    Description: Class D β-lactamases with carbapenemase activity are emerging as carbapenem-resistance determinants in Gram-negative bacterial pathogens, mostly Acinetobacter baumannii and Klebsiella pneumoniae. Carbapenemase activity is an unusual feature among class D β-lactamases, and the structural elements responsible for this activity remain unclear. Based on structural and molecular dynamics data, we previously hypothesized a potential role of the residues located in the short-loop connecting strands β5 and β6 (the β5–β6 loop) in conferring the carbapenemase activity of the OXA-48 enzyme. In this work, the narrow-spectrum OXA-10 class D β-lactamase, which is unable to hydrolyze carbapenems, was used as a model to investigate the possibility of evolving carbapenemase activity by replacement of the β5–β6 loop with those present in three different lineages of class D carbapenemases (OXA-23, OXA-24, and OXA-48). Biological assays and kinetic measurements showed that all three OXA-10–derived hybrids acquired significant carbapenemase activity. Structural analysis of the OXA-10loop24 and OXA-10loop48 hybrids revealed no significant changes in the molecular fold of the enzyme, except for the orientation of the substituted β5–β6 loops, which was reminiscent of that found in their parental enzymes. These results demonstrate the crucial role of the β5–β6 loop in the carbapenemase activity of class D β-lactamases, and provide previously unexplored insights into the mechanism by which these enzymes can evolve carbapenemase activity.

    Description: The 2016 Mw 6.5 earthquake struck a large region of the Apennines in central Italy and produced impressive and complex surface ruptures. These ruptures were investigated and mapped by field geologists for years after the earthquake thanks also the use of remote sensing data. Here we present detailed maps of the ground deformation pattern produced by the Vettore normal fault activated during the Mw 6.5 mainshock, using Differential Synthetic Aperture Radar Interferometry. The DInSAR analysis has been retrieved from ALOS-2 SAR data, via the Parallel Small BAseline Subsets (P-SBAS) algorithm. At the local scale, we automatically identify many surface ruptures, most of which already observed in the field. At the large scale, we trace a set of five geological cross-sections to inspect a possible link between the coseismic vertical displacement, the lithology distribution and the tectonic structures of the area (i.e., thrusts, normal faults). On these sections, we also project the seismicity distribution recorded during October 2016.The integration of such datasets allows the recognition of an important geological control in the overall distribution of the deformation, which shows maximum values in correspondence of the carbonatic multilayer and minimum values within the clastic succession. The distribution of seismicity allows also us to distinguish seismogenic by aseismic slip associated with fault ruptures.The results of this work demonstrate that the integration of surface geology, remote sensing data and seismicity, can lead to a better understanding of the influence of geological structures on the distribution of the ground deformation associated with earthquakes.

    Description: Starting from 24 August 2016, a long seismic sequence, including nine Mw 〉 5.0 earthquakes, struck a wide area of the Central Italy. A large amount of geological, geodetic, and seismological data envisages a complex system of NNW-SSE trending, seismogenic normal faults. These active tectonic structures are well known at the surface and consistent with previous seismotectonic studies. In order to improve the comprehension of the seismotectonic framework of this seismic sequence, we provide a novel reconstruction of the subsurface geology of the area close to the NorciaMw 6.5 mainshock (30 October 2016), based on previously unpublished seismic reflection profiles and available geological data. All the data have been synthesized along a 47 km long, WSW-ENE trending geological cross section, interpreted down to a depth of 12 km. Comparing the subsurface geological model with the available seismological data, we find that the majority of seismicity is confined within the sedimentary sequence and does not penetrate the underlying basement. The basement has been constrained at depths of 8 to 11 km and coincides with the cutoff of the seismicity. We have also traced the trajectories of the seismogenic normal faults activated during this seismic sequence, reconciling the high-angle (dip〉65°) normal faults exposed at the surface, with their angle (dip 〈 50°) at hypocentral depths. The results of this study may be useful for better understanding the rheological properties of the seismogenic rock volume, as well as the coseismic deformations of the topographic surface observed by geodetic techniques and field mapping.
