    Description: Inflammation conveys the development of glomerular injury and is a major cause of progressive kidney disease. NF-B signaling is among the most important regulators of proinflammatory signaling. Its role in podocytes, the epithelial cells at the kidney filtration barrier, is poorly understood. Here, we inhibited NF-B signaling in podocytes by specific ablation of the NF-B essential modulator (NEMO, IKK). Podocyte-specific NEMO-deficient mice (NEMO pko ) were viable and did not show proteinuria or overt changes in kidney morphology. After induction of glomerulonephritis, both NEMO pko and control mice developed significant proteinuria. However, NEMO pko mice recovered much faster, showing rapid remission of proteinuria and restoration of podocyte morphology. Interestingly, quantification of infiltrating macrophages, T-lymphocytes, and granulocytes at day 7 revealed no significant difference between wild-type and NEMO pko . To further investigate the underlying mechanisms, we created a stable NEMO knockdown mouse podocyte cell line. Again, no overt changes in morphology were observed. Translocation of NF-B to the nucleus after stimulation with TNFα or IL-1 was sufficiently inhibited. Moreover, secretion of proinflammatory chemokines from podocytes after stimulation with TNFα or IL-1 was significantly reduced in NEMO-deficient podocytes and in glomerular samples obtained at day 7 after induction of nephrotoxic nephritis. Collectively, these results show that proinflammatory activity of NF-B in podocytes aggravates proteinuria in experimental glomerulonephritis in mice. Based on these data, it may be speculated that immunosuppressive drugs may not only target professional immune cells but also podocytes directly to convey their beneficial effects in various types of glomerulonephritis.

    Description: In certain sporadic, familial, and infectious prion diseases, the prion protein misfolds and aggregates in skeletal muscle in addition to the brain and spinal cord. In myocytes, prion aggregates accumulate intracellularly, yet little is known about clearance pathways. Here we investigated the clearance of prion aggregates in muscle of transgenic mice that develop prion disease de novo . In addition to neurodegeneration, aged mice developed a degenerative myopathy, with scattered myocytes containing ubiquitinated, intracellular prion inclusions that were adjacent to myocytes lacking inclusions. Myocytes also showed elevated levels of the endoplasmic reticulum chaperone Grp78/BiP, suggestive of impaired protein degradation and endoplasmic reticulum stress. Additionally, autophagy was induced, as indicated by increased levels of beclin-1 and LC3-II. In C2C12 myoblasts, inhibition of autophagosome maturation or lysosomal degradation led to enhanced prion aggregation, consistent with a role for autophagy in prion aggregate clearance. Taken together, these findings suggest that the induction of autophagy may be a central strategy for prion aggregate clearance in myocytes. IMPORTANCE In prion diseases, the prion protein misfolds and aggregates in the central nervous system and sometimes in other organs, including muscle, yet the cellular pathways of prion aggregate clearance are unclear. Here we investigated the clearance of prion aggregates in the muscle of a transgenic mouse model that develops profound muscle degeneration. We found that endoplasmic reticulum stress pathways were activated and that autophagy was induced. Blocking of autophagic degradation in cell culture models led to an accumulation of aggregated prion protein. Collectively, these findings suggest that autophagy has an instrumental role in prion protein clearance.

    Description: Chromatin-rich smears are well-recognised artefacts in H&E sections, which may result from nuclear fragility and tissue crushing. Less well known among surgical pathologists is a novel, recently discovered form of cell death called ‘NETosis’, which at the light microscopical level closely resembles such crush artefacts. Figure 1 A shows an H&E stain of thrombus obtained from human aorta. Besides platelets, fibrin and red blood cells, this type of specimens often contain large numbers of neutrophils. 1 In this image, deep purple stained treads of variable length and often clustered can be noticed, which will easily be interpreted as a crush artefact. However, these structures are, in fact, neutrophil extracellular traps (NETs).     Notes: Sequential sputter deposition of amorphous 4-nm-thick Ta and 7-nm-thick Si layers leads to the formation of a periodic multilayer that can be regarded as a synthetic one-dimensional crystal. A simple kinematical diffraction model is presented to discuss x-ray diffraction patterns obtained from the multilayer structure during annealing. Post-deposition annealing up to temperatures above 530 °C, where the lattice structure disappears, leads to Si diffusion into the Ta layers forming a nonstoichiometric Ta-Si phase. The high diffusion rate of Si in Ta leads to a homogeneous distribution of Si in the Ta (silicide) regimes. As shown by cross-sectional transmission electron microscopy, the interfaces remain sharp during the interdiffusion process. This makes the observation of low-angle Bragg reflections up to the 12th order possible. The local atomic structure around the Ta atoms was analyzed by measuring the extended x-ray-absorption fine structure above the Ta LIII edge at different probing depth conditions. During annealing the local Ta environment was found to be TaSi2 like, even if only a small portion of the Si has diffused into the Ta layers, and the silicide formation is far from stoichiometry. The formation of a Ta suboxide was detected in the surface region of the top Ta layer.

    Notes: The migraine prophylactic effect of 10 mmol magnesium twice-daily has been evaluated in a multicentre, prospective, randomized, double-blind, placebo-controlled study. Patients with two to six migraine attacks per month without aura, and history of migraine of at least 2 years, were included. A 4-week baseline period without medication was followed by 12 weeks of treatment with magnesium or placebo. The primary efficacy end-point was a reduction of at least 50% in intensity or duration of migraine attacks in hours at the end of the 12 weeks of treatment compared to baseline. With a calculated total sample size of 150 patients, an interim analysis was planned after completing treatment of at least 60 patients, which in fact was performed with 69 patients (64F, 5M), aged 18–64 years. Of these, 35 had received magnesium and 34 placebo. The number of responders was 1 in each group (28.6% under magnesium and 29.4% under placebo). As determined in the study protocol, this was a major reason to discontinue the trial. With regard to the number of migraine days or migraine attacks there was no benefit with magnesium compared to placebo. There were no centre-specific differences, and the final assessments of treatment efficacy by the doctor and patient were largely equivocal. With respect to tolerability and safety, 45.7% of patients in the magnesium group reported primarily mild adverse‘ events like soft stool and diarrhoea in contrast to 23.5% in the placebo group.
