    Kurzfassung: A shift-down response in Escherichia coli cells has been brought about by moderate concentrations of azide or cyanide. Early events of the response included a preferential inhibition of ribonucleic acid relative to deoxyribonucleic acid synthesis, a degradation of polyribosomes, and an inhibition of protein synthesis followed by a transient relief. These changes were entirely comparable to those observed with nutrient-induced shift-downs. The influences of various nutrient supplements on an azide shift-down were examined, and methionine was found to relieve effectively the inhibition of ribonucleic acid synthesis in some strains of cells.

    Kurzfassung: Properties of the ribosome-bound β-galactosidase were examined in Escherichia coli cells after prolonged induction. This fraction of enzyme was not chased from ribosomes by removal of inducer, or by treatments with hydroxylamine, puromycin, chloramphenicol, and azide. However, the metabolic turnover of this fraction could be demonstrated by means of a pulsed exposure to the phenylalanine analogue β-2-thienylalanine, and this fraction was enriched in heavy forms relative to the soluble enzyme. These observations indicated a tight coupling of the release of ribosome-bound enzyme to nascent enzyme synthesis, and it is suggested that the ribosome-bound enzyme is related to an intermediate stage in the assembly of quarternary enzyme structures.

    Kurzfassung: A procedure is described for the quantitative determination of amino compounds separated by paper chromatography. The procedure is of general applicability, and more specifically has been extensively applied to the standard P-E-PP and P-E-T chromatograms. Recovery data from these chromatograms have been obtained, and the various factors influencing the accuracy of analysis are discussed.

    Kurzfassung: When cells of a pyrimidine-requiring strain of Escherichia coli were starved of uridine, there occurred a sequential turn-off in the synthesis of different ribosomal proteins. This observation provided a basis for distinguishing between ribosomal proteins exhibiting an 'early' turn-off from those exhibiting a 'late' turn-off. Both the 'early' and 'late' classes were represented on both the 30 S and 50 S subunits, but the 30 S subunit was enriched in 'early' ribosomal proteins relative to the 50 S subunit.

    Kurzfassung: Quenching of the fluorescence of the complex between horse liver alcohol dehydrogenase (alcohol:NAD + oxidoreductase (EC 1.1.1.1)) and auramine O complex is inconsistent with a simple competitive displacement of auramine O by ethanol. Instead, the action of ethanol requires an explanation in terms of a solvent effect, or the formation of an enzyme – auramine O – ethanol ternary complex. The latter complex would have to be the low-affinity variety similar to the enzyme–NADH–ethanol ternary complex encountered in the kinetic system.

    Kurzfassung: The general mechanism for enzymic group-transfer reactions is shown to be of considerable complexity. In order to deal with this complexity, an analysis has been made of the structure of steady-state rate equations, which permits the establishment of direct relationships between specified features of mechanism and of rate behavior. This has opened up the possibility of a featurewise approach for the progressive elucidation of complex enzymic mechanisms. In particular, a theoretical basis has been provided for the comprehensive consideration and systematic differentiation of mechanisms of enzymic group-transfer reactions.

    Kurzfassung: The 3 biological domains delineated based on small subunit ribosomal RNAs (SSU rRNAs) are confronted by uncertainties regarding the relationship between Archaea and Bacteria, and the origin of Eukarya. The similarities between the paralogous valyl-tRNA and isoleucyl-tRNA synthetases in 5398 species estimated by BLASTP, which decreased from Archaea to Bacteria and further to Eukarya, were consistent with vertical gene transmission from an archaeal root of life close to Methanopyrus kandleri through a Primitive Archaea Cluster to an Ancestral Bacteria Cluster, and to Eukarya. The predominant similarities of the ribosomal proteins (rProts) of eukaryotes toward archaeal rProts relative to bacterial rProts established that an archaeal parent rather than a bacterial parent underwent genome merger with bacteria to generate eukaryotes with mitochondria. Eukaryogenesis benefited from the predominantly archaeal accelerated gene adoption (AGA) phenotype pertaining to horizontally transferred genes from other prokaryotes and expedited genome evolution via both gene-content mutations and nucleotidyl mutations. Archaeons endowed with substantial AGA activity were accordingly favored as candidate archaeal parents. Based on the top similarity bitscores displayed by their proteomes toward the eukaryotic proteomes of Giardia and Trichomonas, and high AGA activity, the Aciduliprofundum archaea were identified as leading candidates of the archaeal parent. The Asgard archaeons and a number of bacterial species were among the foremost potential contributors of eukaryotic-like proteins to Eukarya.

    Kurzfassung: The reaction mechanism for the major isoenzyme C 1 of the alcohol dehydrogenase (EC 1.1.1.1) of horse liver has been established through kinetic analysis utilizing direct correlations between rate and mechanistic features. The mechanism consists of random sequences of reaction of the enzyme with ethanol and NAD, and an obligatory sequence of reaction with NADH and acetaldehyde. At high concentrations of ethanol or NADH, excess-ethanol and excess-NADH pathways become evident involving the formation of an enzyme/NADH/ethanol complex. Even after dissociation of NADH from this complex the enzyme/ethanol complex remains imprinted with an altered conformation and differs from the regular binary complex formed under conditions of low ethanol concentrations. A range of elementary rate constants pertaining to the binding and dissociation of reactants from binary as well as ternary complexes of the mechanism has been estimated by means of a combination of algebraic analysis, curve simulation, and sensitivity analysis.
