    Notes: Didymella rabiei grew saprophytically on pieces of artificially and naturally infected chickpea stem debris under artificial incubation conditions, and formed pseudothecia and pycnidia. The extent of growth was not significantly affected by temperature of incubation within the range 5–25°C, but was significantly reduced as relative humidity (RH) decreased from 100% to 86%, when no growth occurred. Pseudothecia matured at 10°C and constant 100% RH, or at 5 and 10°C and alternating 100%/34% RH. Under these conditions, pseudothecial maturation, assessed by a pseudothecia maturity index, increased over time according to the logistic model. For temperatures higher than 10°C or RH lower than 100%, pseudothecia either did not form ascospores, or ascopores did not mature and their content degenerated. When pseudothecia that initially developed to a given developmental stage were further incubated at a constant 100% RH, temperature became less limiting for complete pseudothecial development as the developmental stage was more advanced. Pycnidia of the fungus developed and formed viable conidia in all environmental conditions studied, except at 86% RH. However, the density of pycnidia formed and the number of viable conidia per pycnidium were significantly influenced by temperature, RH and the type of debris (artificially or naturally infected) used.

    Notes: The shrub Cistus ladanifer (gum cistus, rock rose) is a key component of the Maquis in the Mediterranean forests of southern Spain and Portugal. Between 1996 and 1998, and coinciding with exceptionally high rainfall rates, a disease characterized by foliar wilting and death of branches, leading to death of the whole plant, affected C. ladanifer in the Mediterranean oak and pine forests in Andalucía (southern Spain). Field surveys carried out in 15 forest areas showed that diseased branches were associated with the presence of strips of necrotic bark. Botryosphaeria dothidea was consistently obtained from the necrotic tissues. Growth of isolates on potato dextrose agar was very rapid at high temperatures, with a growth rate of 25 mm day−1 at the optimum temperature (about 28°C). Germination rate of conidia increased with temperature, with the optimum temperature being over 35°C. Pathogenicity tests were conducted on cut branches, 3-year-old seedlings and mature plants in the field. In every situation, disease symptoms were reproduced in the inoculated branches and Koch’s postulates were fulfilled. The results demonstrate the behaviour of B. dothidea as a primary pathogen of C. ladanifer. The present work constitutes the first description of fungal canker of C. ladanifer caused by B. dothidea, being the first report of a disease affecting this shrub species.

    Notes: Several attenuation studies have established a frequency dependence law of the anelastic attenuation factor Q in the form Q=Qo(f/fo)v for the approximate 1–10 Hz frequency range. We propose a method that leads to the determination of Qo, which is a function of the reference frequency fo, and the real exponent v with a single station. to carry out the problem we determine a set of master curves as a function of v. We discuss the method, and the different features of the master curves, when it is applied to the complicated regions of the Iberian Peninsula and to several instruments with different responses. Using this new method and the seismographic stations available in the Iberian Peninsula we have mapped iso-Q0 lines, at a reference frequency of 1 Hz, applying inversion methods. the Q0 values determined for Iberia vary between about 100 and about 600. Values close to 100 correspond to the southern part of Iberia. In general, Qo values increase from south to north with values about 600 near the NW part of Iberia. the Pyrenees Mountains and adjacent areas present Q0 values between about 200 and about 350. These results suggest a strong Q0 lateral variation in Iberia. A considerable frequency dependence of coda-Q has also been determined. the v values vary between 0.3 and 0.8. the Q0 values obtained in the Iberian Peninsula show very good agreement with several Q0 values obtained in other regions of the world. Comparison between the iso-Q0 lines and other geophysical parameters, like regional variations of Pn velocities, heat flow, isoseimal intensity distribution and crustal thickness, indicates that lower Q0 values are associated with higher isoseismal intensity attenuation, higher heat flow, lower Pn velocities and thinner crust.

    Notes: This study was conducted with the objective of characterizing the genetic variation among a representative set of 37 barley cultivars currently grown in Spain, using restriction fragment length polymorphism (RFLP) markers. Thirty-two RFLP probes, in combination with three restriction enzymes, were used to analyse polymorphism at the molecular level. Genetic distances (GD), based on RFLP band patterns, and coancestry coefficients (f), based on pedigree records, were calculated. Of the 95 clone-enzyme combinations analysed, 71 (74.7%) were polymorphic, representing 246 RFLP patterns. A cluster analysis of GD split the sample into five distinct germplasm groups that were consistent with the history of the cultivars (winter European, spring European, CIMMYT-ICARDA materials, the single cultivar ‘Dobla’ and Spanish local materials). The Spanish group was the most distinct one and had unique alleles at markers close to major loci determining phonological adaptation. The probes which best distinguished among groups were also identified. Genetic similarity estimates were moderately consistent with f (for cultivars with complete pedigrees). The implications for integration of diversity studies into breeding programmes are discussed.

    Notes: Didymella rabiei grew saprophytically on pieces of infested chickpea stems and pods, and formed pycnidia and pseudothecia. The extent of saprophytic growth and production of viable spores were determined by the incubation conditions. On debris left on the soil surface under natural conditions, the fungus rapidly colonized the tissues, formed abundant pseudothecia and pycnidia, and remained viable throughout the 2 years of the study. When plant debris was buried, D, rabiei was restricted to the original lesions, in which it formed new pycnidia and was viable for 2 to 5 months. Under controlled conditions in the laboratory, D. rabiei extensively colonized plant debris spread over the soil surface. On the other hand, the fungus did not grow on buried debris, or showed only very limited development when the artificially infested debris was buried between two layers of sterilized soil. Incubation temperature was the principal factor associated with the production of conidia and especially ascospores.

    Notes: Background  The optimal vehicle to ensure adequate penetration of 5-aminolaevulinic acid (ALA) for its use in photodynamic therapy (PDT) of skin lesions has not been determined. Objectives We aimed to study the effects of ALA in various vehicle formulations [saline lotion with and without dimethylsulphoxide (DMSO), cream, liposomes and vaseline] after topical application in a murine subcutaneous adenocarcinoma model. Methods The effect of DMSO on porphyrin synthesis and ALA penetration through the skin was studied by measuring the uptake of 14C label from ALA, ALA and porphobilinogen accumulation, and some haem enzyme activities. The tissue distribution and kinetics of porphyrin synthesis after topical application of ALA entrapped in large multilamellar liposomes was also determined. Results ALA in saline lotion, alone or with 10% DMSO, proved to be the most efficient vehicle for tumour porphyrin accumulation (mean ± SD 1·75 ± 0·25 and 2·09 ± 0·39 µg g−1, respectively), whereas cream and liposomes induced lower levels and identical porphyrin accumulation (0·60 µg g−1). Using ALA + DMSO saline lotion, a higher porphyrin accumulation was found in skin overlying the tumour tissue and in the first 2 mm of tumour, probably due to increased ALA penetration, or greater interconversion to porphyrins, or greater retention of ALA and/or porphyrins. Conclusions These findings reinforce the importance of the vehicle in topical ALA-based PDT, and explain the mechanism of action of DMSO in enhancing protoporphyrin IX biosynthesis in superficial lesions.

    Notes: Abstract  Induced mutants in the barley cultivar Triumph have been screened for reduced dormancy. One line, which germinated readily 2 weeks after harvest, was classified as ABA-insensitive, since it could tolerate a ten-fold increase in ABA, compared to its parent, before germination was inhibited. This mutant, designated TL43, was genotypically similar to Triumph and phenotypically similar under Scottish growing conditions, except for a slightly reduced grain size. In Spain, it showed considerable reductions in both grain yield and plant height, suggesting that it was less widely adapted than its parent. Levels of α-amylase activity were increased at both sites. The mutant appeared to be different from those with ABA insensitivity or altered dormancy previously documented in either barley or Arabidopsis.
