Notes: Abstract The period since death was estimated at the scene in 72 consecutive cases using the temperature-based nomogram method as the primary method and supplemented by examination of criteria such as lividity, rigor mortis, mechanical and electrical excitability of skeletal muscle and chemical excitability of the iris. A case-oriented, computer-assisted selection of the non-temperature-based methods and integration of the results into a common result of the compound method was made following a special logistic. The limits of the period since death as estimated by the nomogram were improved in 49 cases by including the non-temperature-based methods and also provided results in 4 cases where the temperature method could not be used. In a further 6 cases the non-temperature-based methods confirmed the limits estimated by the temperature method but in 14 cases a useful result could not be obtained. In only one of the cases investigated was the upper limit of the period since death, as estimated by the criterion re-establishment of rigor (8 h post-mortem), in contradiction with the period determined by the police investigations (9.4 h post-mortem).

Notes: Abstract In a series of 100 sharp force fatalities – 70 homicides and 30 suicides – the differential diagnostic value of classical findings indicating homicidal or self-infliction was examined. The sensitivity of morphological discrimination criteria is not very high in most cases: tentative or hesitation injuries 63%, defense injuries 47%, exposure of the injured area 30%, stabbing through clothing 75% and rib injuries in the case of stab wounds to the chest 57%. If tentative or hesitation injuries (specifity 93%) or defense injuries (specificity 93%) are present, or if the injured area is exposed (specificity 100%) then these findings can be attributed to a large degree to either homicide or suicide. Case studies are presented to demonstrate the possibility of false positive interpretations of findings including threatening situations that precede a homicide or attempts at intimidation, sadomasochistic practices, static defense constellations, the use of a blunt knife when a victim is unable to react, or ritual actions which may be mistaken for tentative or hesitation injuries. Cuts on the inner surface of the hand that result from clumsy handling with a tool may be mistaken for defense injuries. In severe fight situations the clothing might have ridden up at the time of stabbing. Other findings cannot be attributed with a high significance due to a lack of specificity.

Notes: Abstract The temperature-based nomogram method for estimation of the time period since death was used at the scene of death as the primary method within a compound method in 72 consecutive cases. The situation and cooling conditions inspected and evaluated by the forensic pathologist at the scene are described as far as necessary to ¶enable handling of the method. A comparison of the ¶estimated period since death with the period determined by the police investigations demonstrates the reliability ¶of the method. There were no contradictions in any of the 60 cases between the period of death estimated by this method and that determined by the police investigations. The criminal investigations were effectively supported in the earliest stages in 11 cases despite the fact that the period estimated was of considerable duration.

    Keywords: Heart β-adrenoceptor ; 3′,4′-Dihydroxy-α-methyl propiophenone (U-0521) ; Structure-activity ; cAMP ; Chronotropism
Source: Springer Online Journal Archives 1860-2000
    Topics: Medicine
Notes: Summary The effect of 3′,4′-dihydroxy-α-methylpropiophenone (U-0521) on the rate of spontaneously contracting cultured rat heart cells and right atria of rats and kittens was investigated. The action of U-0521 on the cellular content of cyclic AMP and on the adenylyl cyclase of heart membrane particles was also studied. 1. U-0521 caused positive chronotropic effects on single cultured heart cells and right atria of the rat. U-0521 was about 105 times less potent than (-)-isoprenaline. The maximum effect of U-0521 was smaller than the maximum effect of (-)-isoprenaline. A small positive chronotropic effect of U-0521 was also observed on kitten atria. 2. The β-adrenoceptor blocker (-)-bupranolol antagonized the positive chronotropic effects of U-0521 to the same extent as the effects of (-)-isoprenaline on single cells and atria of the rat. The effects of both U-0521 and (-)-isoprenaline appear therefore mediated through the same β-adrenoceptors. The positive chronotropic effects of U-0521 on kitten atria were also blocked by (-)-bupranolol. 3. Up to 0.1 mM U-0521 did not block the effects of (-)-isoprenaline on rat atria, not even in the presence of corticosterone or hydrocortisone. 4. 1 min incubations with equieffective (increase in cellular beating rate) concentrations of U-0521 (0.1 mM) and (-)-isoprenaline (1 nM) caused a significant increase in the cellular content of cAMP; this effect of both drugs was antagonized by 10 nM (-)-bupranolol. 5. 0.1–3.3 mM U-0521 did not stimulate the adenylyl cyclase of cell-free membrane particles of kitten ventricles. The cyclase was depressed by 10 mM U-0521. 3.3 mM U-0521 caused a 20% decrease of the maximum cyclase-stimulating effect of (-)-isoprenaline and a 1.6-fold increase of its apparent K m. 6. The results with U-0521 suggest that β-adrenoceptors can be activated by agonists devoid of nitrogen. However, the affinity of U-0521 for the β-adrenoceptor is very low (KU-0521 ≈ 5.5 mM). The concentration of U-0521 (0.1 mM) causing maximal increases in beating rate of cultured cells probably occupies less than 7% of the available β-adrenoceptors.
