MNase shows no differences in the nucleosomal organization in the coding region of the gene in the two ages. The gene has a DNase I hypersensitive site which is distinctly less sensitive in old rats. Its 5′-CCGG-3′ sequences are more methylated in the old in which its rate of transcription is also lower.

    Notes: Abstract Age-related changes occur in histones of the liver and oviduct of the female quail. In the liver an extra histone band, named HCX, between H2A and H4, is observed that increases with age. In the oviduct, a large complex of histones is seen which is tissue-specific. This complex declines with increasing age. The changes in the histones of the oviduct of adult and old birds in response to estradiol and progesterone are age-related. In the adult, the histone-complex increases after administration of either one of the hormones. In old birds, however, it is seen only after progesterone administration. Thus, the alterations in histones in the birds are not only tissue- and age-related, but also vary in response to steroid hormones.
