    Abstract: Objective. Many stroke victims have severe swallowing problems. Previous neuroimaging studies have found that several brain regions scattered in the frontal, temporal, and parietal lobes, such as Brodmann’s areas (BA) 6, 21, and 40, are associated with swallowing function. This study sought to investigate changes in swallowing function and resting-state functional magnetic resonance imaging (rs-fMRI) in stroke patients with dysphagia following action observation treatment. It also sought to detect changes in brain regions associated with swallowing in stroke patients. Methods. In this study, 12 healthy controls (HCs) and 12 stroke patients were recruited. Stroke patients were given 4 weeks of action observation therapy. In order to assess the differences in mfALFF values between patients before treatment and HCs, the fractional amplitude of low-frequency fluctuations (fALFF) in three frequency bands (conventional frequency band, slow-4, and slow-5) were calculated for fMRI data. The significant brain regions were selected as regions of interest (ROIs) for subsequent analysis. The mfALFF values were extracted from ROIs of the three groups (patients before and after treatment and HCs) and compared to assess the therapeutic efficacy. Results. In the conventional band, stroke patients before treatment had higher mfALFF in the inferior temporal gyrus and lower mfALFF in the calcarine fissure and surrounding cortex and thalamus compared to HCs. In the slow-4 band, there was no significant difference in related brain regions between stroke patients before treatment and HCs. In the slow-5 band, stroke patients before treatment had higher mfALFF in inferior cerebellum, inferior temporal gyrus, middle frontal gyrus, and lower mfALFF in calcarine fissure and surrounding cortex compared to HCs. We also assessed changes in aberrant brain activity that occurred both before and after action observation therapy. The mfALFF between stroke patients after therapy was closed to HCs in comparison to the patients before treatment. Conclusion. Action observation therapy can affect the excitability of certain brain regions. The changes in brain function brought about by this treatment may help to further understand the potential mechanism of network remodeling of swallowing function.

    Abstract: As the most prevalent internal eukaryotic modification, N6-methyladenosine (m6A) is installed by methyltransferases, removed by demethylases, and recognized by readers. However, there are few studies on the role of m6A in clear cell renal cell carcinoma (ccRCC). In this study, we researched the RNA-seq transcriptome data of ccRCC in the TCGA dataset and used bioinformatics analyses to detect the relationship between m6A RNA methylation regulators and ccRCC. First, we compared the expression of 18 m6A RNA methylation regulators in ccRCC patients and normal tissues. Then, data from ccRCC patients were divided into two clusters by consensus clustering. LASSO Cox regression analysis was used to build a risk signature to predict the prognosis of patients with ccRCC. An ROC curve, univariate Cox regression analysis, and multivariate Cox regression analysis were used to verify this risk signature’s predictive ability. Then, we internally validated this signature by random sampling. Finally, we explored the role of the genes in the signature in some common pathways. Gene distribution between the two subgroups was different; cluster 2 was gender-related and had a worse prognosis. IGF2BP3, IGF2BP2, HNRNPA2B1, and METTL14 were chosen to build the risk signature. The overall survival of the high- and low-risk groups was significantly different ( p = 7.47 e − 12 ). The ROC curve also indicated that the risk signature had a decent predictive significance ( AUC = 0.72 ). These results imply that the risk signature has a potential value for ccRCC treatment.

    Abstract: Agriculture is the foundation of human civilization. However, the rapid increase of the global population poses a challenge on this cornerstone by demanding more food. Modern autonomous greenhouses, equipped with sensors and actuators, provide a promising solution to the problem by empowering precise control for high-efficient food production. However, the optimal control of autonomous greenhouses is challenging, requiring decision-making based on high-dimensional sensory data, and the scaling of production is limited by the scarcity of labor capable of handling this task. With the advances of artificial intelligence (AI), the internet of things (IoT), and cloud computing technologies, we are hopeful to provide a solution to automate and smarten greenhouse control to address the above challenges. In this paper, we propose a smart agriculture solution named iGrow, for autonomous greenhouse control (AGC): (1) for the first time, we formulate the AGC problem as a Markov decision process (MDP) optimization problem; (2) we design a neural network-based simulator incorporated with the incremental mechanism to simulate the complete planting process of an autonomous greenhouse, which provides a testbed for the optimization of control strategies; (3) we propose a closed-loop bi-level optimization algorithm, which can dynamically re-optimize the greenhouse control strategy with newly observed data during real-world production. We not only conduct simulation experiments but also deploy iGrow in real scenarios, and experimental results demonstrate the effectiveness and superiority of iGrow in autonomous greenhouse simulation and optimal control. Particularly, compelling results from the tomato pilot project in real autonomous greenhouses show that our solution significantly increases crop yield (+10.15%) and net profit (+92.70%) with statistical significance compared to planting experts. Our solution opens up a new avenue for greenhouse production. The code is available at https://github.com/holmescao/iGrow.git.

    Abstract: Trachypithecus leucocephalus , the white-headed langur, is a critically endangered primate that is endemic to the karst mountains in the southern Guangxi province of China. Studying the genomic and transcriptomic mechanisms underlying its local adaptation could help explain its persistence within a highly specialized ecological niche. Results In this study, we used PacBio sequencing and optical assembly and Hi-C analysis to create a high-quality de novo assembly of the T. leucocephalus genome. Annotation and functional enrichment revealed many genes involved in metabolism, transport, and homeostasis, and almost all of the positively selected genes were related to mineral ion binding. The transcriptomes of 12 tissues from three T. leucocephalus individuals showed that the great majority of genes involved in mineral absorption and calcium signaling were expressed, and their gene families were significantly expanded. For example, FTH1 primarily functions in iron storage and had 20 expanded copies. Conclusions These results increase our understanding of the evolution of alkali tolerance and other traits necessary for the persistence of T. leucocephalus within an ecologically unique limestone karst environment.

    Abstract: Benign monomelic amyotrophy of lower limb (BMALL) is a neurogenic syndrome representing an unclear field. Further studies might be helpful to elucidate uncertainties regarding causation, outcome, and the risk of progression to amyotrophic lateral sclerosis (ALS). Methods According to the inclusion and exclusion criteria, 37 patients with BMALL were retrospectively collected in three neuromuscular centers from January 2012 to October 2018. The detailed medical data were summarized. Multiple laboratory tests were examined. Routine electrophysiological examinations, muscle MRI of lower limbs, and muscle biopsy were conducted. Results The cohort included 24 male and 13 female cases with median age of onset 47 years. Muscle MRI revealed that the distribution of involved muscles matched with the extent of fat infiltration, so the pattern muscle atrophy can be divided into the following four types: six patients with thigh atrophy (type I), 14 patients with leg atrophy (type II); 10 patients with disproportionate atrophy in both thigh and leg (type III); and seven patients with well‐proportionate atrophy in both thigh and leg (type IV). Electrophysiological findings showed neurogenic pattern, spontaneous activity, and abnormal H reflex, which suggested a disorder of spinal anterior horn cell in the patients with types I‐III. However, no electrophysiological abnormalities were found in the patients with type IV. Muscle pathology varied from almost normal pattern to advanced neurogenic pattern in nine biopsied patients. Follow‐up showed that two patients with type II developed to ALS four years later, and all patients with type IV were in stable condition without any complaints. Conclusion Muscle MRI was useful to exactly localize the distribution of involved muscles in BMALL patients. The distribution of atrophic muscles can be roughly divided into four types based on the MRI features. The classification of distributing types might be as an indicator for the prognosis of BMALL.

    Abstract: To improve the sensitivity of surface‐enhanced Raman spectroscopy (SERS) by constructing smaller hot spots and making the target molecules more accessible, a liquid–liquid interface assembly method is used to construct a three‐layer Ag nanoparticle film structure, wherein the nanoparticles between the adjacent upper and lower layers form natural smaller gaps of 1–3 nm. The target solution is pumped into these gaps using the principle of natural transpiration (the nanopump effect). The resulting hot spot density and efficiency are significantly higher than that of the normal dry‐state method. In addition, the detection limit reaches 1 amol, which is a reduction of 2–3 orders of magnitude compared to the dry‐state method. It is also demonstrated that this dynamic detection process is suitable for detecting the substance changes occurring during sperm–oocyte binding. This method therefore provides a new means for target molecules to actively enter naturally interbedded smaller gaps and provides essentially unlimited possibilities for SERS detection.

    Abstract: Serum Golgi protein 73 (GP73) is a promising marker for significant fibrosis in adults. However, current diagnostic value of serum GP73 for liver fibrosis in children is unknown. To investigate the relationship between levels of serum GP73 and liver fibrosis in children, we measured serum GP73 in 86 healthy controls and 183 patients with liver diseases using commercially available double-antibody sandwich enzyme-linked immunosorbent assay (ELISA) kit. The value of serum GP73 in fibrosis stage assessment was compared with aspartate transaminase to platelet ratio index (APRI). We found that serum GP73 was decreasing with age in healthy controls, while it was increasing with the extent of inflammation and fibrosis in patients with liver diseases. Though area under the receiver operating curve (AUROC) of serum GP73 for diagnosing significant fibrosis was nearly equal to APRI (0.62 vs 0.64) in patients aged 3 years or older, AUROC for serum GP73 was superior to APRI (0.76 vs 0.67) in patients aged below 3 years, indicating that serum GP73 is comparable to APRI for diagnosing significant fibrosis in children.
