    Abstract: This study was conducted to investigate whether different dietary Cyberlindnera jadinii levels affect growth performance, serum immunity, antioxidant capacity, and intestinal microbiota in growing raccoon dogs. Forty-five healthy male raccoon dogs were randomly assigned to three treatment groups, with 15 raccoon dogs per group. Each raccoon dog was housed in an individual cage. The raccoon dogs in the three groups were fed diets supplemented with Cyberlindnera jadinii at dosages of 0 (N group), 1 × 10 9 (L group) and 5 × 10 9 CFU/g (H group). A 7-day pretest period preceded a formal test period of 30 days. The results showed that Cyberlindnera jadinii in the L and H groups improved average daily gain (ADG) ( P & lt; 0.05) and decreased the ratio of feed to weight (F/G) ( P & lt; 0.05). Serum immunoglobulins A and G levels were increased in the L and H groups compared to the N group ( P & lt; 0.05). Cyberlindnera jadinii in the L and H groups increased serum superoxide dismutase activity ( P & lt; 0.05), and serum glutathione peroxidase activity was increased in the L group compared to the N group ( P & lt; 0.05). The relative abundance of Firmicutes and Actinobacteriota were increased, and the relative abundance of Bacteroidota was decreased in the L and H groups compared to the N group ( P & lt; 0.05). The relative abundance of Proteobacteria and Cyanobacteria was increased in the H group compared to the other two groups ( P & lt; 0.05). The ratio of Firmicutes to Bacteroidetes in the Cyberlindnera jadinii supplementation groups increased compared with the N group ( P & lt; 0.05). The relative abundance of Megasphaera and Bifidobacterium were increased, and the relative abundance of Prevotella was decreased in the L and H groups compared to the N group ( P & lt; 0.05). The relative abundance of Dialister was increased, while the relative abundance of Blautia was decreased in the H group compared to the other two groups ( P & lt; 0.05). The relative abundance of Agathobacter was decreased in the H group compared to the N group ( P & lt; 0.05). In conclusion, dietary supplementation with Cyberlindnera jadinii increased growth performance, serum immunity, antioxidant capacity, and improved intestinal microbiota in growing raccoon dogs. Cyberlindnera jadinii can therefore be used as a growth promoter in raccoon dogs.

    Abstract: The objectives of this study were to compare the energy values of poultry byproduct meal (PBM) as feed for adult beagle dogs using the direct, difference, and regression methods to examine dogs’ nitrogen metabolism, energy utilization, gaseous metabolism, and body health. Five groups of six 12 mo old female beagles with an average body weight of 9.67 ± 0.52 kg were tested in a 5 × 6 incomplete Latin square design, with six repetitions in each group. Five experimental diets were tested consisting of 100% PBM; three substitution diets containing either 15%, 30%, or 45% PBM (termed 15PBM, 30PBM, and 45PBM, respectively); and a basal diet (included 6.90% PBM). Each experimental period lasted for 10 d, comprising 4 d of dietary acclimation followed by 6 d of testing (including 3 d feeding period and 3 d fasting period), during which the heat production (HP) was determined and feces and urine were collected. Results showed that, in the feeding state, the nitrogen intake, urinary nitrogen, apparent nitrogen digestibility, retained nitrogen, andHP increased significantly (P  & lt; 0.05) as the PBM level increased. The net protein utilization, biological value of protein, and total apparent digestibility of amino acids did not differ between the 30PBM and 45PBM diets (P  & gt; 0.05). The O2 consumption and CO2 production of beagles during the fasting period were not influenced by the PBM level (P  & gt; 0.05). The digestible energy and metabolizable energy values of the PBM estimated by the regression method were 20.16 and 18.18 MJ/kg dry matter (DM), respectively, and did not differ from those determined by the direct method (P  & gt; 0.05). The fecal DM percentages and fecal PBM scores were significantly higher in the PBM diet than in the difference method groups (P  & lt; 0.05). The direct method group had a significantly higher fecal score (4.63) than the other groups (P  & lt; 0.05), The fecal score of the 45PBM diet (3.50) was significantly higher than the 30PBM diet (2.90; P  & lt; 0.05). In summary, the direct and difference methods of determining the effective energy value of PBM for beagles, produce significantly different results. Under the conditions of this test, the best proportion of PBM in beagle feed for optimum energy provision is 30%.

    Abstract: The fruits of Physalis (Solanaceae) have a unique structure, a lantern-like fruiting calyx known as inflated calyx syndrome (ICS) or the Chinese lantern, and are rich in steroid-related compounds. However, the genetic variations underlying the origin of these characteristic traits and diversity in Physalis remain largely unknown. Here, we present a high-quality chromosome-level reference genome assembly of Physalis floridana (~1.40 Gb in size) with a contig N50 of ~4.87 Mb. Through evolutionary genomics and experimental approaches, we found that the loss of the SEP -like MADS-box gene MBP21 subclade is likely a key mutation that, together with the previously revealed mutation affecting floral MPF2 expression, might have contributed to the origination of ICS in Physaleae, suggesting that the origination of a morphological novelty may have resulted from an evolutionary scenario in which one mutation compensated for another deleterious mutation. Moreover, the significant expansion of squalene epoxidase genes is potentially associated with the natural variation of steroid-related compounds in Physalis fruits. The results reveal the importance of gene gains (duplication) and/or subsequent losses as genetic bases of the evolution of distinct fruit traits, and the data serve as a valuable resource for the evolutionary genetics and breeding of solanaceous crops.

    Abstract: This paper aims to investigate the physical and 3D printing properties of arrowroot starch (AS), a natural biopolymer with many potential health benefits. Scanning electron microscopy images showed that AS granules had mixed spherical and elongated geometries, with average sizes of 10.5 ± 2.5 μm. The molecular weight of AS measured by gel permeation chromatography (GPC) was 3.24 × 107 g/mol, and the amylose/amylopectin ratio of AS was approximately 4:11. AS has an A-type crystal structure, with a gelatinization temperature of 71.8 ± 0.2 °C. The overlap concentration (C*) of AS in aqueous solutions was 0.42% (w/v). Temperature-dependent dynamic rheological analyses of 10% to 30% (w/v) AS fluids showed that the storage modulus (G’) reached the maximum values around the gelatinization temperatures, while the yield stress (τy) and flow stress (τf) values all increased with the increase in AS concentration. The printing accuracy of AS gels was found to be associated with the interplay between the G’ values and the restorability after extrusion, determined by the three-interval thixotropy tests (3ITT). The optimum 3D printing condition occurred at 20% (w/v) AS, the nozzle diameter of 0.60 mm, the printing speed of 100 mm/s and the extrusion speed of 100 mm/s. Our research provides a promising biopolymer to be used in the design of novel personalized functional foods.

    Abstract: Lung ultrasound can reliably diagnose pulmonary atelectasis. The object of this study is to determine the most efficient region to assess changes in atelectasis in children with congenital heart disease under general anesthesia. Design: Randomized controlled trial. Setting: Operating room at university-affiliated children’s hospital. Patients: Children between 3 months and 3 years old, scheduled for elective congenital heart disease surgery under general anesthesia. Interventions: Forty children with congenital heart disease were randomly allocated to either a 5 cm H 2 O positive end-expiratory pressure group or a standard therapy control group. Measurements and Main Results: Preoperative lung ultrasound was performed twice in each patient—after 1 and 15 minutes of mechanical ventilation. Atelectatic areas and B-lines were compared between two examinations. Different ultrasound regions were evaluated using Bland-Altman plots. The occurrence rate of atelectasis was much higher in inferoposterior lung regions (Scans 4–6) than in anterior and lateral regions (Scans 1–3). The median (interquartile range) lung ultrasound scores were lower in the positive end-expiratory pressure group than in the control group after treatment: 8 (3.3–9.8) versus 13 (8.3–17.5; p 〈 0.001). The atelectatic area was significantly decreased after treatment in the positive end-expiratory pressure group: 128 mm 2 (34.5.5–213.3 mm 2 ) versus 49.5 mm 2 (5.3–75.5 mm 2 ; p 〈 0.001). Bland-Altman plots revealed concordance between measurements in Scans 1–6 and those in Scans 4–6. In the posterior axillary line regions, changes in atelectatic area were significantly larger in the positive end-expiratory pressure group than in the control group ( p = 0.03, 0.007, and 0.018). Conclusions: Lung ultrasound in inferoposterior lung regions may be more likely to reflect changes in atelectasis and save examination time; 5 cm H 2 O positive end-expiratory pressure may be useful in lung reaeration and can reduce, but not eliminate, atelectasis in children with congenital heart disease.

    Abstract: Exosomes play critical roles in regulating various physiological and pathological processes, including immune stimulation, immune suppression, cardiovascular diseases, and cancers. Recent studies show that exosomes that transport specific microRNAs (miRNAs) are involved in tumor development. However, the molecular mechanism by which tumor invasion and migration are regulated by exosomes from non-small cell lung cancer (NSCLC) is not well understood. Here, we show that exosomes shuttling low levels of miR-34c-3p are involved in NSCLC progression. Our results showed that exosomes derived from NSCLC cells carrying low levels of miR-34c-3p could be transported into the cytoplasm of NSCLC cells and accelerate NSCLC invasion and migration by upregulating integrin α2β1. A luciferase assay revealed that integrin α2β1 was the direct target of miR-34c-3p, and overexpression of integrin α2β1 could promote the invasion and migration of NSCLC cells. The analysis of exosomes derived from clinical serum samples indicated that the expression of miR-34c-3p was significantly downregulated in exosomes from NSCLC patients compared with that of normal controls. A549-derived exosomes promoted NSCLC cells lung metastases in vivo. Exosomes shuttling low levels of miR-34c-3p were associated with the progression of NSCLC in vitro and in vivo. Our data demonstrate that exosomes shuttling low levels of miR-34c-3p can accelerate the invasion and migration of NSCLC by upregulating integrin α2β1. MiR-34c-3p can be a diagnostic and prognostic marker for NSCLC. High expression of integrin α2β1 is positively related to the migration and metastasis of NSCLC cells.

    Abstract: Diet energy is a key component of pet food, but it is usually ignored during pet food development and pet owners also have limited knowledge of its importance. This study aimed to explore the effect of diet energy on the body condition, glucolipid metabolism, fecal microbiota and metabolites of adult beagles and analyze the relation between diet and host and gut microbiota. Eighteen healthy adult neutered male beagles were selected and randomly divided into three groups. Diets were formulated with three metabolizable energy (ME) levels: the low-energy (Le) group consumed a diet of 13.88 MJ/kg ME; the medium-energy (Me) group consumed a diet of 15.04 MJ/kg ME; and the high-energy (He) group consumed a diet of 17.05 MJ/kg ME. Moreover, the protein content of all these three diets was 29%. The experiment lasted 10 weeks, with a two-week acclimation period and an eight-week test phase. Body weight, body condition score (BCS), muscle condition score (MCS) and body fat index (BFI) decreased in the Le group, and the changes in these factors in the Le group were significantly higher than in the other groups (p 〈 0.05). The serum glucose and lipid levels of the Le and He groups changed over time (p 〈 0.05), but those of the Me group were stable (p 〉 0.05). The fecal pH of the Le and He groups decreased at the end of the trial (p 〈 0.05) and we found that the profiles of short-chain fatty acids (SCFAs) and bile acids (BAs) changed greatly, especially secondary BAs (p 〈 0.05). As SCFAs and secondary BAs are metabolites of the gut microbiota, the fecal microbiota was also measured. Fecal 16S rRNA gene sequencing found that the Me group had higher α-diversity indices (p 〈 0.05). The Me group had notably higher levels of gut probiotics, such as Faecalibacterium prausnitzii, Bacteroides plebeius and Blautia producta (p 〈 0.05). The diet–host–fecal microbiota interactions were determined by network analysis, and fecal metabolites may help to determine the best physical condition of dogs, assisting pet food development. Overall, feeding dogs low- or high-energy diets was harmful for glucostasis and promoted the relative abundance of pathogenic bacteria in the gut, while a medium-energy diet maintained an ideal body condition. We concluded that dogs that are fed a low-energy diet for an extended period may become lean and lose muscle mass, but diets with low energy levels and 29% protein may not supply enough protein for dogs losing weight.

    Abstract: Hepatitis B virus (HBV) infection is still one of the most dangerous viral illnesses. HBV infects around 257 million individuals worldwide. Hepatitis B in many individuals ultimately develops hepatocellular carcinoma (HCC), which is the sixth most common cancer and the third leading cause of cancer-related deaths worldwide. The innate immunity acts as the first line of defense against HBV infection through activating antiviral genes. Along with the immune responses, pro-inflammatory cytokines are triggered to enhance the antiviral responses, but this may result in acute or chronic liver inflammation, especially when the clearance of virus is unsuccessful. To a degree, the host innate immune and inflammatory responses dominate the HBV infection and liver pathogenesis. Thus, it is crucial to figure out the signaling pathways involved in the activation of antiviral factors and inflammatory cytokines. Here, we review the interplay between HBV and the signal pathways that mediates innate immune responses and inflammation. In addition, we summarize current therapeutic strategies for HBV infection via modulating innate immunity or inflammation. Characterizing the mechanisms that underlie these HBV-host interplays might provide new approaches for the cure of chronic HBV infection.
