    Notes: . The α- and the β-tubulin genes of the hypotrichous ciliate Euplotes octocarinatus were isolated from a size-selected macronuclear DNA library. The α-tubulin gene is located on a 1,587 bp macronuclear DNA molecule and the β-tubulin gene on a 1,524 bp macronuclear DNA molecule. Sequencing revealed that all the cysteine residues of the two genes are encoded by the common cysteine codons UGU and UGC and none by an UGA codon. This is in contrast to the genes of E. octocarinatus sequenced so far, where some of the cysteines are encoded by the opal codon UGA. The tubulin genes end like other Euplotes genes with a TAA. They do not contain introns. The last codon for an amino acid in the α-tubulin gene is a GAA which codes for glutamic acid. This is in contrast to what has been reported for most α-tubulin genes, but it supports findings for other hypotrichous ciliates. No evidence for the existence of more than one type of α- and one type of β-tubulin genes could be obtained.

    Notes: We report a method that allows us to grow and maintain the freshwater ciliate Euplotes octocarinatus in large quantities. Frequent exchange of culture fluid proved more effective than aeration in obtaining high cell densities (4200 cells/ml) and reasonable doubling times in large-scale cultures. For harvesting gamone 1, the cell density was raised to 10,000 cells/ml. Under these conditions, the cells continued to produce and secrete gamone; they were slightly starved, but they no longer divided. Cell-free fluid with a steady and relatively high yield of gamone was obtained from two such cultures over a period of five months. We isolated gamone 1 also from cell homogenates and compared it with secreted gamone 1, but found no differences in the gamones from these two sources.

    Notes: . Concanavalin A (≥ 50 μg/ml) inhibits pair formation in both of the two complementary mating types of Euplotes octocarinatus studied in this investigation. This effect can be reversed by methyl-α-d-mannose. Concanavalin A is accessible for methyl-α-d-mannose until pairs are formed. Methyl-α-d-mannose as well as methyl-α-d-glucose and 2-acetamido-2-deoxy-d-glucose alone do not inhibit pair formation unless applied in concentrations ≥ 60 mM. The Concanavalin A-sensitive phase of preconjugant interaction starts 2 h after cells are induced to conjugate. Based on these observations we suggest that Concanavalin A might exhibit its action by binding to carbohydrate moieties of preconjugation-specific adhesion molecules and thereby might allosterically block interactions with their counterparts. To identify preconjugation-specific alterations in number or localization of Concanavalin A-binding glycoconjugates, we probed western blots of total cell proteins or fixed cells, respectively, with digoxigenin-labeled Concanavalin A. On Concanavalin A blots 20 different Concanavalin A-binding glycoconjugates were identified in mating-competent cells. Localization of Concanavalin A-binding sites on mating-competent cells by light microscopy resulted in predominant labeling of a comma-shaped structure near the paroral membranelle. During the preconjugation period no changes in number or localization of Con A-binding glycoconjugates were detected. Possible reasons are discussed.

    Notes: The hypothesis is advanced that all freshwater Euplotes species with a 9 type 1 fronto-ventral cirrus pattern (E. patella type) depend upon bacteria-like endosymbionts. Aposymbiotic cells of these species are unable to divide. The hypothesis is based on the investigation of 40 different freshwater Euplotes stocks collected in Germany, France, the USA, and Japan. No symbionts were found in E. crenosus and E. palustris, freshwater species with 10 fronto-ventral cirri, nor in E. muscicola, a representative of the freshwater Euplotes group with a 9 type 2 fronto-ventral cirrus pattern (E. affinis type). Characteristic for the essential endosymbionts are multiple nucleoids, a feature described earlier for omikron, an indispensable symbiont of E. aediculatus. Although the symbionts differ from omikron and among each other in size, shape, and their average number per host, they are believed to be related to omikron. In two stocks a different type of bacterium was found in which no defined nucleoids can be detected. Transfer of this symbiont into aposymbiotic cells, originally carrying omikron, revealed that it can restore the ability to multiply. Similarly, omikron was also able to restore the ability to divide in cells freed of this symbiont. It is assumed that this different type of symbiont is a secondary invader of Euplotes which displaced the original omikron-like endosymbiont. Some of the stocks were found to carry, in addition to omikron-like symbionts, other symbiotic bacteria; E. daidaleos carries in addition an alga. The findings suggest that the freshwater Euplotes species with a 9 type 1 cirrus pattern are closely related to each other and evolved from an ancestor (probably of cirrotype 10) which already was dependent upon endosymbionts of the omikron type. It supports the view that the two subgroups of freshwater Euplotes forms with a cirrotype of 9 have evolved independently from each other from species with 10 fronto-ventral cirri by losing a cirrus at different positions.

    Notes: SYNOPSIS. Eight interfertile strains of Euplotes minuta collected from two localities in the Mediterranean Sea are shown to comprise a single breeding system of multiple mating types. One strain regularly passes through autogamy and, in crosses to non-autogamous strains, preliminary evidence for the genic control of autogamy was obtained. The cytogenetic events of conjugation and autogamy are briefly described. Three strains are “killers.” Killer cells liberate a substance into the surrounding medium which has a lethal effect on cells of other strains called “sensitives.” This substance, “euplotein,” is not produced by sensitives. The inheritance of the killer trait is apparently under the control of a cytoplasmic factor but present attempts to identify the factor microscopically were without success. The discussion stresses the potential usefulness of this species for studies of protozoan genetics and development.

    Notes: ZUSAMMENFASSUNG. Der Süßwasserciliat Euplotes aediculatus beherbergt in seinem Cytoplasma 900–1000 stäbchenförmige Symbionten, die für seine Vermehrung essentiell zu sein scheinen. Durch Wachstum in Gegenwart von Penicillin kann Euplotes von seinen Symbionten befreit werden, es führt dies jedoch stets auch zu einem Verlust der Teilungsfähigkeit. Eine Reinfektion ist bei etwa 4% der Zellen möglich, die dann nach 4–5 Tagen sich wieder zu vermehren beginnen.Die Endosymbionten wurden sowohl lichtmikroskopisch als auch elektronenmikroskopisch untersucht. In ihrer Feinstruktur und Färbbarkeit gleichen sie, wie kappa und andere von Paramecium her bekannte “killer”-Partikel, gramnegativen Bakterien. Sie unterscheiden sich von diesen Symbionten jedoch dadurch, daß ihre DNA im Cytoplasma nicht homogen verteilt ist, sondern sich in 3–9 schon lichtmikroskopisch erkennbaren Zentren konzentriert. Die in Euplotes vorkommenden Symbionten lassen keine “killer”-Aktivität erkennen. Sie werden omikron-Partikel genannt.SYNOPSIS. The fresh water ciliate Euplotes aediculatus contains in its cytoplasm 900–1000 rod-shaped symbiotes which appear to be essential for division. Growth of Euplotes in the presence of penicillin results in loss of these symbiotes and simultaneously in a loss of the ciliate's ability to divide. Reinfection with the symbiotes can be achieved in 4% of the cells which then resume growth after a lag period of 4–5 days.The endosymbiotes have been studied by light and electron microscopy. In their fine structure and staining reaction they resemble gram-negative bacteria as do kappa and other killer particles of Paramecium. The symbiotes of Euplotes, however, are unusual in that their DNA is not distributed throughout the cytoplasm but is localized in 3–9 areas (nucleoids), which are visible even in the light microscope. No killing activity seems to be associated with the symbiotes. Following the practice of referring to those endosymbiotes by Greek letters they are here designated omikron particles.

    Notes: SYNOPSIS. The killer strains of Euplotes minuta contain cytoplasmic epsilon particles absent in sensitive strains. The toxic principle of killers is associated with large particles which sediment readily in the centrifuge and may be identical with epsilon. Killing particles are inactivated by heat, certain proteolytic enzymes, and lysis with the French press.

    Notes: . Three macronuclear genes encoding putative nuclear protein kinases of the ciliate Euplotes octocarinatus syngen I were isolated and sequenced. All three deduced gene products share significant properties with a group of recently identified nuclear serine/ threonine protein kinases named Ndr. The three predicted proteins contain the twelve conserved catalytic subdomains of protein kinases and 22 near universally-conserved amino acids residues that are characteristic of serine/threonine protein kinases. In addition, there is an—30 amino acid-peptide insertion between subdomains VII and VIII that contains a potential nuclear localization signal. Sequence analysis suggests that expression of the Eondr2 gene requires a +1 programmed translational frameshift for its translation. Comparison of the deduced EoNdr2 with other known Ndr protein kinases implies that a +1 ribosomal frameshift occurs at the motif AAATAA.
