    Notizen: While vaccines are effective in adults, they are less successful in newborns and infants. Neonatal unresponsiveness to vaccines could be owing to immaturity of lymphocytes and/or to inhibition by maternal antibodies. Unresponsiveness of newborn to vaccines can be overcame by genetic immunization. In the present study we investigated the effect of maternal antibodies on the anti-influenza virus protective response in progeny born to dams immunized with plasmid containing the hemagglutinin gene or UV-inactivated virus. The effect of maternal antibodies was studied in plasmid immunized F1 mice born to BALB/c dams, previously immunized with virus or plasmid and crossed with C57BL/6 males, as well as in offspring born to BALB/c dams immunized with plasmid and then immunized with UV-inactivated WSN virus. We have found that the inhibition period of the anti-HA antibody response in offspring born to dams immunized with DNA is shorter than that of offspring born to dams immunized with virus. Furthermore, there is a persistent inhibitory effect on B cells from offspring born to dams immunized with virus or injected with antiviral monoclonal antibodies (MoAb), after the decline of maternal antibody titers. The analysis of the haemagglutinin-specific clonotype reactivity pattern of offspring born to dams immunized with inactivated influenza virus or with a plasmid showed that clonotypes producing antibodies specific for the immunizing virus strain were predominant in offspring born to dams immunized with DNA compared to those born to dams immunized with virus. Maternal antibodies do not affect cell-mediated immunity. These findings might be used to design efficient vaccination schedules for newborns and infants.

    Notizen: Background Allergen-specific immunotherapy (SIT) leads to long-term amelioration of T-helper type 2 (Th2)-mediated allergic symptoms and is therefore recommended as a first line therapy for allergies. The major disadvantage of SIT is its low efficiency, requiring treatment over years.Objective In this study, we evaluated the potential of Toll-like receptor (TLR) ligands to facilitate Th1-type immune responses.Methods The immunogenicity and therapeutic potential of the major bee venom allergen phospholipase A2 (PLA2) combined with various TLR ligands were tested in mice and compared with immune responses induced by conventional aluminium-based preparations.Results Regarding total IgG against PLA2, TLR2/4-binding lipopolysaccharide and TLR3-binding polyriboinosinic polyribocytidylic (PolyI:C) were the superior adjuvants for prophylactic vaccination. However, TLR9-binding phosphorothioate-modified cytosine–guanosine-rich oligonucleotide (CpG), TLR-3-binding PolyI:C, and TLR2/6-binding peptidoglycan skewed the immune responses more towards IgG2a isotype and Th1 cytokines. Furthermore, in a therapeutic approach, CpG, PolyI:C and TLR7/8-binding 3M003 had immune modulating properties as they suppressed established IgE titres.Conclusion The potential of TLR ligands to adjuvate the immunogenicity of bee venom PLA2 and to skew the Th1–Th2 balance proved very heterogeneous. With respect to SIT, CpG, PolyI:C, and 3M003 were very promising. Hence, TLR ligands should be considered as adjuvants or immune modulators in SIT in human as to improve its efficiency regarding the Th1–Th2 balance of the immune response with a likely effect on therapy duration.

The effects of some relevant electrochemical parameters (the electrolyte, the polymerization current density, and the deposition time) on the reflectance spectra of polypyrrole are reported. The optical constants for polypyrrole films were calculated from the reflectance spectra by Kramers–Kronig technique. The decrease of the electropolymerization current density and film thickness results in an increase of the reflectance and dc conductivity values for PPY(TSO).

    Notizen: Abstract Twenty-five children (23 girls and 2 boys) with central precocious puberty were treated with the LH-RH agonistd-Ser (TBU)6-LHRH (1–9) EA (HOE) 766, Buserelin) by daily subcutaneous injection for a period of 11–18 months. Eight girls and 2 boys previously treated with cyproterone acetate (CPA, 100–150 mg/m2 body surface per day) and the first seven newly diagnosed patients received 2×10 μg Buserelin/kg bodyweight per day for 1 week, followed by a maintenance therapy of 1×10 μg/kg per day. After an initial marked increase, oestrogen (E2) serum levels in girls and testosterone (T) values in boys decreased. After a treatment period of 6–20 weeks the patients received 2×20 μg Buserelin/kg per day for 1 week and thereafter a maintenance dosage of 20 μg/kg per day to obtain full suppression (i.e. E2〈50 pmol/l; T〈1 nmol/l). The remaining eight patients started directly on 2×20 μg Buserelin/kg per day followed by 1×20 μg/kg per day. All eight girls with menarche before therapy had no further menses. In all girls there was a reduction of palpable breast tissue. Decrease of pubic hair development was observed in 3 girls, an increase was seen in 5 girls, whereas in the remaining 15 girls no change was observed. Both boys had a reduction of testicular volume and of pubic hair. The basal growth rate during the half year preceding Buserelin treatment was elevated in the newly diagnosed patients as compared to the growth rate of the patients previously treated with CPA and to the median growth rate of healthy children. During the first 6 months of treatment the mean growth rate decreased slightly in the patients who had received no previous treatment and increased in the patients previously treated with CPA. During the subsequent year a marked fall of the mean growth rate was observed. Paired comparison of the mean SDS (height/chronological age and height/bone age) and of the mean final height prediction (according to Bayley and Pinneau) of 15 children treated for 18 months revealed no significant change. We conclude that during an observation period of 18 months the daily subcutaneous administration of the LH-RH agonist Buserelin has been a safe and effective method of selectively inhibiting gonadal activity in children with central precocious puberty. With prolonged treatment an improvement of the final height prediction is expected.

    Notizen: Abstract Sixteen girls with Turner syndrome (TS) were treated for 4 years with biosynthetic growth hormone (GH). The dosage was 4IU/m2 body surface s.c. per day over the first 3 years. In the 4th year the dosage was increased to 61 U/m2 per day in the 6 girls with a poor height increment and in 1 girl oxandrolone was added. Ethinyl oestradiol was added after the age of 13. Mean (SD) growth velocities were 3.4 (0.9), 7.2 (1.7), 5.3 (1.3), 4.3 (2.0) and 3.6 (1.5) cm/year before and in the 1st, 2nd, 3rd and 4th year of treatment. Skeletal maturation advanced faster than usual in Turner patients especially in the youger children. Although the mean height prediction increased by 5.6 cm and 11 of the 16 girls have now exceeded their predicted height, the height of the 4 girls who stopped GH treatment exceeded the predicted adult height by only 0 to 3.4 cm.
