    Notes: Abstract: The β-amyloid peptide (Aβ), a main constituent in both senile and diffuse plaques in Alzheimer's disease brains, was previously shown to be neurotoxic and to be able to interact with several macromolecular components of brain tissue. Previous investigations carried out in our laboratory demonstrated free radical species formation in aqueous solutions of Aβ(1–40) and its C-end fragment, Aβ(25–35). Toxic forms of Aβ rapidly inactivate the oxidation-sensitive cytosolic enzyme glutamine synthetase (GS). In this regard, we suggested and subsequently demonstrated that Aβ radicals can cause an oxidative damage of cell proteins and lipids resulting in disruption of membrane functions, enzyme inactivation, and cell death. Because GS can be a substrate for Aβ-derived oxidizing species, the present study was conducted to determine if GS could protect against Aβ neurotoxicity. In contrast to this initial hypothesis, we here report that GS significantly enhances the neurotoxic effects of Aβ(1–40). The Aβ-mediated inactivation of GS was found to be accompanied by the loss of immunoreactive GS and the significant increase of Aβ(1–40) neurotoxicity.

    Notes: Phosphorus is one of several dopants that electronically compensate the native deep donor responsible for the yellow coloration observed in bismuth silicon oxide (BSO). Low-temperature optical absorption measurements of a series of Czochralski-grown P-doped BSO crystals show that ∼0.1–0.15 at. % P is needed in the sample to fully remove the yellow coloration. The absorption cutoff in the fully compensated P-doped sample was at 3.2 eV while compensated Al- and Ga-doped samples cutoff at 3.35 eV. Excitation at 10–15 K with near band-edge light produces photochromic absorption bands. In the lightly-doped (partially bleached) samples these bands were identical to those observed in undoped BSO. In the fully bleached sample a new spectrum was observed. Its major contribution was a band centered near 1.8 eV with a weaker absorption in the blue-green. By comparison with the spectra observed in undoped and in Al-doped material before and after photoexcitation it is believed that the 1.8 eV band is due to the [PO4]− center and that the broad 2.45 eV band observed in Al- and Ga-doped BSO is due to the [BiO4]0 center. © 1995 American Institute of Physics.

    Notes: An investigation was made of the defect characteristics of single-crystal Bi12SiO20 (BSO) grown by both Czochralski (Cz) and hydrothermal transport methods. Only Cz-grown BSO is photorefracting using a coherent pumping source centered around 500 nm, as undoped hydrothermal BSO is transparent throughout the visible spectrum. Thermally stimulated current (TSC) studies in conjunction with temperature-dependent optical-absorption measurements and room-temperature photoconductivity data all indicate that the hydrothermal material is near intrinsic in terms of its low defect content. TSC measurements made below room temperature indicate that concentrations of traps of activation energy 〈0.7 eV are a factor of 103 smaller in hydrothermal than in Cz BSO. At least six different defects were identified in the TSC measurements. Temperature-dependent optical-absorption measurements indicate two Urbach band tails for Cz-grown materials that are not observed in the hydrothermal materials. Cz material of lower purity also possesses an impurity band tail which can be observed through temperature dependent optical absorption measurements. Comparison of Cz material with hydrothermal BSO of similar impurity content suggests that the BiSi defect responsible for the 500 nm absorption may be complexed with an impurity such as Fe or V. In the absence of this defect, however, these impurities have no effect on the absorption. An additional defect in the TSC data is also related to a transition metal impurity. The results indicate that the photoconductivity associated with the photorefractive effect in Cz material must proceed via a trap-hopping conduction mechanism that is missing in the intrinsic material.

    Notes: Objective— To compare the cumulative pregnancy rates after gamete intrafallopian transfer (GIFT) with the cumulative spontaneous pregnancy rates in couples with unexplained infertility.Design— A contemporaneous study in a single group of patients.Setting— Northern Regional Fertility Centre.Subjects— 76 couples with unexplained infertility of more than 3 years durationInterventions— Successful pregnancies were recorded during at least 3 months before GIFT and up to 21 months after a maximum of three cycles of GIFT treatment.Main outcome measures— Pregnancy resulting in a live birth.Results— Average monthly fecundability without treatment was 0.021 and after GIFT was 0.14 (P〈0.001). This was reflected as a cumulative pregnancy rate of 52% after three cycles of GIFT and 30% after 24 months without treatmentConclusions— The chance of having a baby after one cycle of GIFT is significantly greater than the chance in a spontaneous cycle. However, considering the cumulative pregnancy rates, we suggest that if GIFT is to be a realistic treatment option, it should be offered for more than one cycle.

    Notes: Objective— To compare GIFT, intrauterine insemination (IUI) with, and without, ovarian hyperstimulation in the treatment of unexplained infertility.Design— Women randomly allocated to one of three treatment protocols.Setting— Northern Regional Fertility Centre.Subjects— 59 couples with unexplained infertility of more than 3 years duration.Interventions— Three cycles of either GIFT, IUI after ovarian hyperstimulation or IUI in a spontaneous cycle.Main outcome measures— Pregnancy resulting in a live birth.Results— Fecundabilities were 0.12 after GIFT, 0.018 after ovarian hyperstimulation and IUI, and 0.018 after IUI in a spontaneous cycle. The fecundability after IUI was no different from that which would be expected without treatment in these couples but fecundability was significantly better (P〉0.02) after GIFT.Conclusions— This trial does not support the use of IUI in the treatment of unexplained infertility but confirms the value of GIFT.
