    Abstract: In recent years, the guiding effect of highly charged ions (HCIs) through insulating nanocapillary membrane has received extensive attention. It is found that slow highly charged ions at keV energies can be guided along the capillary even when the title angle of membrane is a few degrees and larger than geometry opening angle of the capillary. Initially, Stolterfoht et al. (2002 Phys. Rev. Lett. 88 133201), according to the incident ions deposit positive charges on the capillary surface in a self-organizing manner, proposed scattering and guiding regions to explain this guiding phenomenon. Hereafter, a detailed experiment and simulation performed by Skog et al. (2008 Phys. Rev. Lett. 101 223202) provided clear evidence that the guiding process is actually attributed to the self-organized charge patches formed on the inner capillary walls. HCIs entering into a capillary may hit the surface, leaving their charge on the inner wall of the capillary. When the capillary axis is tilted with respect to the beam incidence direction, a charge patch is formed in the capillary entrance, simultaneously a repulsive electric field is created. After sufficient charge deposition this field is strong enough to deflect the subsequent ions in the direction of the capillary exit. Therefore, the ions are guided through the capillary. The deflection at the charge patch only occurs at relatively large distances from the capillary wall so that the incident charge state of the ions is kept during the passage through the capillary. Further experimental and theoretical studies on various target materials, such as polyethylene terephthalate (PET), polycarbonate (PC), SiO2, and Al2O3 indeed found that a single or a small number of charge patches near the entrance formed in the charge up process dominate the observed oscillatory variations of the ion emission angle and the final guiding process. Besides, measurements and simulations of the steering of swift ions at MeV energies have shown that the transmission mechanism of the high energy ions in a tapered tube is primarily dominated by multiple random inelastic collisions below the surface and the charge patches are not responsible for the transmission process. However, the studies of the transmission of hundreds keV ions through nanocapillaries are still lacking so far. In this work, we observe the evolution of the angular distribution, charge state distribution, FWHM and transmission rate of 100 keV H+ ions incident on a polycarbonate (PC) membrane at +1 tilt angle. It is found that the transmitted particles are located around the direction along the incident beam, not along the capillary axis, which suggests that the mechanism of hundreds keV (E/q~100 kV) protons through capillaries is significantly different from that for the guiding effect of keV protons. We present a qualitative explanation based on the data: that the 100 keV H+ are transmitted by multiple random inelastic collisions below the surface is attributed to the absence of the deposited charges on the surface of the capillary at the beginning of the experiment. After the equilibrium, several charge patches are formed on the inner wall of the capillary, which suppresses the ions to penetrate into the surface of the capillary, while the H+ is transmitted via specular scattering above the surface (or closest to the surface) assisted by the charge patches, and finally is emitted in the incident direction through twice specular scattering. This finding increases the knowledge of charged ions through nanocapillaries, which is conducible to the applications of nanosized beams produced by capillaries or tapered glass within hundreds keV energies in many scientific fields.

    Abstract: Adolescent idiopathic scoliosis (AIS) is a complex spine deformity, affecting approximately 1–3% adolescents. Earlier diagnosis could increase the likelihood of successful conservative treatment and hence reduce the need for surgical intervention. We conducted a serum metabonomic study to explore the potential biomarkers of AIS for early diagnosis. Serum metabolic profiles were firstly explored between 30 AIS patients and 31 healthy controls by ultra high-performance liquid chromatography coupled with quadrupole time-of-flight mass spectrometry. Then, the candidate metabolites were validated in an independent cohort including 31 AIS patients and 44 controls. The results showed that metabolic profiles of AIS patients generally deviated from healthy controls in both the discovery set and replication set. Seven differential metabolites were identified as candidate diagnostic biomarkers, including PC(20:4), 2-hexenoylcarnitine, beta-D-glucopyranuronicacid, DG(38:9), MG(20:3), LysoPC(18:2) and LysoPC(16:0). These candidate metabolites indicated disrupted lipid metabolism in AIS, including glycerophospholipid, glycerolipid and fatty acid metabolism. Elevated expressions of adipose triglyceride lipase and hormone sensitive lipase in adipose tissue further corroborated our findings of increased lipid metabolism in AIS. Our findings suggest that differential metabolites discovered in AIS could be used as potential diagnostic biomarkers and that lipid metabolism plays a role in the pathogenesis of AIS.

    Abstract: It’s a challenge for detecting the therapeutic targets of a polypharmacological drug from variations in the responsed networks in the differentiated populations with complex diseases, as stable coronary heart disease. Here, in an adaptive, 31-center, randomized, double-blind trial involving 920 patients with moderate symptomatic stable angina treated by 14-day Danhong injection(DHI), a kind of polypharmacological drug with high quality control, or placebo (0.9% saline), with 76-day following-up, we firstly confirmed that DHI could increase the proportion of patients with clinically significant changes on angina-frequency assessed by Seattle Angina Questionnaire (ΔSAQ-AF ≥ 20) (12.78% at Day 30, 95% confidence interval [CI] 5.86–19.71%, P  = 0.0003, 13.82% at Day 60, 95% CI 6.82–20.82%, P  = 0.0001 and 8.95% at Day 90, 95% CI 2.06–15.85%, P  = 0.01). We also found that there were no significant differences in new-onset major vascular events ( P  = 0.8502) and serious adverse events ( P  = 0.9105) between DHI and placebo. After performing the RNA sequencing in 62 selected patients, we developed a systemic modular approach to identify differentially expressed modules (DEMs) of DHI with the Z summary value less than 0 compared with the control group, calculated by weighted gene co-expression network analysis (WGCNA), and sketched out the basic framework on a modular map with 25 functional modules targeted by DHI. Furthermore, the effective therapeutic module (ETM), defined as the highest correlation value with the phenotype alteration (ΔSAQ-AF, the change in SAQ-AF at Day 30 from baseline) calculated by WGCNA, was identified in the population with the best effect (ΔSAQ-AF ≥ 40), which is related to anticoagulation and regulation of cholesterol metabolism. We assessed the modular flexibility of this ETM using the global topological D value based on Euclidean distance, which is correlated with phenotype alteration ( r 2 : 0.8204, P  = 0.019) by linear regression. Our study identified the anti-angina therapeutic module in the effective population treated by the multi-target drug. Modular methods facilitate the discovery of network pharmacological mechanisms and the advancement of precision medicine. (ClinicalTrials.gov identifier: NCT01681316).

    Abstract: In recent years, by using the etching techniques with great precision, the ion tracks in materials were converted into insulator and metal nanocapillaries. The physical and chemical properties of the inner surface on micro and nano-scales of these capillaries can be investigated by the interaction of ions with the surfaces. Stolterfoht et al. (2002 Phys. Rev. Lett. 88 133201) have found the evidence for capillary guiding in studying the transmission of 3 keV Ne7+ ions (energy/charge E/q100 kV) through the polymer nanocapillaries. The self-organized charge-up process was thought to inhibit close contact between the ions and the inner capillary walls. Skog et al. (2008 Phys. Rev. Lett. 101 223202) investigated the guiding effect of 7 keV Ne7+ ions (E/q100 kV) transmitted through SiO2 nanocapillaries, and found the evidence of sequentially formed charge patches along the capillary. For these keV highly charged ions with E/q100 kV, the charge patches were formed in a very short time, and then the repulsive electric field rapidly becomes strong enough to deflect the ions, then the ions move along the capillary axis without charge exchange. Zhou et al. (2016 Acta Phys. Sin. 65 103401) have investigated the transmission of 100 keV protons (E/q102 kV) through the nanocapillaries in polycarbonate (PC) membrane. It was found that the transmitted ions are located around the direction of the incident beam, rather than along the capillary axis. This indicated that the transmission mechanism of hundreds of keV protons through nanocapillaries is significantly different from that for keV highly charged ions. For 100 keV protons, several charge patches suppress the protons to penetrate into the surface, and the protons are transmitted via twice specular scattering near the surface and finally emitted along the incident direction. However, the study of the transmission of E/q101 kV ions through nanocapillaries is still lacking. In this work, we measure the time evolution of the relative transmission rate, charge state and angular distribution as well as the full width at half maximum of 20 keV protons (E/q101 kV) transmitted through the nanocapillaries in PC membrane at a tilt angle of +1. We observe a very long time pre-guiding period before the stable guiding process is established. During the pre-guiding period the direction of the transmitted H+ ions changes to the direction of capillary axis gradually. The transmitted H0 particles are composed of two peaks:the higher and sharper one is nearly in the beam direction, the wider and lower one is around the guiding direction. With the continuous charging-up process, the intensities of the narrow and sharp peak of transmitted H0 near the beam direction will decrease and disappear at the end. The data indicate that the scattering and guiding forces are both important for E/q101 kV ions during the period of pre-guiding process, and the guiding force is dominant till a long time pre-guiding period is ended. This finding will fill in the gap between E/q100 kV and 102 kV of previous studies of ions transmitted through nanocapillaries. It is also helpful for finding the applications of nano-and micro-sized ion beams produced by tapered glass capillary with E/q101 kV.

    Abstract: 〈 b 〉 〈 i 〉 Background: 〈 /i 〉 〈 /b 〉 Human adenoviruses (HAdV) are a common cause of viral conjunctivitis, making surveillance of them from sporadic cases of conjunctivitis important. 〈 b 〉 〈 i 〉 Methods: 〈 /i 〉 〈 /b 〉 To acquire a better understanding of the epidemic genotypes of HAdV in outpatient children with adenoviral conjunctivitis in Jiangxi Province, China (2011–2012), 179 samples from cases with a high suspicion of HAdV were analyzed by PCR. Samples confirmed to be HAdV-positive by PCR were cultured in Hep-2 cells to isolate the viruses, which were then identified through hexon gene sequencing. 〈 b 〉 〈 i 〉 Results: 〈 /i 〉 〈 /b 〉 The adenoviral conjunctivitis positivity rate was 74.86% (134/179), from which 71.64% (96/134) were infections in boys, and 92.54% (124/134) were infections in children under 5 years of age. Sixty-nine HAdV strains were isolated from the positive samples and 69 sequences were obtained. Phylogenetic analysis indicated that 33 strains (47.82%) clustered with HAdV-B7, 21 (30.43%) with HAdV-B3, 6 (8.70%) with HAdV-B55, 6 (8.70%) with HAdV-E4, 1 with HAdV-B21 (1.45%), 1 with HAdV-D37 (1.45%), and 1 with HAdV-D64 (1.45%). 〈 b 〉 〈 i 〉 Conclusions: 〈 /i 〉 〈 /b 〉 This is the first identification of HAdV-B55 relating to adenoviral conjunctivitis in China. These findings provide a firm basis for future surveillance of adenoviral conjunctivitis in China or other East Asian regions.
