    Abstract: Xanthium sibiricum is a well-known traditional Chinese medicine (TCM) that has been commonly used to treat rhinitis and related nasal diseases. The aim of this study was to develop a comprehensive analytical method based on high-performance liquid chromatographyelectrospray ionization coupled with triple quadrupole-linear ion trap mass spectrometry (LC-ESIQTRAP- MS/MS) for the simultaneous determination of phenolic acids, anthraquinones, and flavonoids in the aerial part and fruit of Xanthium sibiricum. Methods: The separation was completed on Agilent ZORBAX SB-C18 column (250 × 4.6 mm, 5μm) using methanol and 0.2% (v/v) aqueous formic acid as the mobile phase. The target components were analyzed in negative ion mode with accurate and sensitive multiple reaction monitoring (MRM) mode. Results: The correlation coefficients of all the calibration curves were higher than 0.9994. Relative standard deviations of intra- and inter-day precisions of the eighteen components were all lower than 2.87% and the recoveries were in the range from 97.73% to 101.82%. The validated method was successfully applied to possess forty Xanthium sibiricum samples (Xanthii Herba, Xanthii Fructus, and processed Xanthii Fructus) collected from different places in P. R. China. Furthermore, principal component analysis (PCA) was performed to evaluate and classify the samples according to the contents of the eighteen bioactive components. Conclusion: All the results demonstrated that the developed method was useful and could be applied for the overall assessment of the quality of Xanthii Herba and Xanthii Fructus.

    Abstract: MYH9 has dual functions in tumors. However, its role in inducing tumor stemness in hepatocellular carcinoma (HCC) is not yet determined. Here, we found that MYH9 is an effective promoter of tumor stemness that facilitates hepatocellular carcinoma pathogenesis. Importantly, targeting MYH9 remarkably improved the survival of hepatocellular carcinoma-bearing mice and promoted sorafenib sensitivity of hepatocellular carcinoma cells in vivo. Mechanistic analysis suggested that MYH9 interacted with GSK3β and reduced its protein expression by ubiquitin-mediated degradation, which therefore dysregulated the β-catenin destruction complex and induced the downstream tumor stemness phenotype, epithelial–mesenchymal transition, and c-Jun signaling in HCC. C-Jun transcriptionally stimulated MYH9 expression and formed an MYH9/GSK3β/β-catenin/c-Jun feedback loop. X protein is a hepatitis B virus (HBV)-encoded key oncogenic protein that promotes HCC pathogenesis. Interestingly, we observed that HBV X protein (HBX) interacted with MYH9 and induced its expression by modulating GSK3β/β-catenin/c-Jun signaling. Targeting MYH9 blocked HBX-induced GSK3β ubiquitination to activate the β-catenin destruction complex and suppressed cancer stemness and EMT. Based on TCGA database analysis, MYH9 was found to be elevated and conferred poor prognosis for hepatocellular carcinoma patients. In clinical samples, high MYH9 expression levels predicted poor prognosis of hepatocellular carcinoma patients. These findings identify the suppression of MYH9 as an alternative approach for the effective eradication of CSC properties to inhibit cancer migration, invasion, growth, and sorafenib resistance in HCC patients. Our study demonstrated that MYH9 is a crucial therapeutic target in HCC.

    Abstract: Magnoliae Officinalis Cortex is a well-known traditional Chinese herbal medicine with vast clinical consumption owing to its positive effects. However, little attention has been devoted to analyzing the dynamic changes of the chemical constituents of Magnoliae Officinalis Cortex in different growth periods. Methods: In this study, all analyses were performed on UFLC -20ADXR system and a SynergiTM Hydro-RP 100 Å column (100 mm×2.0 mm, 2.5µm). The mobile phase consisted of water containing 0.1% formic acid (A) and acetonitrile containing 0.1% formic acid (B). The target constituents, including two lignans, two alkaloids, two flavonoids, one phenylpropanoid glycoside, and one organic acid, were analyzed in both positive and negative ion modes with accurate and sensitive multiple reaction monitoring (MRM) mode. Results: The correlation coefficients of all the calibration curves were higher than 0.9992. Relative standard deviations of intra- and inter-day precisions of the eight analytes were all lower than 4.01% and the recoveries were in the range from 98.62% to 102.46%. Grey relational analysis was performed to evaluate the samples according to the contents of 8 constituents. The results showed that the quality of Magnoliae Officinalis Cortex collected at traditional harvest time was much better, and the higher the age, the better the quality. Conclusion: The proposed method is useful for the assessment on the quality of Magnoliae Officinalis Cortex, and this study provides the basis for exploring the quality forming mechanism of Magnoliae Officinalis Cortex medicinal materials and choosing the suitable harvesting period.

    Abstract: With the rapid and vigorous growth of forest tourism, the irresponsible environmental behavior of tourists has caused enormous strain on forests’ ecological systems. Carrying out environmental education in forest parks is conducive to promoting the sustainable development of forest tourism. To explore the impact of human–place emotion on environmental education effects, this study took Fuzhou National Forest Park as an example to construct a structural equation model composed of landscape perception, environment interpretation, place attachment, and the effects of environmental education (EEE). The relationship between the four elements and the mechanism of action was clarified. A questionnaire was used with 480 visitors. Statistical analysis showed that: (1) The value of scientific research and education (0.774) influences landscape perception. Reliability (0.770) and tangibility (0.718) contribute to environmental interpretation. Place identification and dependence are represented by environmental identity (0.771) and are activity-dependent (0.792), respectively. Knowledge (0.860) and behavior (0.869) are essential factors in driving the EEE. (2) Place attachment and environment interpretation had a significant positive impact on the environmental education effect (p 〈 0.001), and there was no direct effect between landscape perception and EEE. (3) Landscape perception and environmental interpretation indirectly influence EEE with place attachment as full and partial mediators, respectively. This paper aims to provide theoretical support for better synergistic growth of forest park ecology, economy, and environment.

    Abstract: Prunellae Spica (PS) is an important traditional Chinese herbal medicine (TCM) with massive consumption because of its therapeutical and seasoning effects. However, little attention has been paid to simultaneous analyze its bioactive components for quality control of PS based on its different growing habitats and different growth periods. In this study, the quality of PS was evaluated based on simultaneous determination of multiple bioactive components combined with grey relational analysis (GRA). A reliable method based on high-performance liquid chromatography tandem triple quadrupole mass spectrometry (HPLC-QTRAP-MS/MS) was established to simultaneously determine the contents of 21 components in PS, including 9 phenolic acids, 3 coumarins, 8 flavonoids and 1 pentacyclic triterpene. Furthermore, GRA was performed to evaluate the quality of PS samples according to the contents of these 21 components. The results showed that the quality of PS harvested on withering period, cultivated in Anhui was better than that of other PS samples. The proposed method is useful for the overall assessment on the quality of PS, and this study provides valuable information for revealing the dynamic change laws of metabolite accumulation in PS and choosing the most suitable harvesting time and genuine producing area of PS to obtain the best quality.

    Abstract: To explore rapidly the potential chemical markers for differentiating Pseudostellariae Radix from different cultivated fields and germplasms, a method is proposed based on ultra-performance liquid chromatography-triple time-of-flight mass/mass spectrometry (UPLC-Triple TOF-MS/MS) coupled with multivariate statistical analysis. Peak matching, peak alignment, and noise filtering were used in analyzing mass spectrometric data. Accurate m/z value analysis of MS data based on software of database search and MS/MS fragment analysis were applied to identify constituents. The obtained data were statistically analyzed with hierarchical cluster analysis (HCA), principal component analysis (PCA), and partial least squared-discriminant analysis (PLS-DA) to compare the differences among these samples. The PLS-DA loading plot obtained from all mass data showed that 21 compounds were identified as the potential chemical markers to characterize the samples. The results provide experimental data to reveal the influence of ecological environments and germplasms on metabolite biosynthesis in Pseudostellariae Radix.
