    Abstract: ABSTRACTLysozymes play an important role in the first line of defense in fish and potentially used as an immunity status biomarker and immune responses evaluation in fish, which often found in two types, i.e. chicken-type and goose-type (c- and g-type, respectively). To recent, the information related to the sequences and the expression analysis of the c- and g-type lysozyme genes in African catfish is still limited. In the present study, we report a partial cloning and mRNA expression analysis of c-type and g-type lysozymes in African catfish Clarias gariepinus. We have successfully cloned and partially identify the c-type, and g-type lysozyme genes of C. gariepinus, which consist of 594 and 560 of coding sequences, respectively. Catalytic and other conserved residues were identified by multiple sequences alignment and they showed high similarity with other teleost fish species. mRNA levels of the genes were analyzed by using qPCR method and their expressions in the spleen, liver, and head kidney were rapidly modulated after Aeromonas hydrophila injection, with different patterns were observed in each organ. These results confirmed that c- and g-type lysozymes played an important role in non-specific immunity against A. hydrophila infection. This study provided valuable information that can be used to understand the African catfish immune systems for better disease and stress management in C. gariepinus culture.Keywords: lysozymes, gene identification, gene expression, bacterial infection, African catfish ABSTRAKLisozim berperan dalam sistem pertahanan dini pada ikan dan sangat potensial digunakan sebagai marka status imunitas dalam evaluasi respons imun. Lisozim umum ditemukan dalam dua tipe pada ikan: tipe-ayam (tipe-c) dan tipe-angsa (tipe-g). Informasi terkait sekuens gen dan ekspresi gen kedua tipe lisozim pada ikan lele dumbo sangat terbatas. Pada penelitian ini, kami melaporkan kloning gen secara parsial, dan analisis ekspresi gen dari kedua tipe lisozim pada ikan lele dumbo C. gariepinus. Sekuens parsial gen lisozim tipe-c dan tipe-g yang berhasil diidentifikasi adalah sepanjang 594 dan 560 pasang basa. Situs katalitik dan residu khas memiliki tingkat kesamaan yang tinggi dengan spesies ikan yang lain. Analisis mRNA dilakukan dengan metode quantitative PCR (qPCR). Ekspresi kedua gen di jaringan ginjal depan, limpa, dan hati dengan cepat terinduksi pasca infeksi bakteri A. hydrophila dengan pola yang berbeda. Hasil ini menunjukkan bahwa lisozim tipe-c dan tipe-g memiliki peran yang sangat penting dalam sistem imun ikan lele dumbo terhadap infeksi A. hydrophila. Penelitian ini menghasilkan informasi penting yang dapat digunakan untuk mempelajari sistem imun ikan lele dumbo dan sebagai acuan dalam penanganan dan manajemen penyakit pada budidaya ikan lele dumbo.Kata kunci: lisozim, identifikasi gen, ekspresi gen, infeksi bakteri, ikan lele dumbo

    Abstract: Background: A large number graduates of midwife even now accompanied with low competency isues in associated with quality of clinical learning. Now implementation of clinical learning with caseload model has problems. Clinical learning model in clinical practice that student, teacher and clinical midwife experienced provide greater insight to develop an effective clinical teaching strategy in midwifery education. The main objective of this study was to investigate student midwife, teacher and clinical midwife’ insight about an ideal clinical learning model in midwifery education.Method: A qualitative study by Focus Group Discussion (FGDs). By selected randomly, this study was conducted 32 from 76 final year midwifery students at Estu Utomo Boyolali Midwifery Academy, 14 lecturer and 13 clinical midwife who involve in Estu Utomo Boyolali Midwifery Academy clinical practice program at 2013-2014. There are 4 groups of student midwife, 2 groups of lecturer and 2 groups of clinical midwife. FGDs were arranged in 3 session differently between students, lecturer and clinical midwife. FGDs were facilitated by researcher and 3 research assistant. Data were analyzed using Atlas.Ti 6.1 software to support the coding process and identifying the main categories from verbatim transcripts.Results: Six themes emerged from the focus group data, “student caseload”, “duration of clinical practice”, “clinical placement”, “clinical mentorship”, “documentation”, and “clinical assessment”. From the sixth themes, demonstrated that an ideal clinical learning model in midwifery education to design preparation clinical learning should consider about that components. An ideal clinical learning model in midwifery education should give priority to quality than quantity especially to suggest decreasing student caseload.Conclusion: This study showed that midwife student, teacher and clinical midwife suggest that clinical learning model should give priority to quality than quantity to gain midwifery care competence. Clinical learning model therefore applied with student case loading that each student have different need. They are suggest that midwifery clinical learning should be consistent with the midwifery care philosophy “women center care” that provide midwifery continuity of care.

    Abstract: One indirect causes of maternal mortality in Indonesia is "Three Delays", delays in: (1) deciding to seek appropriate medical help for an obstetric emergency; (2) reaching an appropriate obstetric facility; and (3) receiving adequate care when a facility is reached, which has relation with the lack of women's ability in empowering herself to actively participate and to make suitable decisions about midwifery care they need. The research aims to explore the partnership between woman and midwife in midwifery care. A descriptive qualitative study was conducted using a convenience sample of six low-risk women after they had given birth and six midwives from six geographically distinct midwife-led care clinic in Bantul Regency. Data were collected through semi‑structured in‑depth interviews. The interview was conducted two  to three times and recorded by audio record. Interviews were transcribed and analyzed by the thematic analysis according to Cresswell approach. Seven major themes were identified: (a) midwives ability to partner with women, (b) equality and reciprocity, (c) negotiation, (d) empowerment, (e) trust and time, (f) sharing power and responsibility, and (g) professional friendship. Most of the women are not capable to empower themselves such as less participating in decision making, less considerate in choosing a birthing position, and worsen by the limitations of midwives availability during the delivery process. Midwives need to reflect more the philosophy meaning inside the midwifery care and strengthen the midwifery professionals curriculum in order to persuade women as a partner who capable to empower themselves and to participate actively.

    Abstract: Gumanano Village, Mawasangka District is an area that has biodiversity, one of which is medicinal plants. Communities in Gumanano Village have used medicinal plants to prevent and treat various types of diseases that have been passed down from generation to generation. aims to inventory the types of medicinal plants used by the people in Gumanano Village, to know the plant organs used, the method of processing and utilization (ethno-medical). This type of research is classified as qualitative descriptive research, namely research that describes the research data qualitatively. The method used in this research is interview and exploration methods, namely field investigations to collect comprehensive data/information about the types of medicinal plants, plant organs used, how to process and use medicinal plants in Gumanano Village, Mawasangka District. The results showed 37 types of medicinal plant species consisting of 22 families. Parts of plants used as medicine in Gumanano Village are leaves, fruit, tubers, roots, stems, bark, rhizomes, tuber skin and seeds. Methods of processing plants as medicine are boiled, grated, grown, kneaded, scraped, drunk, squeezed, chewed, mashed, and cooked. The benefits of medicinal plants are used for various diseases such as fever, external injuries, red eyes, high blood pressure, measles, constipation, vomiting blood, coughs, diabetes, sarmpa and antidote, kidney stones, gout, canker sores and toothache, headache , tinea versicolor, fever, dysentery, heart disease, intestinal worms, ulcers, vomiting blood, eye medicine, cough, stomach pain, lowering high blood pressure, vaginal discharge, broken bones, kidney stones, postpartum, ulcers, and jaundice.

    Abstract: Aim: Diabetes mellitus increases the risk of peripheral artery disease (PAD) 2 times. PAD is diagnosed by ABI (Ankle Brachial Index). PAD increases mortality and morbidity of patient with Diabetes Mellitus (DM) Type 2. This study aims to determine the prevalence and risk factors of PAD in the population of type 2 diabetes mellitus (DM) in Yogyakarta, Indonesia.   Methods: This study was a community-based descriptive and analytic observational study that examines the prevalence and risk factors of PAD in type 2 diabetes mellitus. Diagnosis of PAD was detected by ABI examination using VaSera VS-1500N. All patients with type 2 diabetes mellitus in Yogyakarta who meet the inclusion criteria will be enrolled in this study. The research subjects were taken in multi-stage, cluster-random sampling in Yogyakarta. The patient will undergo an interview of demographic data which were assessed by one assessor.  Data from interviews and ABI examinations were analysed statistically.   Results: Two hundred and fifty-six (256) patients with type 2 DM consisted of 188 patients (73.4%) women and 68 patients (26.6%) men. Prevalence of PAD diagnosed by abnormal ABI was found in 41 (16%) of 256 patients. Subjects with age more than 67 years old was significantly associated with PAD occurrence (P=0,001) in type 2 DM population.   Conclusion: The prevalence of PAD was 16% of 256 type 2 DM patients. Age more than 67 years old was the strong risk factor of PAD in type 2 DM.   Key words: Ankle Brachial Index, Peripheral Arterial Disease, Diabetes mellitus, Prevalence, Risk Factor

    Abstract: A bridge structure should have a resistance to wind loads. The purpose of research is determine the coefficient of static wind load using computational calculations with FLUENT. The objects were modeled into a square 1x1 m, rectangle 2x1 m and circle diameter of 1 m with velocity of wind 10 m/s, 20 m/s and 30 m/s. The results described the value of the coefficient of drag force (CD) for square shape larger than rectangle. The coefficient of drag force (CD) for rectangular shape larger than circle. The value of coefficient of static wind do not depend on dimensional and wind speed. The coefficient value of wind static for Suramadu bridge which calculated by FLUENT Program and wind tunnel test have a similar chart patterns.

    Abstract: Tempeh is a traditional fermented Indonesian food from white soybean. Tempe has better nutritional value than non-fermented white soybean. The aim of this study was to extract tempeh oil and analyze the inhibitory potency of Alzheimer-related gene expression in LPS-induced neuronal Schwann cells. Tempeh oil was extracted with Bligh Dyer method and was analyzed with PUFA identification, anticholinesterase activity, antioxidant activity, and quantitative PCR. Tempeh oil had a total yield of 7.14%, and PUFA identification found 8.37% omega-3. The anti-acetylcholinesterase activity showed that tempeh oil 25 µg/mL had the highest activity and 500 µg/mL in anti-butyrylcholinesterase activity. The quantitative PCR showed that tempeh oil had downregulated the gene expression of PSEN1, Gsk3b, cdk5, and TNF . From this study, tempeh oil may have the potential to lower the risk of Alzheimer’s disease by regulating certain gene traits.

    Abstract: The superconducting material BPSCCO-2212 with doping Pb = 0,2 has been synthesized by the wet mixing method. Calculation is caried out at 800 °C for 10 hours. While sintering is done at 825 °C, 830 °C, 835 °C and 840 °C for 20 hours. The synthesis result were characterized by XRD X-Ray Diffraction) and SEM (Scanning Electron Microscopy). The XRD spectrum analysis results that have been processed using celref, show that in general the samples produced have formed the BPSCCO-2212 phase (indicated the presence of Bi-2212 peaks) and have been oriented (indicated the existence of peaks with h = k = 0, l even number). For the calculation result obtanced the highest volume fraction value at 830 °C sintering temperature with a value FV = 66,97 % and the highest degree of orientation at 825 °C with a value of P = 76,54 %. The results of the SEM photo recorder generally show that the crystals formed are oriented.
