    Abstract: Pulmonary right-to-left shunt (P-RLS) and patent foramen ovale right-to-left shunt (PFO-RLS) often appear in combination, and there are often differences and connections between them. Intrapulmonary arteriovenous anastomoses (IPAVAs), as part of P-RLS, are often overlooked because there are no technologies to detect and identify them. This study aimed to further clarify the incidence and characteristics of P-RLS with the help of contrast transesophageal echocardiography (c-TEE) and contrast transthoracic echocardiography (c-TTE), providing a reference for clinically relevant research and patent foramen ovale (PFO) management disposal decisions. Methods We retrospectively investigated 414 subjects who came to our hospital for c-TEE from October 2021 to July 2022, and all subjects completed c-TTE simultaneously. 7 Patients who were newly diagnosed with an atrial septal defect were excluded. Eventually, 407 patients were included in this study. Among them, 157 patients with PFO (58 patients were treated with PFO closure subsequently) and 250 patients without PFO confirmed by c-TEE were finally enrolled. In the process, we observed and analysed the presence of P-RLS. Results A total of 407 patients were included in the final analysis and divided into PFO group ( N  = 157) and non-PFO group ( N  = 250) according to the results of c-TEE. Whether at rest or after Valsalva maneuver, the incidence of P-RLS was significantly higher under c-TEE than under c-TTE in the two groups ( P   & lt; 0.001). For both c-TTE and c-TEE, the incidence of P-RLS was slightly higher after Valsalva maneuver than at rest, but the difference was not significant (c-TTE: rest vs. Valsalva maneuver, P  = 0.214; c-TEE: rest vs. Valsalva maneuver, P  = 0.076). The Valsalva maneuver increased the incidence of P-RLS in the group without PFO, which was more significant in c-TEE (c-TTE: rest vs. Valsalva maneuver, P  = 0.591; c-TEE: rest vs. Valsalva maneuver, P  = 0.008). In both groups, the P-RLS semiquantitative grading was statistical significance under different states and examinations ( P   & lt; 0.001). Conclusion The vast majority of P-RLS are grade 1–2 and are derived from physiological IPAVAs. Even so, attention should be given to the differentiation between P-RLS and PFO-RLS. c-TEE is an effective method to detect P-RLS; however, the recruitments of c-TEE and Valsalva maneuver to P-RLS should be noted.

    Abstract: The novel severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) pandemic has been posing a severe threat to global public health. Although broadly neutralizing antibodies have been used to prevent or treat corona virus disease 2019 (COVID-19), new emerging variants have been proven resistant to these antibodies. Methods In this study, we isolated receptor binding domain (RBD)-specific memory B cells using single-cell sorting method from two COVID-19 convalescents and expressed the antibody to test their neutralizing activity against diverse SARS-CoV-2 variants. Then, we resolved antibody-RBD complex structures of potent RBD-specific neutralizing antibodies by X-ray diffraction method. Finally, we analyzed the whole antibody repertoires of the two donors and studied the evolutionary pathway of potent neutralizing antibodies. Results and discussion We identified three potent RBD-specific neutralizing antibodies (1D7, 3G10 and 3C11) from two COVID-19 convalescents that neutralized authentic SARS-CoV-2 WH-1 and Delta variant, and one of them, 1D7, presented broadly neutralizing activity against WH-1, Beta, Gamma, Delta and Omicron authentic viruses. The resolved antibody-RBD complex structures of two antibodies, 3G10 and 3C11, indicate that both of them interact with the external subdomain of the RBD and that they belong to the RBD-1 and RBD-4 communities, respectively. From the antibody repertoire analysis, we found that the CDR3 frequencies of the light chain, which shared high degrees of amino acid identity with these three antibodies, were higher than those of the heavy chain. This research will contribute to the development of RBD-specific antibody-based drugs and immunogens against multiple variants.

    Abstract: Dynamic response analysis has potential for increasing fatigue life of the components in the transmission of a multistage main transmission system. The calculated data can demonstrate the influence of shaft torsional stiffness on dynamic characteristics of the system. Detecting key shafts of the system and analyzing their sensitivity are important for the design of four-stage helicopter gear box. Lumped mass method is applied for dynamic modeling and Fourier method is used to solve differential equation of the system. Results of the analysis indicate that key shafts can be designed carefully to improve the performance of the transmission system.

    Abstract: We shall investigate the asymptotic stability, toward a nonlinear wave, of the solution to an outflow problem for the one-dimensional compressible Navier–Stokes–Poisson equations. First, we construct this nonlinear wave which, under suitable assumptions, is the superposition of a stationary solution and a rarefaction wave. Then it is shown that the nonlinear wave is asymptotically stable in the case that the initial data are a suitably small perturbation of the nonlinear wave. The main ingredient of the proof is the [Formula: see text]-energy method that takes into account both the effect of the self-consistent electrostatic potential and the spatial decay of the stationary part of the nonlinear wave.

    Abstract: Endothelial-to-mesenchymal transition (EndMT) is a pathophysiological change in the vascular endothelium commonly seen in the cardiovascular system. Elevated serum Growth differiention factor 15 (GDF15) has been reported in VTE patients, but the relationship and mechanism between GDF15, EndMT and VTE are still unclear. Methods We performed a retrospective clinical study, and human serum GDF15 expression levels were detected. The mouse DVT model was established through subtotal ligation of the mouse inferior vena cava, and then we detected intimal changes and thrombi in the stenotic inferior vena cava by haematoxylin-eosin (HE) staining, Masson staining, and Sirius Red staining. The expression levels of GDF15 and SM22 were detected by immunohistochemistry and RT‒qPCR. Serum samples of mice were collected, and the expression level of GDF15 in serum was detected. Human umbilical vein endothelial cells (HUVECs) were stimulated with a cytokine mixture (TGF-β1 + TNF-α + IL-1β). The role and mechanism of GDF15 in EndMT and VTE were detected in HUVECs and in a DVT mice model. Results We found that serum GDF15 levels in both VTE patients and mouse DVT models were higher than those in the control group. EndMT was increased in the stenotic vascular tissue of mice. Further experiments showed that GDF15 could promote the EndMT of HUVECs and reduce their anticoagulation and antifibrinolytic ability through the smad2/p-smad2/snail pathway. Inhibition of mature GDF15 release can significantly reduce venous thrombotic fibre deposition in mice. Conclusions GDF15 positively promotes EndMT through activation of the Smad2/psmad2/snail pathway, and inhibition of GDF15 expression can alleviate the EndMT process, further improving the coagulation and fibrinolytic function of endothelial cells and thus reducing the local fibre deposition of venous thrombi.

    Abstract: Rodent outbreak is the main biological disaster in grassland ecosystems. Traditional rodent damage monitoring approaches mainly depend on costly field surveys, e.g., rodent trapping or hole counting. Integrating an unmanned aircraft system (UAS) image acquisition platform and deep learning (DL) provides a great opportunity to realize efficient large-scale rodent damage monitoring and early-stage diagnosis. As the major rodent species in Inner Mongolia, Brandt’s voles (BV) ( Lasiopodomys brandtii ) have markedly small holes, which are difficult to identify regarding various seasonal noises in this typical steppe ecosystem. Methods In this study, we proposed a novel UAS-DL-based framework for BV hole detection in two representative seasons. We also established the first bi-seasonal UAS image datasets for rodent hole detection. Three two-stage (Faster R-CNN, R-FCN, and Cascade R-CNN) and three one-stage (SSD, RetinaNet, and YOLOv4) object detection DL models were investigated from three perspectives: accuracy, running speed, and generalizability. Results Experimental results revealed that: 1) Faster R-CNN and YOLOv4 are the most accurate models; 2) SSD and YOLOv4 are the fastest; 3) Faster R-CNN and YOLOv4 have the most consistent performance across two different seasons. Discussion The integration of UAS and DL techniques was demonstrated to utilize automatic, accurate, and efficient BV hole detection in a typical steppe ecosystem. The proposed method has a great potential for large-scale multi-seasonal rodent damage monitoring.
