    Abstract: The 12-oxo-phytodienoic acid reductases (OPRs) are classified into the two subgroups OPRI and OPRII. The latter proteins participate in jasmonic acid synthesis, while the function of the former ones is as yet unclear. We describe here the characterization of the OPRI gene TaOPR1, isolated from the salinity-tolerant bread wheat (Triticum aestivum) cultivar SR3. Salinity stress induced a higher level of TaOPR1 expression in the seedling roots of cv SR3 than in its parental cultivar, JN177. This induction was abolished when abscisic acid (ABA) synthesis was inhibited. The overexpression of TaOPR1 in wheat significantly enhanced the level of salinity tolerance, while its heterologous expression in Arabidopsis alleviated root growth restriction in the presence of salinity and oxidants and raised the sensitivity to ABA. In Arabidopsis, TaOPR1 promoted ABA synthesis and the ABA-dependent stress-responsive pathway, partially rescued the sensitivity of the Arabidopsis aba2 mutant defective in ABA synthesis to salinity, and improved the activities of reactive oxygen species scavengers and the transcription of their encoding genes while reducing malondialdehyde and reactive oxygen species levels. TaOPR1 did not interact with jasmonate synthesis or the jasmonate signaling pathway. Rather than serving purely as an antioxidant, we believe that TaOPR1 acts during episodes of abiotic stress response as a signaling compound associated with the regulation of the ABA-mediated signaling network.

Corn root growth and nutrient accumulation improved by five years of repeated cattle manure addition to eroded Chinese Mollisols. Can. J. Soil Sci. 92: 521–527. The use of fertilizers with additional cattle manure application on eroded soil has been reported to improve cereal yields. Limited research exists on the long-term effect of cattle manure on root growth and nutrient uptake by corn (Zea mays L.) grown on eroded soils. A field experiment was established in Hailun city, Northeast China to determine the impact of long-term cattle manure addition on corn production in eroded Mollisols. There were five levels of simulated-erosion, which removed 0, 5, 10, 20, 30 cm of topsoil. Two soil amendments were: (1) chemical fertilizer at the rate normally used by farmers in the region and (2) chemical fertilizer plus 15 000 kg ha −1 (dry weight basis) of cattle manure. Root growth (length, surface area and dry weight) was assessed at the three-leaf stage. Nitrogen (N), phosphorus (P) and potassium (K) uptake and accumulation by corn were evaluated at the three-leaf stage and at harvest. Compared with chemical fertilizer alone, 5 yr of repeated cattle manure addition significantly increased root surface area by 18–35%, and root dry weight by 45–129% in soil with simulated-erosion. The improvement of root growth by manure application was mainly correlated with the changes in larger size aggregate. N content increased by 12–59%, P by 31–129%, and K by 297–494% in corn at the three-leaf stage, and the same trend was found at harvest. Long-term cattle manure addition increased corn yield by 7% in soils with 5 cm topsoil removal, and gave similar yields in soils with 10 and 20 cm topsoil removal as non-eroded plots receiving chemical fertilizer only. Our results suggest that the increased corn yield in manure-amended soils was related to greater N, P and K accumulation due to larger root surface area and biomass. Addition of cattle manure with chemical fertilizer would be a practical and effective approach to restore soil productivity and improve corn yields in eroded Chinese Mollisols.

    Abstract: Background. Non-small cell lung cancer (NSCLC) is still a slightly less orphan disease after immunotherapy, and routine treatment has low efficiency and adverse events. Ginseng is commonly used in the treatment of NSCLC. The purpose of this study is to assess the efficacy and hemorheological indexes of ginseng and its active components in patients with non-small cell lung cancer. Methods. A comprehensive literature search was performed in PubMed, the Cochrane Library, Medline (Ovid), the Web of Science, Embase, CKNI, Wan Fang, VIP, and SinoMed up to July 2021. Only randomized controlled trials evaluating ginseng in combination with chemotherapy versus chemotherapy alone in NSCLC patients were included. Primary outcomes included patients’ condition after using ginseng or its active components. Secondary outcomes included changes in immune cells, cytokines, and secretions in serum. Data were extracted by two independent individuals, and the Cochrane Risk of Bias tool version 2.0 was applied for the included studies. Systematic review and meta-analysis were performed by RevMan 5.3 software. Results. The results included 1480 cases in 17 studies. The results of the integration of clinical outcomes showed that the treatment of ginseng (or combination of ginseng with chemotherapy) can improve the quality of life for patients with NSCLC. Analysis of immune cell subtypes revealed that ginseng and its active ingredients can upregulate the percentages of antitumor immunocyte subtypes and downregulate the accounts of immunosuppressive cells. In addition, a reduction of the inflammatory level and an increase of antitumor indicators in serum were reported. Meta-analysis showed that Karnofsky score: WMD = 16, 95% CI (9.52, 22.47); quality-of-life score: WMD = 8.55, 95%CI (6.08, 11.03); lesion diameter: WMD = −0.45, 95% CI (−0.75, −0.15); weight: WMD = 4.49, 95% CI (1.18, 7.80); CD3+: WMD = 8.46, 95% CI (5.71, 11.20); CD4+: WMD = 8.45, 95% CI (6.32, 10.57)+; CD8+: WMD = −3.76, 95% CI (−6.34, −1.18); CD4+/CD8+: WMD = 0.32, 95% CI (0.10, 0.53); MDSC: WMD = −2.88, 95% CI (−4.59, −1.17); NK: WMD = 3.67, 95% CI (2.63, 4.71); Treg: WMD = −1.42, 95% CI (−2.33, −0.51); CEA: WMD = −4.01, 95% CI (−4.12, −3.90); NSE: WMD = −4.00, 95% CI (−4.14, −3.86); IL-2: WMD = 9.45, 95% CI (8.08, 10.82); IL-4: WMD = −9.61, 95% CI (−11.16, −8.06); IL-5: WMD = −11.95, 95% CI (−13.51, −10.39); IL-6: WMD = −7.65, 95% CI (−8.70, −6.60); IL-2/IL-5: WMD = 0.51, 95% CI (0.47, 0.55); IFN-γ: WMD = 15.19, 95% CI (3.16, 27.23); IFN-γ/IL-4: WMD = 0.91, 95% CI (0.85, 0.97); VEGF: WMD = −59.29, 95% CI (−72.99, −45.58); TGF-α: WMD = −10.09, 95% CI (−12.24, −7.94); TGF-β: WMD = −135.62, 95% CI (−147.00, −124.24); TGF-β1: WMD = −4.22, 95% CI (−5.04, −3.41); arginase: WMD = −1.81, 95% CI (−3.57, −0.05); IgG: WMD = 1.62, 95% CI (0.18, 3.06); IgM: WMD = −0.45, 95% CI (−0.59, −0.31). All results are statistically significant. No adverse events were reported in the included articles. Conclusion. It is a reasonable choice to use ginseng and its active components as adjuvant therapy for NSCLC. Ginseng is helpful for NSCLC patients’ conditions, immune cells, cytokines, and secretions in the serum.

    Abstract: Background: Humans are constantly exposed to low concentrations of 4-tert-octylphenol (OP). However, studies investigating the effects of low-dose OP on the liver are scarce, and the mechanism of these effects has not been thoroughly elucidated to date. Methods: Adult male institute of cancer research (ICR) mice were exposed to low-dose OP (0, 0.01 and 1 μg/kg/day) for 7 consecutive days. Weights of mice were recorded daily during the experiment. Blood serum levels of OP, alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were determined, and haematoxylin-eosin (HE) staining of the liver was performed. We applied an integrated metabolomic and enzyme gene expression analysis to investigate liver metabolic changes, and the gene expression of related metabolic enzymes was determined by real-time PCR and ELISA. Results: OP in blood serum was increased after OP exposure, while body weights of mice were unchanged. Liver weight and its organ coefficient were decreased significantly in the OP (1 μg/kg/day) group, but ALT and AST, as well as the HE staining results, were unchanged after OP treatment. The levels of cytidine, uridine, purine and N-acetylglutamine were increased significantly, and the level of vitamin B6 was decreased significantly in mice treated with OP (1 μg/kg/day). The mRNA and protein levels of Cda and Shmt1 were both increased significantly in OP (1 μg/kg/day)-treated mice. Conclusions: Through metabolomic analysis, our study firstly found that pyrimidine and purine synthesis were promoted and that N-acetylglutamine was upregulated after low-dose OP treatment, indicating that the treatment disturbed nucleic acid and amino acid metabolism in mice liver.

    Abstract: Atherosclerosis (AS) is a disease dangerous to human health and the main pathological cause of ischemic cardiovascular diseases. Although its pathogenesis is not fully understood, numerous basic and clinical studies have shown that AS is a chronic inflammatory disease existing in all stages of atherogenesis. It may be a common link or pathway in the pathogenesis of multiple atherogenic factors. Inflammation is associated with AS complications, such as plaque rupture and ischemic cerebral infarction. In addition to inflammation, apoptosis plays an important role in AS. Apoptosis is a type of programmed cell death, and different apoptotic cells have different or even opposite roles in the process of AS. Unlike linear RNA, circular RNA (circRNA) a covalently closed circular non-coding RNA, is stable and can sponge miRNA, which can affect the stages of AS by regulating downstream pathways. Ultimately, circRNAs play very important roles in AS by regulating inflammation, apoptosis, and some other mechanisms. The study of circular RNAs can provide new ideas for the prediction, prevention, and treatment of AS.

    Abstract: Introduction. To date, identifying emergent large vessel occlusion (ELVO) patients in the prehospital stage is important but still challenging. In this present study, we aimed to design a modified prehospital acute stroke severity (mPASS) scale to identify ELVO patients and compared the scale to the PASS scale which has been published. Methods. We retrospectively evaluated a consecutive cohort of acute ischemic stroke (AIS) in our stroke unit who visited the emergercy department. These patients underwent CT angiography (CTA), MR angiography (MRA), or digital subtraction angiography (DSA) at admission. The mPASS scale was calculated based on the National Institutes of Health Stroke Scale (NIHSS) items retrospectively, including the level of consciousness commands, gaze, arm weakness, and aphasia/dysarthria. Receiver operating characteristic (ROC) analysis was used to obtain the area under the curve (AUC) of the mPASS scale, NIHSS, and PASS scale. U -statistics was used to compare the AUC of the mPASS scale to the NIHSS and PASS scale. Results. A total of 382 AIS patients were enrolled. The AUC and specificity of the mPASS scale (0.92, 84.4) were all higher than those of the PASS scale. Cortical symptoms such as gaze palsy and consciousness disorder were more specific indicators for ELVO than motor deficits. Conclusions. The mPASS scale had a better discrimination for identifying ELVO than the PASS scale in our retrospective cohort. It might predict ELVO in an effective and simple way for paramedics in the prehospital triage stage or emergency stage. Moreover, cortical symptoms might have relatively high specificities to predict ELVO on their own.
