    Notes: Dowling-Degos' anomaly is characterized by a reticular and spotted pigmentation of the skin folds; this pigmentary disturbance, occurring most often in women, is a dominant inherited genodermatosis which worsens progressively and may exhibit in the early phase rapid changes in severity after emotional stress. In a 9-year-old girl, an electronmicroscopic study of pigmented lesions showed a strong melanocytic activity with quantitative increase of the melanosomes; the average size of the melanosomes was not different when compared to normal Caucasoid skin, but in the keratinocytes they were distributed according to a dispersed pattern as in black skin. The pigmentary state of Dowling-Degos' disease is another example of melanocyte-keratinocyte interaction where the epidermal melanin pattern and the size of pigment granules are not in striking correlation. An accelerated rate of melanogenesis and pigmentation may be another factor determining a non-aggregated distribution of the melanosomes within the keratinocytes. The nature of the stimulus of pigmentation in these skin areas which are not sun-exposed is still unclear.

    Notes: Langerhans cells (LC) play an important role in the skin immune system. They are bone marrow-derived and function as the only accessory and antigen-presenting cells in the skin. Several techniques for enriching these cells have been devised, and four, including density gradient centrifugation, use of cell sorter, panning and immunomagnetic separation, are discussed. It is concluded that the most satisfactory method for isolation of LC is based on density gradient centrifugation and the most satisfactory for depletion of epidermal cell preparations for LC is based on the immunomagnetic principle.

    Notes: Numerous tumors with follicle-like direction of differentiation have been described. At least three of them connected to the overlying epidermis are related to the outer root sheath or trichilemma: the tumor of follicular infundibulum (Mehregan & Butler 1961) and the inverted follicular keratosis (Helwig 1955) or follicular poroma (Duperrat & Mascaro 1963) have been related to the infundibulum, the tricholemmoma (Headington & French 1961) to the lower outer root sheath. On the occasion of a case first diagnosed as proliferating trichilemmal cyst of the scalp, we think we have individualized a tumor exhibiting ability to undergo trichilemmal keratinization. We describe its histogenesis and stress its peculiarities compared with other lesions showing trichilemmal keratinization.

    Notes: T cell tolerance induction to foreign and self-antigens has occupied research since the beginning of the understanding of the immune system. Much controversy still exists on this question even though new methods became available to investigate immunoregulatory mechanisms. Antigen-presenting cells play a pivotal role in transferring information from the periphery of the organism to lymphoid organs. There, they initiate not only the activation of naive T cells but seem to deliver important signals which result in T cell unresponsiveness with antigen-specific tolerance induction.

    Description: CD1e is the only human CD1 protein existing in soluble form in the late endosomes of dendritic cells, where it facilitates the processing of glycolipid antigens that are ultimately recognized by CD1b-restricted T cells. The precise function of CD1e remains undefined, thus impeding efforts to predict the participation of this protein in the presentation of other antigens. To gain insight into its function, we determined the crystal structure of recombinant CD1e expressed in human cells at 2.90-Å resolution. The structure revealed a groove less intricate than in other CD1 proteins, with a significantly wider portal characterized by a 2 Å-larger spacing between the α1 and α2 helices. No electron density corresponding to endogenous ligands was detected within the groove, despite the presence of ligands unequivocally established by native mass spectrometry in recombinant CD1e. Our structural data indicate that the water-exposed CD1e groove could ensure the establishment of loose contacts with lipids. In agreement with this possibility, lipid association and dissociation processes were found to be considerably faster with CD1e than with CD1b. Moreover, CD1e was found to mediate in vitro the transfer of lipids to CD1b and the displacement of lipids from stable CD1b–antigen complexes. Altogether, these data support that CD1e could have evolved to mediate lipid-exchange/editing processes with CD1b and point to a pathway whereby the repertoire of lipid antigens presented by human dendritic cells might be expanded.

    Description: Lipids are important antigens that induce T cell-mediated specific immune responses. They are presented to T lymphocytes by a specific class of MHC-I like proteins, termed CD1. The majority of the described CD1-presented mycobacterial antigens are presented by the CD1b isoform. We previously demonstrated that the stimulation of CD1b-restricted T cells by the hexamannosylated phosphatidyl-myo-inositol (PIM6), a family of mycobacterial antigens, requires a prior partial digestion of the antigen oligomannoside moiety by α-mannosidase and that CD1e is an accessory protein absolutely required for the generation of the lipid immunogenic form. Here, we show that CD1e behaves as a lipid transfer protein influencing lipid immunoediting and membrane transfer of PIM lipids. CD1e selectively assists the α-mannosidase-dependent digestion of PIM6 species according to their degree of acylation. Moreover, CD1e transfers only diacylated PIM from donor to acceptor liposomes and also from membranes to CD1b. This study provides new insight into the molecular mechanisms by which CD1e contributes to lipid immunoediting and CD1-restricted presentation to T cells.

    Description: Background: Experts agree that one of the more promising strategies in cancer management is early detection coupled with early intervention. In this study, we evaluated an early cancer detection strategy of cancer presence based on serum levels of the cancer-specific transcript variants of ENOX2 in serum coupled with an ENOX2-targeted nutraceutical preparation of green tea concentrate plus Capsicum (Capsol-T(R)) as a strategy of Curative Prevention(R) involving early detection coupled with early intervention in early stage cancer when in its most susceptible and manageable stages.Experimental design: One hundred ten (110) subjects were tested for cancer presence using the ONCOblot(R) Tissue of Origin 2-D gel/western blot protocol for detection of serum presence of transcription variants of the ENOX2 protein. Subjects testing positive for ENOX2 received 350 mg of Capsol-T(R) in capsule form every 4 h including during the night for periods of at least 3 to 6 months or longer after which they were again tested for ENOX2 presence using the ONCOblot(R) Tissue of Origin Cancer Test protocol. Results: Of the 110 subjects, both male and female, ages 40 to 84, with no evidence of clinical symptoms of cancer, 40% were positive for ENOX2 presence in the ONCOblot(R) Tissue of Origin Cancer Test. After completion of 3 to 17 months of Capsol-T(R) use, 94% of subjects subsequently tested negative for ENOX2 presence. Conclusions: Oral Capsol-T(R) is well tolerated and, for ENOX2 presence in serum in the absence of clinical cancer symptoms, is consistently effective in reducing the serum ENOX2 levels to below detectable limits.

    Description: Neurturin (NTN) was previously described for its neuronal activities, but recently, we have shown that this factor is also involved in asthma physiopathology. However, the underlying mechanisms of NTN are unclear. The aim of this study was to investigate NTN involvement in acute bronchial Th2 responses, to analyze its interaction with airway structural cells, and to study its implication in remodeling during acute and chronic bronchial inflammation in C57BL/6 mice. We analyzed the features of allergic airway inflammation in wild-type and NTN –/– mice after sensitization with two different allergens, OVA and house dust mite. We showed that NTN –/– dendritic cells and T cells had a stronger tendency to activate the Th2 pathway in vitro than similar wild-type cells. Furthermore, NTN –/– mice had significantly increased markers of airway remodeling like collagen deposition. NTN –/– lung tissues showed higher levels of neutrophils, cytokine-induced neutrophil chemoattractant, matrix metalloproteinase 9, TNF-α, and IL-6. Finally, NTN had the capacity to decrease IL-6 and TNF-α production by immune and epithelial cells, showing a direct anti-inflammatory activity on these cells. Our findings support the hypothesis that NTN could modulate the allergic inflammation in different mouse asthma models.
