    Notes: We have grown unstrained Al0.66In0.34As0.85Sb0.15/Ga0.64In0.36As0.84Sb0.16 multiple-quantum-well (MQW) structures on InP substrates by metalorganic vapor phase epitaxy. Low-temperature photoluminescence was performed for these MQW structures. By comparing the luminescence peak energies with the theoretical calculations, we estimated the conduction-band offset ratio to be 0.75±0.10 for the Al0.66In0.34As0.85Sb0.15/Ga0.64In0.36As0.84Sb0.16 heterostructure. © 1999 American Institute of Physics.

    Notes: Abstract A simple, sensitive, non-stimulated assay was developed to measure the superoxide anion concentration in whole blood, using an ultra-sensitive chemiluminescence (CL) analyzer, and lucignin amplification. The assay system can be performed without leukocyte isolation or stimulant administration. The blood CL levels of healthy males (362.8±337.7 counts/10 sec) were not different from those of females (335±308.7 counts/10 sec) (p=0.64), whereas the CL levels in whole blood in patients with acute pancreatitis (2522±2014 counts/10 sec) were significantly higher than those of healthy controls (p〈0.001). This assay system may be valuable in the future for quantitative measurement of reactive oxygen species in various disorders.

    Notes: Summary Thirteen urolithiasis patients with unilateral obstructive uropathy were treated with percutaneous nephrostomy (PCN) either for urinary diversion, endopyelotomy, nephrolithtotmy or chemolysis. After percutaneous nephrostomy, the indvidual urine volume, creatinine clearance (Ccr), urinary absolute and fractional excretions of sodium, potassium, calcium, magnesium and inorganic phosphate were measured separately in timed urine collections from a pigtail catheter and from the urethra. The data showed that Ccr and the absolute urinary excretions of sodium, potassium, calcium, magnesium and inorganic phosphate were significantly lower in the PCN kidney immediately or 2 days after relief of obstruction. The ratio of total urinary calcium excretion to urinary creatinine excretion in the obstructed kidney was significantly greater than that in the contralateral kidney. The fractional excretions of calcium and magnesium increased as renal function decreased. The results showed that when the total Ccr is below normal, the apparent excretion of urinary calcium will be underestimated. However, when the total Ccr of patients is within normal range, hypercalciuria may be detected adequately and thus favors early implementation of an appropriate therapeutic strategy.

    Notes: Abstract. Oxygen-derived free radical injury has been associated with several cytopathic conditions. Oxygen radicals produced by chondrocytes is an important mechanism by which chondrocytes induce matrix degradation. In the present study, we extend these observations by studying oxidative processes against osteoblasts. Osteoblasts were mixed in in vitro culture with 200 μM menadione. The cytotoxic effect of menadione-induced oxidative stress was monitored by lucigenin- or luminol-amplified chemiluminescence, tetrazolium assay and immunocytochemical study. Results showed that adding menadione induces an oxidative stress on osteoblasts, via superoxide and hydrogen peroxide production, that can be eradicated by superoxide dismutase (SOD) and catalase in a dose-dependent manner. Catalase and the appropriate concentration of dimethyl sulfoxide have a protective effect on cytotoxicity induced by menadione, whereas SOD does not. Menadione-treated osteoblasts have a strong affinity for annexin V, and the nuclei are strongly stained by TUNEL (TdT-mediated dUTP nick-end labelling). The results suggest that menadione-triggered production of reactive oxygen species leads to apoptosis of osteoblasts.

    Notes: Abstract Surface structures of the early third-stage larae were examined by scanning electron microscopy. There were hemispherical head-bulbs at the apical ends. These bulbs could be clearly distinguished from the bodies. The head-bulbs (52 × 29 Μm) had four transverse rows of sharp booklets. The number of booklets in each row was 37, 36, 38, and 43, posteriorly. The larvae possessed a pair of lateral lips in the head-bulb. Each of the lips had two labial papillae and an amphid between the papillae. Small unidentate cuticular spines were present along the entire length of larvae on their transverse striations. The number of these cuticular striations was 175–217. A pair of cervical papillae and an excretory pore were present on the anterior part of the body. Another pair of papillae was detected laterally on the posterior one-third of the body. The shape of the posterior body papillae resembled that of cervical papillae. The cuticular spines were absent around the anus.

    Description: Glycogen synthase kinase-3 (GSK-3) facilitates interferon (IFN)- signaling. Because IFN- is involved in inflammatory skin diseases, such as psoriasis, the aim of this study was to investigate the pathogenic role of GSK-3 in 12- O -tetradecanoylphorbol-13-acetate (TPA)-induced IFN--mediated ear skin inflammation. TPA (3 μg per ear) induced acute skin inflammation in the ears of C57BL/6 mice, including edema, infiltration of granulocytes but not T cells, and IFN- receptor 1-mediated deregulation of intercellular adhesion molecule 1 (CD54). TPA/IFN- induced GSK-3 activation, which in turn activated signal transducer and activator of transcription 1. Inhibiting GSK-3 pharmacologically, by administering 6-bromoindirubin-3'-oxime (1.5 μg per ear), and genetically, with lentiviral-based short-hairpin RNA, reduced TPA-induced acute skin inflammation but not T-cell infiltration. It is noteworthy that inhibiting GSK-3 decreased TPA-induced IFN- production and the nuclear translocation of T-box transcription factor Tbx21, a transcription factor of IFN-, in CD3-positive T cells. In chronic TPA-induced skin inflammation, inhibiting GSK-3 attenuated epidermis hyperproliferation and dermis angiogenesis. These results demonstrate the dual role of GSK-3 in TPA-induced skin inflammation that is not only to facilitate IFN- signaling but also to regulate IFN- production. Inhibiting GSK-3 may be a potential treatment strategy for preventing such effects.

    Description: Although formation of urothelial carcinoma of the bladder (UCB) requires multiple steps and proceeds along divergent pathways, the underlying genetic and molecular determinants for each step and pathway remain undefined. By developing transgenic mice expressing single or combinatorial genetic alterations in urothelium, we demonstrated here that overcoming oncogene-induced compensatory tumor barriers was critical for urothelial tumor initiation. Constitutively active Ha-ras (Ras*) elicited urothelial hyperplasia that was persistent and did not progress to tumors over a 10 months period. This resistance to tumorigenesis coincided with increased expression of p53 and all pRb family proteins. Expression of a Simian virus 40 T antigen (SV40T), which disables p53 and pRb family proteins, in urothelial cells expressing Ras* triggered early-onset, rapidly-growing and high-grade papillary UCB that strongly resembled the human counterpart (pTaG3). Urothelial cells expressing both Ras* and SV40T had defective G 1 /S checkpoint, elevated Ras-GTPase and hyperactivated AKT-mTOR signaling. Inhibition of the AKT-mTOR pathway with rapamycin significantly reduced the size of high-grade papillary UCB but hyperactivated mitogen-activated protein kinase (MAPK). Inhibition of AKT-mTOR, MAPK and STAT3 altogether resulted in much greater tumor reduction and longer survival than did inhibition of AKT-mTOR pathway alone. Our studies provide the first experimental evidence delineating the combinatorial genetic events required for initiating high-grade papillary UCB, a poorly defined and highly challenging clinical entity. Furthermore, they suggest that targeted therapy using a single agent such as rapamycin may not be highly effective in controlling high-grade UCB and that combination therapy employing inhibitors against multiple targets are more likely to achieve desirable therapeutic outcomes.
