    Abstract: Alcohol works on the brain to produce its desired effects, e.g., sociability and intoxication, and hence the brain is an important organ for exploring subsequent harms. These come in many different forms such as the consequences of damage during intoxication, e.g., from falls and fights, damage from withdrawal, damage from the toxicity of alcohol and its metabolites and altered brain structure and function with implications for behavioral processes such as craving and addiction. On top of that are peripheral factors that compound brain damage such as poor diet, vitamin deficiencies leading to Wernicke-Korsakoff syndrome. Prenatal alcohol exposure can also have a profound impact on brain development and lead to irremediable changes of fetal alcohol syndrome. This chapter briefly reviews aspects of these with a particular focus on recent brain imaging results. Cardiovascular effects of alcohol that lead to brain pathology are not covered as they are dealt with elsewhere in the volume.

    Abstract: Galvo scanners are popular devices for fast transversal scanning. A triangular signal is usually employed to drive galvo scanners at scanning rates close to the inverse of their response time where scanning deflection becomes a nonlinear function of applied voltage. To address this, the triangular signal is synthesized from several short ramps with different slopes. An optimization algorithm similar to a simulated annealing algorithm is used for finding the optimal signal shape to drive the galvo scanners. As a result, a significant reduction in the nonlinearity of the galvo scanning is obtained.

    Abstract: Photoacoustic imaging (PAI) is an emerging label-free and non-invasive modality for imaging biological tissues. PAI has been implemented in different configurations, one of which is photoacoustic computed tomography (PACT) with a potential wide range of applications, including brain and breast imaging. Hemispherical Array PACT (HA-PACT) is a variation of PACT that has solved the limited detection-view problem. Here, we designed an HA-PACT system consisting of 50 single element transducers. For implementation, we initially performed a simulation study, with parameters close to those in practice, to determine the relationship between the number of transducers and the quality of the reconstructed image. We then used the greatest number of transducers possible on the hemisphere and imaged copper wire phantoms coated with a light absorbing material to evaluate the performance of the system. Several practical issues such as light illumination, arrangement of the transducers, and an image reconstruction algorithm have been comprehensively studied.

    Abstract: Photoacoustic imaging (PAI) is a powerful imaging modality that relies on the PA effect. PAI works on the principle of electromagnetic energy absorption by the exogenous contrast agents and/or endogenous molecules present in the biological tissue, consequently generating ultrasound waves. PAI combines a high optical contrast with a high acoustic spatiotemporal resolution, allowing the non-invasive visualization of absorbers in deep structures. However, due to the optical diffusion and ultrasound attenuation in heterogeneous turbid biological tissue, the quality of the PA images deteriorates. Therefore, signal and image-processing techniques are imperative in PAI to provide high-quality images with detailed structural and functional information in deep tissues. Here, we review various signal and image processing techniques that have been developed/implemented in PAI. Our goal is to highlight the importance of image computing in photoacoustic imaging.

    Abstract: Psychedelic therapy (PT) is an emerging paradigm with great transdiagnostic potential for treating psychiatric disorders, including depression, addiction, post-traumatic stress disorder, and potentially others. ‘Classic’ serotonergic psychedelics, such as psilocybin and lysergic acid diethylamide (LSD), which have a key locus of action at the 5-HT2A receptor, form the main focus of this movement, but substances including ketamine, 3,4-Methylenedioxymethamphetamine (MDMA) and ibogaine also hold promise. The modern phase of development of these treatment modalities in the early 21st century has occurred concurrently with the wider use of advanced human neuroscientific research methods; principally neuroimaging. This can potentially enable assessment of drug and therapy brain effects with greater precision and quantification than any previous novel development in psychiatric pharmacology. We outline the major trends in existing data and suggest the modern development of PT has benefitted greatly from the use of neuroimaging. Important gaps in existing knowledge are identified, namely: the relationship between acute drug effects and longer-term (clinically-relevant) effects, the precise characterisation of effects at the 5-HT2A receptor and relationships with functional/clinical effects, and the possible impact of these compounds on neuroplasticity. A road-map for future research is laid out, outlining clinical studies which will directly address these three questions, principally using combined Positron Emission Tomography (PET) and Magnetic Resonance Imaging (MRI) methods, plus other adjunct techniques. Multimodal (PET/MRI) studies using modern PET techniques such as the 5-HT2A-selective ligand [11 C]Cimbi-36 (and other ligands sensitive to neuroplasticity changes) alongside MRI measures of brain function would provide a ‘molecular-functional-clinical bridge’ in understanding. Such results would help to resolve some of these questions and provide a firmer foundation for the ongoing development of PT.

    Abstract: A low-cost Photoacoustic Computed Tomography (PACT) system consisting of 16 single-element transducers has been developed. Our design proposes a fast rotating mechanism of 360o rotation around the imaging target, generating comparable images to those produced by large-number-element (e.g., 512, 1024, etc.) ring-array PACT systems. The 2D images with a temporal resolution of 1.5 s and a spatial resolution of 240 µm were achieved. The performance of the proposed system was evaluated by imaging complex phantom. The purpose of the proposed development is to provide researchers a low-cost alternative 2D photoacoustic computed tomography system with comparable resolution to the current high performance expensive ring-array PACT systems.

    Abstract: A 49-year-old woman was diagnosed with an ER + , PR-, HER2 + , BRCA- invasive ductal carcinoma which progressed metastatically to include bone, liver, and lymph node involvement. Standardised care included a 26-month treatment period with targeted chemotherapy and a ketogenic diet. The patient also began a course of cannabinoid-based therapy, consisting initially of a titrated high-dose protocol of mixed cannabidiol (CBD) and d9-tetrahydrocannabinol (THC) chemotypes, as well as psilocybin-assisted psychotherapy at macro and intermittent micro-doses. At the end of the five-month treatment period PET/CT investigations revealed no evidence of metastatic disease and chemotherapy was withdrawn. A one year follow up CT investigation concluded no evidence of residual or recurrent disease. A recurrence of disease was noted at 18 months follow up. Over these 18 months the cannabis regimen was titrated down to 60% of the initial protocol. This was subsequently increased to the initial dosing protocol following detection of recurrent disease and this titration occurred over a 10-month period where it remained stable. 16 months following the detection of recurrence of disease, favourable results were observed in the patient with evidence of receding cancer progression. Over the last 15 years there has been a considerable body of in-vitro and in-vivo evidence supporting the anti-neoplastic properties of cannabinoids and more recently psychedelics. Indeed, growing anecdotal and real-world evidence is reported of the therapeutic effect of cannabinoids and psychedelics in reducing both tumour proliferation and aiding as a palliative medicine to treat pain and psychological distress associated with cancer and chemotherapy. The data presented here indicate the potential therapeutic utility of such adjunctive pharmacological interventions in an individual with metastatic breast cancer.

    Abstract: Photoacoustic microscopic images can assist specialists in disease diagnosis by providing vascular information. However, the size of such data is usually extremely large (ie, gigabytes), and thus, a real‐time, efficient compression method can facilitate easy storage and transportation of these images. We have implemented multiple data compression methods in LabVIEW with a high compression ratio and execution times below the repetition rate of the pulsed laser. The qualitative and quantitative results of ex vivo and in vivo imaging with compression showed near‐identical images to uncompressed images, with significantly smaller size.

    Abstract: Cannabis has been legalised for medical use in an ever-increasing number of countries. A growing body of scientific evidence supports the use of medical cannabis for a range of therapeutic indications. In parallel with these developments, concerns have been expressed by many prescribers that increased use will lead to patients developing cannabis use disorder. Cannabis use disorder has been widely studied in recreational users, and these findings have often been projected onto patients using medical cannabis. However, studies exploring medical cannabis dependence are scarce and the appropriate methodology to measure this construct is uncertain. This article provides a narrative review of the current research to discern if, how and to what extent, concerns about problems of dependence in recreational cannabis users apply to prescribed medical users. We focus on the main issues related to medical cannabis and dependence, including the importance of dose, potency, cannabinoid content, pharmacokinetics and route of administration, frequency of use, as well as set and setting. Medical and recreational cannabis use differs in significant ways, highlighting the challenges of extrapolating findings from the recreational cannabis literature. There are many questions about the potential for medical cannabis use to lead to dependence. It is therefore imperative to address these questions in order to be able to minimise harms of medical cannabis use. We draw out seven recommendations for increasing the safety of medical cannabis prescribing. We hope that the present review contributes to answering some of the key questions surrounding medical cannabis dependence.

    Abstract: Photoacoustic microscopy (PAM) is a high‐resolution imaging modality that has been mainly implemented with small field of view applications. Here, we developed a fast PAM system that utilizes a unique spiral laser scanning mechanism and a wide acoustic detection unit. The developed system can image an area of 12.5 cm 2 in 6.4 s. The system has been characterized using highly detailed phantoms. Finally, the imaging capabilities of the system were further demonstrated by imaging a sheep brain ex vivo and a rat brain in vivo.
