    Notes: Summary Tumor DNA from 27 patients with treated or untreated transitional cell carcinomas of the urinary tract was screened for genomic alterations of the multidrug resistance genes in order to determine whether structural changes of these genes are important in primary urothelial tumors. None of the tumors showed evidence of amplification or rearrangements of either mdr1 or mdr2. The lack of amplification or rearrangements observed in these tumors suggests that structural alterations of the mdr1 and mdr2 genes are not important mediators of drug resistance in TCC.

    Notes: Summary No simple screening test for prostatic cancer (CaP) is yet available. Likewise, in most cases, accurate staging requires surgical exploration to determine nodal metastases although there are no data to support the concept that pelvic lymphadenectomy cures nodal disease. Future developments which may significantly alter our ability to diagnose and stage CaP include hybridoma techniques, nuclear magnetic resonance and positron imaging tomography.

    Notes: Summary The encouraging responses noted in some patients with transitional cell carcinoma of the bladder to cis-platinum based chemotherapy have led to the hope that combination therapy utilizing chemotherapy plus limited surgery may make it possible to preserve the bladder in patients with muscle-invading bladder tumors. Despite the conceptual appeal of a bladder-sparing approach, the effectiveness of combined therapy remains unresolved. At present, there are no data to support the routine use of chemotherapeutic agents combined with surgery in the therapy of bladder cancer. The role of neoadjuvant therapy in the treatment of muscle invasive bladder cancer will only be defined by the results of properly conducted and controlled randomized studies.

    Notes: Summary The effect of High energy shock waves (HESW) on the viability and proliferation of normal human bone marrow cells was evaluated. A dose dependent increase in cytotoxicity with an increase in the number of HESW was demonstrated. In general 700 HESW immediately reduced the cell viability of bone marrow cells by 50%. The CFU-GM assay provides a method to evaluate the effect of HESW on the proliferative capacity of bone marrow. Following HESW treatment the colony forming ability of trypan blue excluding cells also felt in a dose dependent fashion, but some variation in sensitivity was noted. By comparing the sensitivity of various cells, the cells of normal human bone marrow were felt to be less sensitive to HESW effects than those of other tissue cultured cells or malignant cell lines.

    Notes: Summary Human prostatic tissue and expressed prostatic secretions (EPS) efrom patients with benign prostatic hyperplasia (BPH) and prostate cancer were incubated with (1 β 3H) androstenedione. The extent of aromatization was determined by measuring the transfer of 3H from the 1 β position into water. The amount of 3H2O recovered corresponds to the estrogens formed. Tissue from 5 patients with BPH yielded 2.13 (±1.05) pmol/mg protein/h while the EPS from the same patients yielded 727 fmol/mg protein/h. In patients with prostate cancer the mean formation of estrogens was 388 fmol/mg protein/h (±75). 4-hydroxyandrostenedione, an aromatase inhibitor, successfully inhibited aromatization in BPH and prostate cancer 53–98%.
