    Notes: Abstract: Parkinson's disease is characterized by massive degeneration of the melanized dopaminergic neurons in the substantia nigra. The functional capacity of the surviving nigral neurons is affected, as indicated by the subnormal levels of tyrosine hydroxylase (TH) mRNA in these neurons and the presence in the parkinsonian mesencephalon of melanized neurons lacking TH immunoreactivity. This is apparently in contradiction with the known overactivity of dopamine synthesis and release that occurs in the remaining dopaminergic terminals. To test the ability of the surviving neurons to express TH protein, a semiquantitative immunocytochemical method was developed. The relative amounts of TH were estimated with a computer-assisted image analysis system in the dopaminergic neurons of representative mesencephalic sections of control and parkinsonian brains and for comparison in brains from patients with Alzheimer's disease. In control brains, the mean TH content per neuron differed from one subject to another and between the different dopaminergic cell groups of the mesencephalon in the same subject. Within a given dopaminergic region, the level of TH was variable among neurons. In patients with Parkinson's disease, the ratio of TH protein content per neuron in the substantia nigra by reference to that of the central gray substance was reduced. In patients with Alzheimer's disease, the amount of TH was selectively reduced in the remaining dopaminergic neurons of the ventral tegmental area, a region characterized by a loss in dopaminergic neurons. The decrease in cellular TH content might therefore be related to the presence of the neurodegenerative process in the area considered. In patients with Parkinson's disease, the incapacity of the surviving neurons to express normal TH levels may reduce the efficiency of the hyperactivity mechanisms that develop in the remaining striatal dopaminergic terminals.

    Notes: Tyrosine decarboxylase activity of different isolates obtained from 20 samples of raw (before the ripening process) and 10 of ripened “salchichón”, a cured sausage produced in Spain, was evaluated. Although 33% of the isolates obtained from raw sausages showed a positive response in the decarboxylation medium described by Joosten and Northolt (JNM), only 8.4% were subsequently confirmed as tyramine formers by thin layer chromatography (TLC). Pseudomonads and enterobacteria showed the highest percentage of positive isolates in JNM, but none was confirmed by TLC. In contrast, 100% of positive enterococci were confirmed. Most of confirmed isolates obtained from raw sausages were identified as Enterococcus faecalis while Enterococcus faecium and Lactobacillus curvatus showed the highest prevalence in ripened sausages. Inoculation with a tyramine forming strain of Lactobacillus brevis, isolated from raw sausage, did not increase the amounts of tyramine formed during the production process of the sausage.

    Notes: Plant reproduction is highly vulnerable to environmental conditions such as temperature and, consequently, planet warming may have significant consequences on the reproductive phase with serious implication in agricultural crops. Although pollen tube growth is clearly affected by temperature, little information is available on its effect on the female side and on flower receptivity. In this work, the effect of temperature has been evaluated on stigmatic receptivity of sweet cherry in vivo, in the laboratory, and in planta, in the field. Results herein show that temperature has a clear effect on the duration of stigmatic receptivity. Thus, whereas high temperature reduced stigmatic receptivity, low temperature enlarged it. The stigma lost the capacity to offer support first for pollen penetration, second for pollen germination and, finally, for pollen adhesion. The effect of temperature was more pronounced on pollen germination and penetration than on pollen adhesion. High temperature reduced the germination capacity of the pollen as early as the first day after anthesis, a time when no apparent signs of stigma degeneration are apparent. This clear effect of temperature on stigmatic receptivity and pollen performance may have clear implication in crop performance and in establishing screening criteria of best-adapted genotypes.

    Notes: More effective measures to control and replace the dopaminergic deficit of Parkinson's disease are being actively sought. One basic problem is how the striatal dopamine loss should be replaced in order to mimic most accurately the physiological state. Animal electrophysiology indicates that the dopaminergic nigrostriatal pathway has a dual tonic and phasic action. Intermittent dopaminergic stimulation is associated with behavioural hyposensitivity both in animal models and in patients with Parkinson's disease. Continuous dopaminergic stimulation provides a tonic background and improves some clinical problems but is also associated with tolerance. None of the available pharmacological approaches can restore the dopamine deficiency of Parkinson's disease to physiological levels. Continuous dopaminergic stimulation for 〈24 h, associated with small doses of levodopa or a short-acting dopamine agonist, appears to be the best, albeit imperfect, therapeutic approach until other, more efficacious remedies are developed.

    Notes: The effects of nigrostriatal denervation and L-dopa therapy on GABAergic neurons were analysed in patients with Parkinson's disease and in monkeys rendered parkinsonian by MPTP intoxication. The expression of the messenger RNA coding for the 67 kDa isoform of glutamic acid decarboxylase (GAD67 mRNA), studied by quantitative in situ hybridization, was used as an index of the GABAergic activity of the striatal neurons. A significant increase in GAD67 mRNA expression, generalized to all GABAergic neurons, was observed in MPTP-treated monkeys compared to control monkeys in the putamen and caudate nucleus (+44 and +67% respectively), but not in the ventral striatum. L-Dopa therapy significantly reduced GAD67 mRNA expression in the putamen and caudate nucleus to levels similar to those found in control monkeys. However, the return to normal of GAD67 mRNA expression was not homogeneous across all neurons since it was followed by an increase of labelling in one subpopulation of GABAergic neurons and a decrease in another. These data suggest that in MPTP-treated monkeys the degeneration of nigrostriatal dopaminergic neurons results in a generalized increase in GABAergic activity in all the GABAergic neurons of the striatum, which is partially reversed by L-dopa therapy. As the expression of GAD67 mRNA is less intense in the ventral than in the dorsal striatum, this increase in striatal GABAergic activity may be related to the severity of nigrostriatal denervation. In parkinsonian patients who had been chronically treated with L-dopa, GAD67 mRNA expression was significantly decreased in all GABAergic neurons, in the caudate nucleus (by 44%), putamen (by 43.5%) and ventral striatum (by 26%). The opposite variation of GAD67 mRNA in patients with Parkinson's disease, compared with MPTP-treated monkeys, might be explained by the combination of chronic nigrostriatal denervation and long-term L-dopa therapy.

    Notes: The objective of this work was to determine the applicability of the dry-weight-rank (DWR) method for evaluating the botanical composition of forest understorey vegetation. An analysis of plant species abundance was carried out, and instead of ranking the three most abundant species, as is commonly used, up to twelve ranks were scored. Concurrently, four models of relative abundance distribution (RAD) of species were compared for their ability to explain the abundance of species in the study area. The Power-fraction model resulted in the best goodness-of-fit and it was subsequently used to produce the coefficients for the DWR method. Lin's concordance correlation coefficient, the adjusted coefficient of determination, the residual standard deviation and Spearman's rank-order correlation coefficient indicated a good performance of the DWR method. Biomass data and the Shannon index for diversity were also considered. Further analyses showed that there was a trade-off between the number of ranks scored and the accuracy of the botanical composition produced by the DWR method. It is concluded that, so long as the RAD model that explains the distribution of plant species is known, the DWR method can be applied to forest understorey vegetation.

    Notes: In canned anchovies the moisture content after salting decreased from 75.5% to 54%.The loss was accompanied by an increase in salt and ash contents. Free fatty acid (FFA) contents increased gradually throughout ripening. The FFA content variability was too wide to consider it as an index to monitor ripening. Although peroxide and TBA values provide an indication of onset of lipid oxidation, alternative methods are recommended based on tertiary compounds of lipid oxidation. Total volatile basic nitrogen (TVB-N) increased linearly in fish muscle and brine during ripening. TVB-N determination in the brine could be a feasible monitoring method for degree of ripening.
