    Abstract: Accumulating mutations in the SARS-CoV-2 Spike (S) protein can increase the possibility of immune escape, challenging the present COVID-19 prophylaxis and clinical interventions. Here, 3 receptor binding domain (RBD) specific monoclonal antibodies (mAbs), 58G6, 510A5 and 13G9, with high neutralizing potency blocking authentic SARS-CoV-2 virus display remarkable efficacy against authentic B.1.351 virus. Surprisingly, structural analysis has revealed that 58G6 and 13G9 both recognize the steric region S 470–495 on the RBD, overlapping the E484K mutation presented in B.1.351. Also, 58G6 directly binds to another region S 450–458 in the RBD. Significantly, 58G6 and 510A5 both demonstrate prophylactic efficacy against authentic SARS-CoV-2 and B.1.351 viruses in the transgenic mice expressing human ACE2 (hACE2), protecting weight loss and reducing virus loads. Together, we have evidenced 2 potent neutralizing Abs with unique mechanism targeting authentic SARS-CoV-2 mutants, which can be promising candidates to fulfill the urgent needs for the prolonged COVID-19 pandemic.

    Abstract: The SARS-CoV-2 Omicron variant evades most currently approved neutralizing antibodies (nAbs) and caused drastic decrease of plasma neutralizing activity elicited by vaccination or prior infection, urging the need for the development of pan-variant antivirals. Breakthrough infection induces a hybrid immunological response with potentially broad, potent and durable protection against variants, therefore, convalescent plasma from breakthrough infection may provide a broadened repertoire for identifying elite nAbs. We performed single-cell RNA sequencing (scRNA-seq) and BCR sequencing (scBCR-seq) of B cells from BA.1 breakthrough-infected patients who received 2 or 3 previous doses of inactivated vaccine. Elite nAbs, mainly derived from the IGHV2–5 and IGHV3-66/53 germlines, showed potent neutralizing activity across Wuhan-Hu-1, Delta, Omicron sublineages BA.1 and BA.2 at picomolar NT 50 values. Cryo-EM analysis revealed diverse modes of spike recognition and guides the design of cocktail therapy. A single injection of paired antibodies cocktail provided potent protection in the K18-hACE2 transgenic female mouse model of SARS-CoV-2 infection.

    Abstract: Background: Lung cancer is the leading source of cancer-caused disability-adjusted life years. Medical cost burden impacts the well-being of patients through reducing income, cutting daily expenses, curtailing leisure activities, and depleting exhausting savings. The COmprehensive Score for Financial Toxicity (COST) was created and validated by De Souza and colleagues. Our study intends to measure the financial burdens of cancer therapy and investigate the link between financial toxicity and health-related quality of life (HRQoL) in an advanced lung cancer population. Methods: Patients aged ≥ 18 years with confirmed stage III to IV lung cancer were eligible. The COST questionnaire verified by de Souza et al. was used to identify financial toxicity. Multivariable linear regression analysis with log transformation univariate analysis and Pearson correlations were used to perform the analysis. Results: The majority of the patients (90.8%, n = 138/152) had an annual income of $50,000 ($7,775). The cohort's insurance situation was as follows: 64.5% of the cohort had social insurance, 20.4% had commercial insurance, and 22.0% had both. Patients who were younger age (50–59, P & lt; 0.001), employed but on sick leave, and had lower income reported increased levels of financial toxicity ( P & lt; 0.05). The risk factors for high financial toxicity: (i) younger age (50–59), (ii) & lt;1 month of savings, and (iii) being employed but on sick leave. Increased financial toxicity is moderately correlated with a decrease in QoL. Conclusion: Poorer psychological status and specific demographics are linked to increased financial toxicity (lower COST). Financial toxicity has a modest relationship with HRQoL and may have a clear link with HRQoL measurements.

    Abstract: The wear condition of steel rails directly affects the safety of railway operations. Line-structured-light visual measurement technology is used for online measurement of rail wear due to its ability to achieve high-precision dynamic measurements. However, in dynamic measurements, the random deviation of the measurement plane caused by the vibration of the railcar results in changes in the actual measured rail profile relative to its cross-sectional profile, ultimately leading to measurement deviations. To address these issues, this paper proposes a method for three-dimensional measurement of steel rail cross-sectional profiles based on binocular line-structured light. Firstly, calibrated dual cameras are used to simultaneously capture the profiles of both sides of the steel rail in the same world coordinate system, forming the complete rail profile. Then, considering that the wear at the rail waist is zero in actual operation, the coordinate of the circle center on both sides of the rail waist are connected to form feature vectors. The measured steel rail profile is aligned with the corresponding feature vectors of the standard steel rail model to achieve initial registration; next, the rail profile that has completed the preliminary matching is accurately matched with the target model based on the iterative closest point (ICP) algorithm. Finally, by comparing the projected complete rail profile onto the rail cross-sectional plane with the standard 3D rail model, the amount of wear on the railhead can be obtained. The experimental results indicate that the proposed line-structured-light measurement method for the complete rail profile, when compared to the measurements obtained from the rail wear gauge, exhibits smaller mean absolute deviation (MAD) and root mean square error (RMSE) for both the vertical and lateral dimensions. The MAD values for the vertical and lateral measurements are 0.009 mm and 0.039 mm, respectively, while the RMSE values are 0.011 mm and 0.048 mm. The MAD and RMSE values for the vertical and lateral wear measurements are lower than those obtained using the standard two-dimensional rail profile measurement method. Furthermore, it effectively eliminates the impact of vibrations during the dynamic measurement process, showcasing its practical engineering application value.

    Abstract: There is increasing incidence of pulmonary nodules due to the promotion and popularization of low-dose computed tomography (LDCT) screening for potential populations with suspected lung cancer. However, a high rate of false-positive and concern of radiation-related cancer risk of repeated CT scanning remains a major obstacle to its wide application. Here, we aimed to investigate the clinical value of a non-invasive and simple test, named the seven autoantibodies (7-AABs) assay (P53, PGP9.5, SOX2, GAGE7, GUB4-5, MAGEA1, and CAGE), in distinguishing malignant pulmonary diseases from benign ones in routine clinical practice, and construct a neural network diagnostic model with the development of machine learning methods. Method A total of 933 patients with lung diseases and 744 with lung nodules were identified. The serum levels of the 7-AABs were tested by an enzyme-linked Immunosorbent assay (ELISA). The primary goal was to assess the sensitivity and specificity of the 7-AABs panel in the detection of lung cancer. ROC curves were used to estimate the diagnosis potential of the 7-AABs in different groups. Next, we constructed a machine learning model based on the 7-AABs and imaging features to evaluate the diagnostic efficacy in lung nodules. Results The serum levels of all 7-AABs in the malignant lung diseases group were significantly higher than that in the benign group. The sensitivity and specificity of the 7-AABs panel test were 60.7% and 81.5% in the whole group, and 59.7% and 81.1% in cases with early lung nodules. Comparing to the 7-AABs panel test alone, the neural network model improved the AUC from 0.748 to 0.96 in patients with pulmonary nodules. Conclusion The 7-AABs panel may be a promising method for early detection of lung cancer, and we constructed a new diagnostic model with better efficiency to distinguish malignant lung nodules from benign nodules which could be used in clinical practice.

    Abstract: In worldwide, the use of chemical pesticides to protect wood has been greatly restricted. In recent years, a large number of researchers devoted to the search for natural, safe and non-polluting bioactive chemical compounds from plants as an alternative to synthetic organic chemical preservative. In Chinese folk, tung oil can be used as paint for wooden furniture to protect them from pests. This study aimed to evaluate the chemical compositions of raw and heated tung oil and their activity against termite. In choice bioassays, weight loss of wood treated with 5% raw or heated tung oil after 4 weeks was significantly less than that of the control group. In no-choice bioassays, there was a significant difference in termite survival and wood weight loss on raw and heated tung oil-treated wood. When tung oil-treatment concentrations increased to 5%, wood weight loss was less than 10%. There was no significant difference in termite survival and wood weight loss between raw and heated tung oil-treated wood. Survival of termites in both tung oil wood treatments was significantly lower than that in the starvation control after 4 weeks. Raw and heated tung oil significantly improved the resistance of pine wood to termites, and have the potential for the development of natural wood preservatives.

    Abstract: Increasing evidence shows that the ubiquitin–proteasome system has a crucial impact on lung adenocarcinoma. However, reliable prognostic signatures based on ubiquitination and immune traits have not yet been established. Methods Bioinformatics was performed to analyze the characteristic of ubiquitination in lung adenocarcinoma. Principal component analysis was employed to identify the difference between lung adenocarcinoma and adjacent tissue. The ubiquitin prognostic risk model was constructed by multivariate Cox regression and least absolute shrinkage and selection operator regression based on the public database The Cancer Genome Atlas, with evaluation of the time-dependent receiver operating characteristic curve. A variety of algorithms was used to analyze the immune traits of model stratification. Meanwhile, the drug response sensitivity for subgroups was predicted by the “pRRophetic” package based on the database of the Cancer Genome Project. Results The expression of ubiquitin genes was different in the tumor and in the adjacent tissue. The ubiquitin model was superior to the clinical indexes, and four validation datasets verified the prognostic effect. Additionally, the stratification of the model reflected distinct immune landscapes and mutation traits. The low-risk group was infiltrating plenty of immune cells and highly expressed major histocompatibility complex and immune genes, which illustrated that these patients could benefit from immune treatment. The high-risk group showed higher mutation and tumor mutation burden. Integrating the tumor mutation burden and the immune score revealed the patient’s discrepancy between survival and drug response. Finally, we discovered that the drug targeting ubiquitin and proteasome would be a beneficial prospective treatment for lung adenocarcinoma. Conclusion The ubiquitin trait could reflect the prognosis of lung adenocarcinoma, and it might shed light on the development of novel ubiquitin biomarkers and targeted therapy for lung adenocarcinoma.
