    Notes: Common statistics of turbulent electrostatic fluctuations observed at the plasma edge and scrape-off layer are analyzed in three tokamak devices that have different configurations. The statistics of experimental data collected using fixed sampling time is the same than the statistics of the time for which the oscillation return to a specified reference interval of values. This observation, in addition to the finding of power-scaling laws for some average quantities with respect to either the sampling time or the size of the reference interval, suggests that turbulence on tokamaks have recurrent characteristics, typical of a recurrent chaotic low-dimensional system. Furthermore, the first Poincaré recurrence time and other dynamical tools are used to simulate the mentioned fluctuation statistical properties. © 2001 American Institute of Physics.

    Notes: Abstract Post-exercise proteinuria is a common phenomenon in healthy subjects. Previous studies have used albumin (Alb) and β2-microglobulin (β2-m) molecules as representatives of high- and low-molecular-weight proteins. Recently, more specific markers of the human kidney proximal tubule have been used to identify the precise site of alterations. Active male subjects underwent two strenuous runs, one 400-m run and one 3000-m run. Urine was collected from the subjects before and after each event. Total protein (TP), Alb, α1-microglobulin (α1-m), β2-m, intestinal alkaline phosphatase (IAP), tissue-nonspecific alkaline phosphatase (TNAP) and N-acetyl-β-d-glucosaminidase (NAG) were determined for each sample. The short-distance run (400 m) resulted in the largest increases (P ≤ 0.05) in TP (31-fold), Alb (100-fold) and β2-m (164-fold) as compared to the long-distance run (3000-m). The α1-m excretion rates were increased to a lesser extent by the exercises. The IAP activity was slightly increased (+90%) by the 400-m run while the TNAP and NAG activities showed a 6.8-fold and a 3.6-fold increase, respectively, after this event. Smaller increases were recorded for the long-distance run (P = 0.05). To conclude, the present investigation showed that: (1) post-exercise proteinuria is related to the absolute intensity of exercise; (2) the impairment of protein reabsorption is revealed better by changes in Alb and β2-m; (3) changes in TNAP and NAG activities could reveal biochemical modifications that occur in the proximal tubule, particularly at the S1-S2 segment.

    Description: This study constitutes a preliminary assessment of probabilistic tsunami inundation in the NE Atlantic region. We developed an event-tree approach to calculate the likelihood of tsunami flood occurrence and exceedance of a specific near-shore wave height for a given exposure time. Only tsunamis of tectonic origin are considered here, taking into account local, regional, and far-field sources. The approach used here consists of an event-tree method that gathers probability models for seismic sources, tsunami numerical modeling, and statistical methods. It also includes a treatment of aleatoric uncertainties related to source location and tidal stage. Epistemic uncertainties are not addressed in this study. The methodology is applied to the coastal test-site of Sines located in the NE Atlantic coast of Portugal. We derive probabilistic high-resolution maximum wave amplitudes and flood distributions for the study test-site considering 100- and 500-year exposure times. We find that the probability that maximum wave amplitude exceeds 1 m somewhere along the Sines coasts reaches about 60 % for an exposure time of 100 years and is up to 97 % for an exposure time of 500 years. The probability of inundation occurrence (flow depth 〉0 m) varies between 10 % and 57 %, and from 20 % up to 95 % for 100- and 500-year exposure times, respectively. No validation has been performed here with historical tsunamis. This paper illustrates a methodology through a case study, which is not an operational assessment.

    Description: Probabilistic Tsunami Hazard Assessment requires the existence of a synthetic earthquake catalogue obeying the geodynamic framework of the source area and a fault model for each event. Usually, we assume a constant fault displacement, eventually filtered by a simple closure condition. The distribution of events by magnitude classes follows a truncated Gutenberg-Richter law with epicentres uniformly distributed in the source domain. This approach makes that the slip on the source structures will extend beyond the generation domain being heterogeneous inside that domain. This contradicts the assumption that the slip is caused by geodynamic constraints. Here, we propose a methodology for creating synthetic earthquake catalogues and we apply it to the Gloria Fault in the Atlantic. With three instrumental tsunamis recorded: 08.05.1939, 25.11.1941 and 26.05.1975 [1]. Additionally, we propose a semi-empirical scaling law for the generation of the tsunamigenic earthquakes

    Description: The Rho GTPase Cdc42 coordinates regulation of the actin and the microtubule cytoskeleton by binding and activating the Wiskott–Aldrich syndrome protein. We sought to define the role of intrinsic expression of Cdc42 by mature B cells in their activation and function. Mice with inducible deletion of Cdc42 in mature B cells formed smaller germinal centers and had a reduced Ab response, mostly of low affinity to T cell–dependent Ag, compared with wild-type (WT) controls. Spreading formation of long protrusions that contain F-actin, microtubules, and Cdc42-interacting protein 4, and assumption of a dendritic cell morphology in response to anti-CD40 plus IL-4 were impaired in Cdc42-deficient B cells compared with WT B cells. Cdc42-deficient B cells had an intact migratory response to chemokine in vitro, but their homing to the B cell follicles in the spleen in vivo was significantly impaired. Cdc42-deficient B cells induced a skewed cytokine response in CD4 + T cells, compared with WT B cells. Our results demonstrate a critical role for Cdc42 in the motility of mature B cells, their cognate interaction with T cells, and their differentiation into Ab-producing cells.

    Description: The method presented here aims to assess the tsunami threat very rapidly after the occurrence of a large earthquake, using as input the parameters of the seismic source, and an approach based on Green's summation. We show that the main weakness of the approach (the need to consider only linear shallow water propagation) is largely compensated by the advantages in terms of computing performance and independence with respect to pre-computed scenarios. To test the approach and to illustrate its implementation in a real environment, we focus on the Sea of Oman, a tsunamigenic area characterized by Makran subduction zone which detailed structure is partially unknown and where secondary tsunami sources must also be taken into account, both for hazard studies and warning purposes. The potential source area is partitioned into a grid of unity water sources. A shallow water (SW) numerical model is used to pre-compute the corresponding empirical Green's functions on several points of interest located on the coasts of Iran, Pakistan and Oman. The comparison between Green's summation and the direct SW computation using the full resolution of the bathymetric grid shows that the accuracy is good enough for practical applications.

    Description: Purpose: Glucocorticoids are part of the therapeutic armamentarium of chronic lymphocytic leukemia (CLL) where it has been suggested that cells with unmutated IGHV genes exhibit higher sensitivity. The mechanisms by which glucorticoids are active in CLL are not well elucidated. We aimed to ascertain the activity of dexamethasone in CLL cells according to prognosis and to identify the molecular mechanisms that are influencing the response to this drug. Experimental Design: Sensitivity to dexamethasone was analyzed ex vivo in 50 CLL and compared according to IGHV mutational status and/or ZAP-70 expression. The response was further compared by gene expression profiling (GEP) of selected cases. Expression of genes of interest was validated by quantitative reverse transcriptase PCR. Results: Response to dexamethasone was higher in cases with unmutated IGHV/high ZAP-70 expression, and the levels of induction of the pro-apoptotic Bim protein correlated with the degree of cell death. GEP analysis showed few genes differentially expressed after dexamethasone treatment between mutated and unmutated cases. However, functional annotation analysis showed that unmutated cases had significant enrichment in terms related to apoptosis. Specific analysis of genes of interest conducted in a large series disclosed that in unmutated IGHV cells, FKBP5 expression was higher at baseline and after dexamethasone exposure and that GILZ was more induced by dexamethasone treatment in these cases. Conclusions: Unmutated IGHV/high ZAP-70 CLL cells exhibit better response to dexamethasone treatment, which is accompanied by a differential expression of genes involved in the glucocorticoid receptor pathway and by an increased induction of genes related to apoptosis.     Description: The Wiskott-Aldrich syndrome protein (WASP) is a key cytoskeletal regulator of hematopoietic cells. Although WASP-knockout (WKO) mice have aberrant B-cell cytoskeletal responses, B-cell development is relatively normal. We hypothesized that N-WASP, a ubiquitously expressed homolog of WASP, may serve some redundant functions with WASP in B cells. In the present study, we generated mice lacking WASP and N-WASP in B cells (conditional double knockout [cDKO] B cells) and show that cDKO mice had decreased numbers of follicular and marginal zone B cells in the spleen. Receptor-induced activation of cDKO B cells led to normal proliferation but a marked reduction of spreading compared with wild-type and WKO B cells. Whereas WKO B cells showed decreased migration in vitro and homing in vivo compared with wild-type cells, cDKO B cells showed an even more pronounced decrease in the migratory response in vivo. After injection of 2,4,6-trinitrophenol (TNP)–Ficoll, cDKO B cells had reduced antigen uptake in the splenic marginal zone. Despite high basal serum IgM, cDKO mice mounted a reduced immune response to the T cell–independent antigen TNP-Ficoll and to the T cell–dependent antigen TNP–keyhole limpet hemocyanin. Our results reveal that the combined activity of WASP and N-WASP is required for peripheral B-cell development and function.
