    Abstract: Intramuscular connective tissue (IMCT) collagen is an important factor in meat quality. This study analyzed the characteristics of type I and III collagen in the IMCT of the semitendinosus (SD) and longissimus dorsi (LD) of Wuzhumuqin sheep at different growth stages (6, 9, 12, and 18 months). Utilizing sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) and Fourier transform infrared spectroscopy (FTIR), collagen types I and III were successfully isolated and shown to contain an intact triple helix structure. Immunofluorescence revealed that these collagens were located in the endomysium and perimysium. Collagen-related genes were significantly expressed in sheep aged 9 and 12 months. The amino acid content increased with age in type I collagen whereas it decreased in type III collagen. Furthermore, type III collagen contained more hydroxyproline (Hyd) than type I collagen. Differential scanning calorimetry (DSC) revealed that the thermal stability of collagen increased with age, accompanied by a decrease in solubility. Semitendinosus muscle had more collagen cross-linkages than LD muscle due to the high pyridinoline (Pyr) content in the endomysium. Finally, a correlation analysis highlighted the multiple correlations between characteristics in different types of collagen during sheep growth. In summary, the collagen characteristics in the IMCT of sheep were impacted by collagen type, muscle type, and age. Furthermore, the various correlations between these characteristics may play an important role in the development of IMCT.

    Abstract: A seeing model based on the Tatarskii equation, which uses wind shear and temperature gradient as inputs, has been proposed for Dome A in our previous study. It has been shown that the seeing was correctly reproduced statistically. In this paper, in order to investigate the ability of the Polar version of the Weather Research and Forecasting (PWRF) model to forecast the seeing, we have tried three different PWRF-simulated wind shear and temperature gradient combinations of two levels for the near-ground. The three combinations all give good performance with the correlation coefficients being higher than 0.75, when compared with the seeing measured by the Kunlun Differential Image Motion Monitor at Dome A during the summer of 2019. Besides this, the gradient Richardson number ( Ri ), which was also simulated by the PWRF model, basically behaved as expected as the Ri is generally large when the measured seeing is small. These results suggest that the PWRF model can be used to forecast the astronomical seeing above Dome A.

    Abstract: Gut microbiota make up the largest microecosystem in the human body and are closely related to chronic metabolic diseases. Herein, 520 fecal samples are collected from different regions of China, the gut microbiome in chronic kidney disease (CKD) is characterized, and CKD classifiers based on microbial markers are constructed. Compared with healthy controls (HC, n = 210), gut microbial diversity is significantly decreased in CKD ( n = 110), and the microbial community is remarkably distinguished from HC. Genera Klebsiella and Enterobacteriaceae are enriched, while Blautia and Roseburia are reduced in CKD. Fifty predicted microbial functions including tryptophan and phenylalanine metabolisms increase, while 36 functions including arginine and proline metabolisms decrease in CKD. Notably, five optimal microbial markers are identified using the random forest model. The area under the curve (AUC) reaches 0.9887 in the discovery cohort and 0.9512 in the validation cohort (49 CKD vs 63 HC). Importantly, the AUC reaches 0.8986 in the extra diagnosis cohort from Hangzhou. Moreover, Thalassospira and Akkermansia are increased with CKD progression. Thirteen operational taxonomy units are correlated with six clinical indicators of CKD. In conclusion, this study comprehensively characterizes gut microbiome in non‐dialysis CKD and demonstrates the potential of microbial markers as non‐invasive diagnostic tools for CKD in different regions of China.

    Abstract: Adipose tissue is a crucial economically significant trait that significantly influences the meat quality and growth performance of domestic animals. To reveal the changes in adipose tissue metabolism during the growth of naturally grazing sheep, we evaluated the thickness, adipocyte morphology, fatty acid profile, and metabolite profile of subcutaneous adipose tissue (SAT) from naturally grazing Sunit sheep at 6, 18, and 30 months of age (referred to as Mth-6, Mth-18, and Mth-30, respectively). The fat thickness and adipocyte number were significantly increased with the growth of the sheep (p 〈 0.05), and the increase of which from Mth-18 to Mth-30 was less than that from Mth-6 to Mth-18. Additionally, the alpha-linolenic acid metabolism was enhanced and fatty acid (FA) elongation increased with growth. The metabolomic analysis revealed 76 differentially expressed metabolites (DEMs) in the SAT in different growth stages. Interestingly, we observed elongation of FAs in lipids correlated with sheep growth. Furthermore, the expression of acylcarnitines was downregulated, and fatty acid amides, aspartic acid, acetic acid and phosphocholine were upregulated in Mth-18 and Mth-30 compared to Mth-6. Altogether, the study found that the difference in SAT in Mth-6 was great compared to Mth-18 and Mth-30. An increase in fat deposition via adipocyte proliferation with the growth of the sheep in naturally grazing. The DEMs of acylcarnitines, fatty acid amides, aspartic acid, acetic acid, and phosphocholine emerged as potential key regulators of adipose tissue metabolism. These findings illustrate the variation in and metabolic mechanism of sheep adipose tissue development under natural grazing, thus providing valuable insights into improving the edible quality of sheep meat and developing the mutton sheep industry.

    Abstract: A model for estimating astronomical seeing at Kunlun Station (Dome A, Antarctica) is proposed. This model is based on the Tatarskii equation, using the wind shear and temperature gradient as inputs, and a seeing model depending directly on the weather data is provided. The seeing and near-ground weather data to build and validate the proposed seeing model were measured at Dome A during the summer of 2019. Two calculation methods were tested from the measured weather data relating the wind shear and temperature gradient to a combination of the two levels for the boundary layer. Both methods performed well, with correlation coefficients higher than 0.77. The model can capture the main seeing trends in which the seeing becomes small when weak wind speed and strong temperature inversion occur inside the boundary layer.

    Abstract: Avian leukosis viruses (ALVs) are important contagious suppressive factors of chicken immunity and growth performance, resulted in enormous economic loss. Although virus eradication programs are applied in breeder flocks, ALVs are still widespread globally. Therefore, other valuable adjunct to reduce the negative effect of ALVs should be considered. Bursin-like peptide (BLP) showed remarkable immunomodulatory effects, whereas their influence on ALV-infected avian groups has not been reported. Here, a designed hybrid BLP was expressed in E. coli . The purified BLP was injected subcutaneously weekly in SPF chickens congenitally infected with a natural ALV strain. Then the influences of this BLP on the growth performance, immune response and virus titer of ALV-infected chickens were determined. Results This BLP injection significantly improved the body weights of ALV-infected birds ( P   〈  0.05). BLP injection significantly enhanced organ index in the BF in ALV-infected birds ( P   〈  0.05). The weekly injection of BLP significantly lengthened the maintenance time of antibodies against Newcastle disease virus (NDV) attenuated vaccine of ALV-infected birds ( P   〈  0.05) and boosted the antibody titer against avian influenza virus (AIV) H 5 inactive vaccine of mock chicken ( P   〈  0.05). BLP injection in mock chickens enhanced the levels of serum cytokines (IL-2, IL-4 and interferon-γ) ( P   〈  0.05). Surprisingly, the novel BLP significantly inhibited expression of the ALV gp85 gene in the thymus ( P   〈  0.05), kidney ( P   〈  0.05) and bursa of Fabricius (BF) ( P   〈  0.01) of ALV-infected chickens. Both viral RNA copy number and protein level decreased significantly with BLP (50 μg/mL) inoculation before ALV infection in DF1 cells ( P   〈  0.05). Conclusions This is the first report investigating the influence of BLP on the growth and immunity performance of chickens infected by ALV. It also is the first report about the antiviral effect of BLP in vivo and in vitro. This BLP expressed in E. coli showed potential as a vaccine adjuvant, growth regulator and antiretroviral drug in chickens to decrease the negative effects of ALV infection.

    Abstract: The timing of the Lhasa–Qiangtang collision following the closure of the Bangong–Nujiang Tethys Ocean has not been well constrained. An integrated study of whole-rock geochemistry and zircon U–Pb–Hf isotopes was carried out for Early Cretaceous quartz diorite-porphyrites and granites from the Yilashan and Amdo areas, northern Tibet. LA–ICP–MS zircon U–Pb dating reveal that the Yilashan and Amdo granitoids were emplaced at ~121–110 Ma. These granitic rocks display selective enrichment of light rare earth elements, large ion lithophile elements (e.g., Rb, U) and Th, but depletion of Sr and high field strength elements (e.g., Nb, Ta, Ti) compared to its neighboring elements. These new data, combined with regional geological setting, show that these igneous rocks were formed under a geodynamic setting of the Lhasa and Qiangtang (–Amdo) collision with oceanic slab breakoff and asthenospheric upwelling. The BNTO had been closed at ~121–110 Ma in the study area. Yilashan-Amdo granitoids roughly yield high (87Sr/86Sr)i ratios and obvious negative εNd(t) and zircon εHf(t) values along with old Nd TDM and zircon Hf TDM2 ages. Together with their variable U–Pb ages, these features indicate a Precambrian “hidden” crustal source beneath the northern Lhasa and Amdo terranes. The YLSS S-type granophyres were derived from partial melting of Paleoproterozoic lower crustal metagraywackes, whereas the YLSZ quartz diorite–porphyrites and the Amdo I- and A-type granites were mainly derived from partial melting of Paleo–Mesoproterozoic lower crustal mafic rocks with a certain amount of addition of mantle-derived melts. Minor amounts of the materials originated from the Amdo orthogneisses may also be involved in the formation of the YLSZ quartz diorite–porphyrites and the Amdo I-type granites. In addition, the Yilashan ophiolite was intruded by the ~112–108 Ma granophyric and quartz diorite–porphyritic intrusions before its final emplacement into the surrounding strata.
