    Notes: The carbon monoxide–water (c–w) cluster produced in an argon matrix at cryogenic temperatures has been reinvestigated by FTIR spectroscopy. In the 4.7 μm region, a line at 2149.4 cm−1 was assigned to the CO stretching mode of the CO–H2O 1–1 cluster. In the 2.7 μm region, two lines, at 3627.8 and 3723.5 cm−1, were assigned to the OH stretching modes of the CO–H2O 1–1 cluster. By the deuterium enriched experiment, the lines of the CO–D2O 1–1 cluster and the CO–HDO 1–1 cluster were identified for the CO stretching vibration and the OD stretching vibration. The tunnel splitting observed in gas phase has not been detected in the present study. Combining with the gas phase data the matrix shifts in the line positions were obtained precisely, with which we have predicted some band centers of the free complexes so far not reported in literature. Migration of trapped molecules, CO and H2O, in the argon matrix has been observed both by annealing and by high temperature deposition. The spectral behavior shows that CO can move at a lower temperature than H2O in the argon matrix. © 2001 American Institute of Physics.

    Notes: Using a combined method of pulse-labelling trees and analysing detailed distribution of 13C tracer within tree rings, we studied how photo-assimilates incorporated on a given day are then distributed in a tree ring. A branch of a 4-year-old Cryptomeria japonica D.Don tree growing in Tsukuba, Japan was pulse-labelled with non-radioactive 13CO2 on two occasions: 29 May 2001 and 18 September 2001. Two discs were cut from the stem on 4 March 2002, one immediately under and the other 0.5 m below the branch and put through high-resolution δ13C analysis. δ13C peaks were observed in both the earlywood and latewood of the concerned tree ring, corresponding to each pulse-labelling date. The earlywood peaks was broader than the latewood peaks, possibly reflecting seasonal variation of the width of wood developing zone. Half-widths of the peaks were measured and used as indicators for the potential time resolution of tree-ring isotope analysis. The half-widths of the peaks indicated a time resolution no finer than 8.7–28 and 33–42 d in the early and latewood, respectively. Holocellulose extraction yielded only a slight change to the shape of the δ13C peaks. 13C tracer pulse-labelled in May and September reached tangentially different locations in the lower disc, suggesting a seasonal change in the pathway of carbohydrates. Local consumption of spring assimilates and long-distance downward transport of autumn assimilates were also suggested.

    Notes: Abstract Winged bean Kunitz chymotrypsin inhibitor (WCI) accumulates abundantly in seeds and tuberous roots, and small amounts of the WCI protein and mRNA can also be detected in stems. In this study, we analyzed the localization of the WCI protein in stems of winged bean. The results demonstrated that the WCI protein was localized in sieve tubes. Furthermore, we showed that the 5′ region of the WCI-3b gene, which exhibited strong transcriptional activity in developing seeds, also promoted transcription of a reporter gene in the phloem of stems of transgenic tobacco.

    Notes: Abstract This study investigated the relation between the resting potential of a 2-year-old Japanese oak (Quercus crispula Blume) sapling and its physiology, especially the radial transport of water containing inorganic ingredients in the stem using the scanning electron microscope-energy dispersive X-ray microanalyzer (SEM-EDXA) method. The resting potential of a sapling could be monitored continuously with our measuring apparatus. Changes in resting potential were due to the light. The hyperpolarization and depolarization peaks of the resting potential, whose absolute voltage was about 10mV, occurred right after lights-off and lights-on, respectively. The resting potential was found to show periodic responses for each day unit. At night (lights-off), the resting potential tended to depolarize with an increase in tangential strain. On the other hand, during the daytime (lights-on) the resting potential tended to hyperpolarize, depolarize, or show a nearly constant value for the tangential strain. The water containing inorganic ingredients was transported, via transpiration, in both directions between the mature xylem zone and the phloem zone through differentiating xylem cells. This water transport within a tree stem had a significant effect on the diurnal changes in resting potential.

    Notes: Abstract In the present study, we purified a neutral thiol proteinase from dog PMN leukocytes and indicated that the proteinase elaborated the chemotactic factor for lymphocytes by cleavage of IgG. The neutral thiol proteinase was purified about 744-fold by ion-exchange chromatographies and affinity chromatography, and the final preparation was over 70% pure. After incubation of dog IgG with the proteinase, three distinct protein peaks were seen by the gel filtration on Sephadex G-200. Only the third peak, perhaps a dialyzable peptide, showed a significant chemotactic activity for dog lymphocytes.

    Description: Aromatase inhibitors (AIs) are considered the gold standard of endocrine therapy for estrogen receptor positive postmenopausal breast cancer patients. AI treatment was reported to result in marked alterations of genetic profiles in cancer tissues but its detailed molecular mechanisms have not been elucidated. Therefore, we profiled miRNA expression before and after the treatment with letrozole in MCF-7 co-cultured with primary breast cancer stromal cells. Letrozole significantly altered the expression profiles of cancer miRNAs in vitro . Among the elevated miRNAs following letrozole treatment, computational analysis identified let-7f , a tumor-suppressor miRNA which targeted the aromatase gene ( CYP19A1 ) expression. Quantitative real time PCR assay using MCF-7 and SK-BR-3 cells as well as clinical specimens of neoadjuvant study demonstrated a significant inverse correlation between aromatase mRNA and let-7f expression. In addition, high let-7f expression was significantly correlated with low aromatase protein levels evaluated by both immunohistochemistry and western blotting method in breast cancer cases. Results of 3'UTR luciferase assay also demonstrated the actual let-7f binding sites in CYP19A1 , indicating that let-7f directly targets the aromatase gene. Subsequent WST-8 and migration assays performed in let-7f transfected MCF-7 and SK-BR-3 cells revealed significant decrement of their proliferation and migration. These findings all demonstrated that let-7f , a tumor suppressor miRNA in breast cancer, directly targeted the gene and were restored by AI treatment. Therefore, AI may exert tumor-suppressing effects upon breast cancer cells by suppressing aromatase gene expression via restoration of let-7f .
