    Abstract: Protein sumoylation is one of the most important post-translational modifications regulating many biological processes (Flotho A & amp; Melchior F. 2013. Ann Rev. Biochem . 82:357–85). Our previous studies have shown that sumoylation plays a fundamental role in regulating lens differentiation ( Yan et al., 2010 . PNAS , 107(49):21034-9.; Gong et al., 2014 . PNAS . 111(15):5574–9). Whether sumoylation is implicated in lens pathogenesis remains elusive. Here, we present evidence to show that the protein inhibitor of activated STAT-1 (PIAS1), a E3 ligase for sumoylation, is implicated in regulating stress-induced lens pathogenesis. During oxidative stress-induced cataractogenesis, expression of PIAS1 is significantly altered at both mRNA and protein levels. Upregulation and overexpression of exogenous PIAS1 significantly enhances stress-induced apoptosis. In contrast, silence of PIAS1 with CRISPR/Cas9 technology attenuates stress-induced apoptosis. Mechanistically, different from other cells, PIAS1 has little effect to activate JNK but upregulates Bax, a major proapoptotic regulator. Moreover, Bax upregulation is derived from the enhanced transcription activity of the upstream transcription factor, p53. As revealed previously in other cells by different laboratories, our data also demonstrate that PIAS1 promotes SUMO1 conjugation of p53 at K386 residue in lens epithelial cells and thus enhances p53 transcription activity to promote Bax upregulation. Silence of Bax expression largely abrogates PIAS1-mediated enhancement of stress-induced apoptosis. Thus, our results demonstrated that PIAS1 promotes oxidative stress-induced apoptosis through positive control of p53, which specifically upregulates expression of the downstream proapoptotic regulator Bax. As a result, PIAS1-promoted apoptosis induced by oxidative stress is implicated in lens pathogenesis.

    Abstract: The function of the transcription factor, cAMP response element‐binding protein (CREB), is activated through S133 phosphorylation by PKA and others. Regarding its inactivation, it is not well defined. cAMP response element‐binding protein plays an essential role in promoting cell proliferation, neuronal survival and the synaptic plasticity associated with long‐term memory. Our recent studies have shown that CREB is an important player in mediating stress response. Here, we have demonstrated that CREB regulates aging process through suppression of αB‐crystallin and activation of the p300‐p53‐Bak/Bax signaling axis. First, we determined that two specific protein phosphatases, PP‐1β and PP‐2Aα, can inactivate CREB through S133 dephosphorylation. Subsequently, we demonstrated that cells expressing the S133A‐CREB, a mutant mimicking constant dephosphorylation at S133, suppress CREB functions in aging control and stress response. Mechanistically, S133A‐CREB not only significantly suppresses CREB control of αB‐crystallin gene, but also represses CREB‐mediated activation of p53 acetylation and downstream Bak/Bax genes. cAMP response element‐binding protein suppression of αB‐crystallin and its activation of p53 acetylation are major molecular events observed in human cataractous lenses of different age groups. Together, our results demonstrate that PP‐1β and PP‐2Aα modulate CREB functions in aging control and stress response through de‐regulation of αB‐crystallin gene and p300‐p53‐Bax/Bak signaling axis, which regulates human cataractogenesis in the aging lens.

    Abstract: Immunotherapy has become an important treatment option for patients with advanced non-small cell lung cancer (NSCLC). At present, none of these existing biomarkers can effectively stratify true responders and there is an urgent need for identifying novel biomarkers. Exosomes derived from the serum of patients with cancer have been proven to be reliable markers for cancer diagnosis. Here, we explored the possibility of using plasma-derived exosomal microRNAs as potential biomarkers for optimal selection of patients with advanced EGFR/ALK negative NSCLC to immunotherapy. Methods From June 2017 to February 2019, 30 patients with advanced EGFR/ALK wild-type (WT) NSCLC who received PD-1/PD-L1 inhibitors were enrolled. The efficacy evaluation was conducted after every three cycles of treatment according to RECIST 1.1. Plasma samples of these patients were collected before the administration of PD-1/PD-L1 inhibitors as baseline, and after every three cycles if the patients achieved partial response (PR) or complete response. Plasma from seven healthy individuals was also collected as normal control. Exosomes were prepared by ultracentrifugation followed by total RNA extraction, and exosome-derived miRNAs were profiled using small RNA next-generation sequencing followed by differential expression analysis. Results In order to identify biomarker for better response, all five patients who achieved PR and four patients with progressive disease (PD) at efficacy evaluation were included for differential expression analysis. Based on unsupervised hierarchical clustering, exosomal miRNA expression profile was significantly altered in patients with NSCLC compared with normal controls with a total of 155 differentially expressed exosomal miRNAs. Interestingly, hsa-miR-320d, hsa-miR-320c, and hsa-miR-320b were identified significantly upregulated in the PD groups compared with the PR group at baseline before the treatment. In addition, we identified that hsa-miR-125b-5p, a T-cell suppressor, showed a trend of increased expression in the PD group at baseline and was significantly downregulated in the post-treatment plasma exosomes compared with pre-treatment samples of the PR patients. Conclusion Patients with NSCLC represent unique plasma exosomal miRNA profiles. Hsa-miR-320d, hsa-miR-320c, and hsa-miR-320b were identified as potential biomarkers for predicting the efficacy of immunotherapy in advanced NSCLCs. When T-cell suppressor hsa-miR-125b-5p was downregulated during the treatment, the patients may obtain increased T-cell function and respond well to immunotherapy.

    Abstract: High-quality large 1 T phase of Ti X 2 ( X = Te, Se, and S) single crystals have been grown by chemical vapor transport using iodine as a transport agent. The samples are characterized by compositional and structural analyses, and their properties are investigated by Raman spectroscopy. Several phonon modes have been observed, including the widely reported A 1 g and E g modes, the rarely reported E u mode (∼183 cm −1 for TiTe 2 , and ∼185 cm −1 for TiS 2 ), and even the unexpected K mode (∼85 cm −1 ) of TiTe 2 . Most phonons harden with the decrease of temperature, except that the K mode of TiTe 2 and the E u and “ A 2 u /Sh” modes of TiS 2 soften with the decrease of temperature. In addition, we also found phonon changes in TiSe 2 that may be related to charge density wave phase transition. Our results on Ti X 2 phonons will help to understand their charge density wave and superconductivity.

    Abstract: In this work, B 4 C porous ceramics reinforced alumium composite was fabricated by the sponge impregnant method and pressureless infiltration technique. The wettability of aluminum on B 4 C, the component and the wear resistance of the composite were studied .The result indicates that the infiltration temperature is determined 1200 centigrade and there are mainly Al, B 4 C, Al 3 BC , no AL 4 C 3 in the experimental composite, the wear resistance of B 4 C/AL composite is 25 to 30 times higher than that of the alumium.

    Abstract: Although water‐soluble graphene quantum dots (GQDs) have shown various promising bio‐applications due to their intriguing optical and chemical properties, the large heterogeneity in compositions, sizes, and shapes of these GQDs hampers the better understanding of their structure‐properties correlation and further uses in terms of large‐scale manufacturing practices and safety concerns. It is shown here that a water‐soluble atomically‐precise GQD (WAGQD‐C 96 ) is synthesized and exhibits a deep‐red emission and excellent sonodynamic sensitization. By decorating sterically hindered water‐soluble functional groups, WAGQD‐C 96 can be monodispersed in water without further aggregation. The deep‐red emission of WAGQD‐C 96 facilitates the tracking of its bio‐process, showing a good cell‐uptake and long‐time retention in tumor tissue. Compared to traditional molecular sonosensitizers, WAGQD‐C 96 generates superior reactive oxygen species and demonstrates excellent tumor inhibition potency as an anti‐cancer sonosensitizer in in vivo studies. A good biosafety of WAGQD‐C 96 is validated in both in vitro and in vivo assays.

    Abstract: Background. Herpes zoster (HZ) is a common infection in individuals with acquired immunodeficiency syndrome (AIDS) patients. Traditional Chinese medicine (TCM) has been used widely in clinical practice for HZ, which remains not supportive of evidence. This review aimed to evaluate the effectiveness and safety of TCM in treating HIV-associated HZ. Methods. Nine electronic databases were searched for randomized controlled trials (RCTs) testing TCM in treating HIV-associated HZ. Data were extracted on citations, interventions, and outcomes, by two authors independently. For the quality evaluation, Cochrane risk-of-bias tool 2.0 was used. Meta-analyses were performed by Revman5.3 software. Effect estimation presented as risk ratio (RR) for dichotomous data and mean difference (MD) for continuous data with their 95% confidence interval (CI). Results. Twelve RCTs (n = 644) were included; the majority of them had a high or unclear risk of bias. Meta-analysis showed that pain intensity (VAS 0–5) in the Chinese herbal medicine (CHM) group was lower than it in the drugs group (MD = −0.87, 95% CI [−1.69, −0.04], two trials, n = 93). Duration of herpes-related pain (days) of patients in the combination group was shorter than those in the drugs group (MD = −9.19, 95% CI [−16.73, −1.65] , n = 144). The incidence of postherpetic neuralgia (PHN) in the combination group was lower than in the drugs group (RR = 0.49, 95% CI [0.25, 0.99], n = 202). As for cure rate (complete absence of pain and herpes), two trials showed that CHM was better than drugs (RR = 1.58, 95% CI [1.13, 2.22] , n = 93), five trials showed combination treatment was better than drugs (RR = 1.40, 95% CI [1.08, 1.82], n = 224). T he cure rate in the acupuncture group was more than that in the drugs group (RR = 1.99, 95% CI [1.18, 3.36], n = 120). Four trials reported adverse effects and found no serious adverse events occurred. Conclusion. CHM and acupuncture demonstrate more benefits than drugs in pain relief, cure rate improvement, and incidence reduction of PHN. However, given the data limitation and TCM therapies’ diversity, the conclusions need to be verified in future trials.

    Abstract: A radial cascaded composite ultrasonic transducer is analyzed. The transducer consists of three short metal tubes and two radially polarized piezoelectric ceramic short tubes arranged alternately along the radial direction. The short metal tubes and the piezoelectric ceramic short tubes are connected in parallel electrically and in series mechanically, which can multiply the input sound power and sound intensity. Based on the theory of plane stress, the electro-mechanical equivalent circuit of radial vibration of the transducer is derived firstly. The resonance/anti-resonance frequency equation and the expression of the effective electromechanical coupling coefficient are obtained. Excellent electromechanical characteristics are determined by changing the radial geometric dimensions. Two prototypes of the transducers are designed and manufactured to support the analytical theory. It is concluded that the theoretical resonance/anti-resonance frequencies are consistent with the numerical and experimental results. When R 2 is at certain values, both the anti-resonance frequency and effective electromechanical coupling coefficient corresponding to the second mode have maximal values. The radial cascaded composite ultrasonic transducer is expected to be used in the fields of ultrasonic water treatment and underwater acoustics.
