    Abstract: To quantify the association between serum sarcosine and diabetic retinopathy (DR) using weighted gene co‐expression network analysis (WGCNA). Methods We measured serum metabolites in 69 pairs of type 2 diabetes (T2D) patients with and without DR matched by age, gender, body mass index(BMI and HbA1c, using a propensity score matching‐based approach. To identify modules and metabolites linked to DR, pathway analysis was performed using WGCNA, the Kyoto Encyclopedia of Genes and Genomes and Small‐Molecule Pathway Database. The association of sarcosine with DR was estimated by restricted cubic spline and conditional logistic regression models. Its joint effects with covariates on DR were also extensively examined. Results With per interquartile range elevation of sarcosine, the adjusted odds ratio (AOR) of DR significantly decreased by 67% (AOR: 0.33, 95% confidence interval [CI]: 0.19‐0.58). Similar results were also found in the tertile analysis. Compared with those in the first tertile of sarcosine, the AOR significantly decreased by 54% (AOR: 0.46, 95% CI: 0.18‐1.17) and 78% (AOR: 0.22, 95% CI: 0.08‐0.59) for subjects in the second and third tertiles, respectively. Compared with subjects with lower sarcosine and lower HDL‐C levels, those with higher sarcosine and lower HDL‐C levels had the lowest odds of DR (OR: 0.13, 95% CI: 0.04, 0.43). Conclusions Serum sarcosine was inversely related to DR, especially in T2D patients with insufficient HDL‐C. This study provides insights on a possible novel target for DR precision prevention and control, as well as a better understanding of the DR mechanism.

    Abstract: Despite advances in diabetic retinopathy (DR) medications, early identification is vitally important for DR administration and remains a major challenge. This study aims to develop a novel system of multidimensional network biomarkers (MDNBs) based on a widely targeted metabolomics approach to detect DR among patients with type 2 diabetes mellitus (T2DM) efficiently. Research design and methods In this propensity score matching-based case-control study, we used ultra-performance liquid chromatography-electrospray ionization-tandem mass spectrometry system for serum metabolites assessment of 69 pairs of patients with T2DM with DR (cases) and without DR (controls). Comprehensive analysis, including principal component analysis, orthogonal partial least squares discriminant analysis, generalized linear regression models and a 1000-times permutation test on metabolomics characteristics were conducted to detect candidate MDNBs depending on the discovery set. Receiver operating characteristic analysis was applied for the validation of capability and feasibility of MDNBs based on a separate validation set. Results We detected 613 features (318 in positive and 295 in negative ESI modes) in which 63 metabolites were highly relevant to the presence of DR. A panel of MDNBs containing linoleic acid, nicotinuric acid, ornithine and phenylacetylglutamine was determined based on the discovery set. Depending on the separate validation set, the area under the curve (95% CI), sensitivity and specificity of this MDNBs system were 0.92 (0.84 to 1.0), 96% and 78%, respectively. Conclusions This study demonstrates that metabolomics-based MDNBs are associated with the presence of DR and capable of distinguishing DR from T2DM efficiently. Our data also provide new insights into the mechanisms of DR and the potential value for new treatment targets development. Additional studies are needed to confirm our findings.

    Abstract: Fatty acid ethyl esters are important flavor chemicals in strong-flavor baijiu. Microorganisms are the main contributors to ester synthesis during baijiu manufacture. However, the ester synthesis was unstable between batches. This was owing to a limited knowledge of the mechanisms for ester synthesis by microorganisms. In this work, a fatty acid ethyl ester synthesizing Aspergillus niger strain CGMCC (China General Microbiological Culture Collection) 3.4309 was identified. The conversion ratios of ethyl valerate, ethyl caproate, ethyl caprylate, and ethyl caprate were 7.87, 29.20, 94.80, and 85.20%, respectively, under the optimized conditions. A comparison of transcriptomes under the initial and optimized ester synthetic conditions indicated that 23 genes were upregulated in transcription level and encoded enzymes with potential abilities for ester synthesis. Eleven of the enzymes were expressed, and three of them, numbered An605, An1097, and An3131, showed the ability to catalyze fatty acid ethyl ester synthesis under aqueous phase, with capric acid as the preferred substrate. The possible enzymatic catalytic mechanism was proposed based on homology modeling and molecular docking. This study reported for the first time that A. niger showed the ability to efficiently catalyze the synthesis of short- and medium-chain fatty acid ethyl esters in aqueous phase, identified the key enzymes, and analyzed the basic enzymatic properties. This is helpful to promote the application of related microorganisms and enzyme resources in the baijiu industry.

    Abstract: Organic acids are widespread in the environment and play an important role in the adsorption, desorption, and migration of soil cadmium (Cd). Under the effect of humic acid (HA) and citric acid, the properties and mechanism of Cd adsorption and desorption in reclaimed soil from the Panyi Mine in the Huainan mining area were investigated (CA). The addition of HA and CA inhibited the adsorption and enhanced the desorption of Cd in reclaimed soil, with HA having a stronger effect than CA. As the concentration of added HA or CA increased, the adsorption capacity of Cd in the reclaimed soil gradually decreased, while the desorption capacity gradually increased. That is, the adsorption inhibition/desorption promotion effect was stronger for higher concentrations of organic acid. Cd adsorption and desorption could be described as a logarithmic function of organic acid concentration. The kinetic curves of Cd adsorption and desorption in reclaimed soil under the influence of organic acids show that both adsorption and desorption involved two stages: a fast reaction stage followed by a slow reaction stage.

    Abstract: Exogenous humus can change the content and migration activity of cadmium (Cd) in soil. Humic acid (HA) is an important soluble humus component in soil. In order to explore the relationship between cadmium pollution mechanism and ecological environment of humic acid in reclaimed soil, the characteristics of humic acid adsorbing cadmium in alkaline conditions were studied. This study employed reclaimed soil from the Huainan mining area, China. The adsorption and desorption characteristics as well as influence mechanisms on the heavy metal cadmium (Cd) were explored under the influence of HA. The results show that: (1) When Cd concentration was low (0.2–10 mg/L), HA had little effect on Cd adsorption and desorption in reclaimed soil. When the Cd concentration was high (15–80 mg/L), HA had a great influence on the adsorption and desorption of Cd in reclaimed soil. The addition of HA can inhibit the adsorption of Cd by reclaimed soil and effectively improve the desorption capacity of Cd by reclaimed soil. (2) The kinetic curves of Cd adsorption and desorption of reclaimed soil with added HA show that both processes (adsorption and desorption) include two stages: rapid reaction and slow reaction. The adsorption of Cd by reclaimed soil under the influence of HA was 18.18% lower than that of normal reclaimed soil, and the increase of Cd desorption was 50.29%. (3) The factors affecting the adsorption and desorption of Cd in the soil were analyzed with gray theory, and their importance can be ordered as follows: Cd concentration  〉  HA concentration  〉  pH  〉  temperature. Considering the influence of HA, a multi-factor coupling function model of adsorption and desorption of Cd in soil is established. This model provides theoretical guidance for the scientific prediction and evaluation of Cd environmental pollution risks in soil and will be useful for developing a new solution for engineering remediation of high concentration Cd contaminated soil.

    Abstract: Despite the improved survival of patients with AIDS and Kaposi's sarcoma (KS), competing events are a non‐negligible issue affecting the survival of such patients. In this study, we explored the prognostic factors of KS‐specific and non‐KS‐specific mortality in patients with AIDS‐related KS (AIDS‐KS), accounting for competing risk. Methods We identified 17 103 patients with AIDS‐KS aged 18–65 years between 1980 and 2016 from the Surveillance, Epidemiology, and End Results (SEER) 18 registry database. Prognostic factors for KS‐specific and non‐KS‐specific mortality were determined by the Fine and Grey proportional subdistribution hazard model. We built competing risk nomograms and assessed their predictive performance based on the identified prognostic factors. Results In total, 12 943 (75.68%) patients died, 1965 (15.50%) of whom died from competing events. The KS‐specific mortality rate was 14 835 per 100 000 person‐years, and the non‐KS specific mortality rate was 2719 per 100 000 person‐years. Specifically, age 〉 44 years was associated with an 11% decrease in the subdistribution hazard of KS‐specific mortality compared with age 〈 43 years but a 50% increase in the subdistribution hazard of non‐KS‐specific mortality. Being male was associated with a 26% increase in the subdistribution hazard of KS‐specific mortality compared with being female but a 32% decrease in the subdistribution hazard of non‐KS‐specific mortality. Notably, being in the antiretroviral therapy (ART) era consistently showed a decrease in the subdistribution hazard of both KS‐specific and non‐KS‐specific mortality than being in the pre‐ART era. Conclusions Competing events commonly occurred among patients with AIDS‐KS, which deserves further attention to improve the prognosis of these patients.
