    Notes: Background  The skin represents an accessible somatic tissue for therapeutic gene transfer. The superficial lipophilic layer of the skin, the stratum corneum, however, constitutes a major obstacle to the cutaneous delivery of charged macromolecules such as DNA.Objectives  To determine whether silicon-based microneedles, microfabricated via a novel isotropic etching/BOSCH reaction process, could generate microchannels in the skin of sufficient dimensions to facilitate access of lipid : polycation : pDNA (LPD) nonviral gene therapy vectors.Methods  Scanning electron microscopy was used to visualize the microconduits created in heat-separated human epidermal sheets after application of the microneedles. Following confirmation of particle size and particle surface charge by photon correlation spectrocopy and microelectrophoresis, respectively, the diffusion of fluorescent polystyrene nanospheres and LPD complexes through heat-separated human epidermal sheets was determined in vitro using a Franz-type diffusion cell. In-vitro cell culture with quantification by flow cytometry was used to determine gene expression in human keratinocytes (HaCaT cells).Results  The diffusion of 100 nm diameter fluorescent polystyrene nanospheres, used as a readily quantifiable predictive model for LPD complexes, through epidermal sheets was significantly enhanced following membrane treatment with microneedles. The delivery of LPD complexes either into or through the membrane microchannels was also demonstrated. In both cases considerable interaction between the particles and the epidermal sheet was observed. In-vitro cell culture was used to confirm that LPD complexes mediated efficient reporter gene expression in human keratinocytes in culture when formulated at the appropriate surface charge.Conclusions  These studies demonstrate the utility of silicon microneedles in cutaneous gene delivery. Further studies are required to elucidate fully the influence of the physicochemical characteristics of gene therapy vectors, e.g. particle diameter and surface charge, on their diffusion through microchannels and to quantify gene expression in vivo.

When aldrin and dieldrin not added to Phaseolus cultures until 10 or 20d after subculture usual extent of conversion of aldrin to dieldrin, but reduced production of other metabolites, and growth inhibition negated. Increasing volumes of 2-methoxyethanol had detrimental effect on growth and uptake and metabolism. Dieldrin production maximal during rapid growth phase and probably independent of other conversions.

    Notes: Summary This paper provides an overview of one aspect of the differentiation of cartilage and bone, namely, the degree of control provided by the extracellular matrix and microenvironment. A brief review of the diagnostic features of cartilage and bone is followed by a discussion of stem cells, emphasizing how to identify them using cytochemical, ultrastructural or experimental procedures. The role of extracellular matrices in theinitiation of differentiation is discussed with referencu to the initiation of chondrogenesis in the vertebral skeleton of the embryonic chick and of osteogenesis in the mandibular skeletons of embryonic chick and mice. The role of extracellular matrices in themaintenance of the differentiated state is discussed with reference to the ability of chondrocytes to compensate for depletion of their extracellular matrices and to the maintenance of altered differentiated states in achondroplasia. Some emphasis is placed on the notion that skeletal cells can neither be considered nor studied in isolation. The epigenetic approach used in studies of growth and morphogenesis needs to be applied to studies on both the initiation and the maintenance of cytodifferentiation.

    Notes: Abstract A study has been made of the probability of exocytosis of synaptic vesicles at different varicosities in single sympathetic terminal axons in the mouse vas deferens. An antibody (SV2Ab) against SV2, a proteoglycan in synaptic vesicles, labelled an area of individual sympathetic varicosities that was slightly less than that occupied by dextran-rhodamine, previously orthogradely transported into the varicosities. In contrast plasma membrane bound protein syntaxin, found at active zones of motor nerve terminals, occupied an area of the varicosity that was approximately one-third that of SV2. This suggests that sympathetic varicosities possess specialized zones for exocytosis on their plasma membranes. Antibodies against the N-terminal sequence of synaptotagmin 1 (SNAb), a sequence exposed within synaptic vesicles, were used to determine the probability of exocytosis at different varicosities of single terminal branches. The area of SNAb labelling was not significantly different from that of the SV2 labelling, which implies vesicles that have undergone exocytosis may eventually return to the main pool of vesicles. Varicosities belonging to the same terminal axon, and identified with SV2Ab, showed different extents of labelling with SNAb when secretion was evoked with high potassium concentrations (80 mM) for 30 min in the presence of SNAb. There was up to an order of magnitude difference in the average intensity of SNAb labelling between different varicosities of the same terminal axon whereas there was little difference in the average intensity of SV2Ab labelling. These observations suggest that there is considerable variability in the probability of exocytosis at the specialized zones in different varicosities.
