    Abstract: Rheum officinale Baill. is an important traditional Chinese medicinal herb, its dried roots and rhizomes being widely utilized to cure diverse diseases. However, previous studies mainly focused on the active compounds and their pharmacological effects, and the molecular mechanism underlying the biosynthesis of these ingredients in R. officinale is still elusive. Here, we performed comparative transcriptome analyses to elucidate the differentially expressed genes (DEGs) in the root, stem, and leaf of R. officinale. A total of 236,031 unigenes with N50 of 769 bp was generated, 136,329 (57.76%) of which were annotated. A total of 5884 DEGs was identified after the comparative analyses of different tissues; 175 and 126 key enzyme genes with tissue-specific expression were found in the anthraquinone, catechin/gallic acid biosynthetic pathway, respectively, and some of these key enzyme genes were verified by qRT-PCR. The phylogeny of the PKS III family in Polygonaceae indicated that probably only PL_741 PKSIII1, PL_11549 PKSIII5, and PL_101745 PKSIII6 encoded PKSIII in the polyketide pathway. These results will shed light on the molecular basis of the tissue-specific accumulation and regulation of secondary metabolites in R. officinale, and lay a foundation for the future genetic diversity, molecular assisted breeding, and germplasm resource improvement of this essential medicinal plant.

    Abstract: Patients with tumors generally present with accompanying activation of the coagulation system, which may be related to tumor stage. To our knowledge, few studies have examined the activation of the coagulation system in reference to lymph node metastasis within gastric cancer. This study aimed to investigate the correlation between multiple coagulation-related factors and lymph node metastasis in patients with gastric cancer after excluding the influence of tumor T stage. Materials and methods We retrospectively evaluated the relationship between lymph node metastasis and coagulation-related factors in 516 patients with T4a stage gastric cancer. We further analyzed influencing factors for lymph node metastasis and verified the predictive value of maximum amplitude (MA, a parameter of thromboelastography which is widely used to assess the strength of platelet-fibrinogen interaction in forming clots) in reference to lymph node metastasis. Results Platelet counts (P=0.011), fibrinogen levels (P=0.002) and MA values (P=0.006) were statistically significantly higher in patients with T4a stage gastric cancer presenting with lymph node metastasis than in those without lymph node metastasis. Moreover, tumor N stage was statistically significantly and positively correlated with platelet count (P & lt;0.001), fibrinogen level (P=0.003), MA value (P & lt;0.001), and D-dimer level (P=0.010). The MA value was an independent factor for lymph node metastasis (β=0.098, 95% CI: 1.020-1.193, P=0.014) and tumor N stage (β=0.059, 95% CI: 0.015-0.104, P=0.009), and could be used to predict the presence of lymph node metastasis in patients with gastric cancer (sensitivity 0.477, specificity 0.783, P=0.006). The independent influencing factors for MA value mainly included platelet levels, fibrinogen levels, D-dimer and hemoglobin levels; we found no statistically significant correlations with tumor diameter, tumor area, and other evaluated factors. Conclusion We conclude that MA value is an independent influencing factor for lymph node metastasis and tumor N stage in patients with T4a stage gastric cancer. The MA value has important value in predicting the presence or absence of lymph node metastasis in patients with gastric cancer.     Abstract: To understand the effect of ultraviolet (UV)-B irradiation on the antioxidant capacity and growth of lettuce (Lactuca sativa), we subjected lettuce plants to UV-B irradiation (15.55 kJ m−2 d−1) for 7 days and measured yield, photosynthetic performance, hydrogen peroxide (H2O2), reduced glutathione (GSH), and ascorbic acid (AsA) contents, and the enzyme activity and expression of genes involving AsA recycling. UV-B exposure did not significantly decrease the fresh/dry weight of the lettuce shoots. The net photosynthesis rate, internal CO2 concentration, transpiration rate, and stomatal conductance decreased during the first 4 days of irradiation and light but recovered at day 7. In UV-B-treated plants, the levels of AsA, GSH, and H2O2 increased significantly and simultaneously, with a positive correlation found between H2O2 and AsA or GSH levels. UV-B exposure upregulated the expression level of most genes encoding the enzymes involving AsA recycling but downregulated the associated enzymatic activities. The increase of AsA content in UV-B-exposed lettuce might contribute to the AsA–GSH cycle, leading to downregulation of ascorbate oxidase (AO) enzymatic activity and gene expression. UV-B irradiation had a greater impact on metabolite levels than time of UV-B treatment. These results suggest that AsA homeostasis in UV-B-treated lettuce is regulated through a feedback loop between the expression and activity of enzymes associated with AsA recycling. Short-term UV-B supplementation (24 h) could be a promising approach to enhance AsA content in lettuce.

    Abstract: Lettuce ( Lactuca sativa L.) cultivated in facilities display low vitamin C (L-ascorbic acid (AsA)) contents which require augmentation. Although UV-B irradiation increases the accumulation of AsA in crops, processes underlying the biosynthesis as well as metabolism of AsA induced by UV-B in lettuce remain unclear. Results UV-B treatment increased the AsA content in lettuce, compared with that in the untreated control. UV-B treatment significantly increased AsA accumulation in a dose-dependent manner up until a certain dose.. Based on optimization experiments, three UV-B dose treatments, no UV-B (C), medium dose 7.2 KJ·m − 2 ·d − 1 (U1), and high dose 12.96 KJ·m − 2 ·d − 1 (U2), were selected for transcriptome sequencing (RNA-Seq) in this study. The results showed that C and U1 clustered in one category while U2 clustered in another, suggesting that the effect exerted on AsA by UV-B was dose dependent. MIOX gene in the myo-inositol pathway and APX gene in the recycling pathway in U2 were significantly different from the other two treatments, which was consistent with AsA changes seen in the three treatments, indicating that AsA accumulation caused by UV-B may be associated with these two genes in lettuce. UVR8 and HY5 were not significantly different expressed under UV-B irradiation, however, the genes involved in plant growth hormones and defence hormones significantly decreased and increased in U2, respectively, suggesting that high UV-B dose may regulate photomorphogenesis and response to stress via hormone regulatory pathways, although such regulation was independent of the UVR8 pathway. Conclusions Our results demonstrated that studying the application of UV-B irradiation may enhance our understanding of the response of plant growth and AsA metabolism-related genes to UV-B stress, with particular reference to lettuce.

    Abstract: Polyester polyol (PP)‐based polyurethanes (PUs) consisting of two difunctional acids [1,4‐cyclohexanedicarboxylic acid (CHDA) and 1,6‐adipic acid (AA)] and also two diols [1,4‐cyclohexanedimethanol (CHDM) and 1,6‐hexanediol (HDO)] were synthesized by a two‐step procedure with a variable feed ratio of CHDA to AA but fixed ratio of CHDM and HDO. The prepared PPs and/or PUs were characterized by Fourier transform infrared spectroscopy, X‐ray diffraction spectroscopy, and atomic force microscopy. The effects of difunctional acids on the thermal, mechanical, and dynamic mechanical thermal properties of PPs or PU films were investigated by thermogravimetry analysis, differential thermogravimetry and dynamic mechanical thermal analysis. The results show that PP exhibits a lowest viscosity with the mole fraction of CHDA and AA at 3 : 7 whereas it delivers a lowest melting point with the mole fraction at 9 : 1. After PPs being cross‐linked by isocyanate trimers, the impact resistance, shear strength and glass transition temperature increase the mixed‐acid formulations with increasing the content of CHDA. In detail, the resultant PU almost simultaneously exhibits the best mechanical and thermal properties when the mole fraction of CHDA and AA is kept constant at 9 : 1, thus giving rise to a high glass transition temperature of 56.4°C and a onset decomposition temperature of 350°C, and also delivering a balanced toughness and hardness with an impact resistance of 100 J/g and storage modulus as high as 10 9 Pa.     Abstract: The role of ecological niches in lineage diversification has been the subject of long-standing interest of ecologists and evolutionary biologists. Specific responses to climate change can arise below the species level, resulting in differentiated adaptation or movement patterns within a given species. Thus, the urgent need to model potential responses to ongoing climate changes among genetically distinct populations within a species is increasingly recognized. In this study, we utilize the information of intraspecific variation within Quercus aquifolioides as a priori, and then focus on the potential distribution ranges and niche dynamics of its three intraspecific lineages (WSP, HDM, and Tibet) under current environmental conditions via ecological niche models (ENMs) and PCA-env ordination method, respectively. Our results indicated that the three lineages have occupied differentiated climatic niches. Although the three lineages have distinctly adaptive strategies for homogeneous environmental conditions, some lineages had sympatric projecting areas. The PCA-env demonstrated that the lineage pair WSP vs. HDM had the largest niche overlap while Tibet vs. HDM showed the smallest one. Moreover, the hypothesis of the niche was indistinguishable within the three lineages rejected, indicating the presence of niche divergence rather than niche conservatism below the species level. Our findings highlight the potential of modeling intraspecific responses to climate change and provide insights into lineage diversification within Q. aquifolioides, permitting the exploration of the information determined by niche evaluations and comparisons to understand plant diversification processes below the species level in biodiversity hotspots.
