    Abstract: Medulloblastoma is the most common pediatric brain malignancy. Patients with the Group 3 subtype of medulloblastoma (MB) often exhibit MYC amplification and/or overexpression and have the poorest prognosis. While Group 3 MB is known to be highly dependent on MYC, direct targeting of MYC remains elusive. Methods Patient gene expression data were used to identify highly expressed EYA2 in Group 3 MB samples, assess the correlation between EYA2 and MYC, and examine patient survival. Genetic and pharmacological studies were performed on EYA2 in Group 3 derived MB cell models to assess MYC regulation and viability in vitro and in vivo. Results EYA2 is more highly expressed in Group 3 MB than other MB subgroups and is essential for Group 3 MB growth in vitro and in vivo. EYA2 regulates MYC expression and protein stability in Group 3 MB, resulting in global alterations of MYC transcription. Inhibition of EYA2 tyrosine phosphatase activity, using a novel small molecule inhibitor (NCGC00249987, or 9987), significantly decreases Group 3 MB MYC expression in both flank and intracranial growth in vivo. Human MB RNA-seq data show that EYA2 and MYC are significantly positively correlated, high EYA2 expression is significantly associated with a MYC transcriptional signature, and patients with high EYA2 and MYC expression have worse prognoses than those that do not express both genes at high levels. Conclusions Our data demonstrate that EYA2 is a critical regulator of MYC in Group 3 MB and suggest a novel therapeutic avenue to target this highly lethal disease.

    Abstract: We evaluated the performance of 12 ground-motion models (GMMs) for earthquakes in the tectonically active shallow crustal region of southern California using instrumental ground-motion observations from the 2019 Ridgecrest, California, earthquake sequence (Mw 4.0–7.1). The sequence was well recorded by the Southern California Seismic Network and rapid response portable aftershock monitoring stations. Ground-motion recordings of this size and proximity are rare, valuable, and independent of GMM development, allowing us to evaluate the predictive powers of GMMs. We first compute total residuals and compare the probability density functions, means, and standard deviations of the observed and predicted ground motions. Next we use the total residuals as inputs to the probabilistic scoring method (log-likelihood [LLH]). The LLH method provides a single score that can be used to weight GMMs in the U.S. Geological Survey (USGS) National Seismic Hazard Model (NSHM) logic trees. We also explore GMM performance for a range of earthquake magnitudes, wave propagation distances, and site characteristics. We find that the Next Generation Attenuation West-2 (NGAW2) active crust GMMs perform well for the 2019 Ridgecrest, California, earthquake sequence and thus validate their use in the 2018 USGS NSHM. However, significant ground-motion residual scatter remains unmodeled by NGAW2 GMMs due to complexities such as local site amplification and source directivity. Results from this study will inform logic-tree weights for updates to the USGS National NSHM. Results from this study support the use of nonergodic GMMs that can account for regional attenuation and site variations to minimize epistemic uncertainty in USGS NSHMs.

    Abstract: Advances in seismic instrumentation have enabled data to be recorded at increasing sample rates. This has in turn created a need to establish higher frequency baselines for assessing data quality, as the widely used new high-noise model (NHNM) and new low-noise model (NLNM) of Peterson (1993) do not extend to frequencies above 10 Hz. To provide a baseline for higher frequencies (10–100 Hz), we examine power spectral density probability density functions (PSDPDFs) for high-sample rate stations available from the Incorporated Research Institutions for Seismology Data Services Modular Utility for STAtistical kNowledge Gathering (IRIS MUSTANG) quality control system. We compute high-frequency high- and low-noise baselines by matching the appropriate composite PSDPDF percentile points to NHNM and NLNM power levels at overlapping frequencies (1–10 Hz) and then extending to higher frequencies (10–100 Hz) with piecewise linear fits to the matching PSDPDF percentile. We find that the Peterson NLNM remains an accurate representation of the lower bound of global ambient Earth noise because it is lower than 99.9% of Global Seismographic Network power spectral densities. We present high-frequency high- and low-noise baselines intended primarily for use by temporary networks targeting high-frequency signals (e.g., monitoring of aftershocks or induced seismicity) based on statistics of PSDPDFs from all publicly available high-sample rate data. Most publicly available high-sample rate data are recorded by temporary deployments, and the experiment design and scientific targets of these deployments strongly influence the observed statistical distribution of high-frequency noise. We anticipate that the noise baselines presented here will be useful in automated quality control of high-sample rate seismic data. However, we note that establishing a low-noise model that accurately represents the lowest possible ambient Earth noise at frequencies up to 100 Hz will require additional continuous high-sample rate data from high-quality permanent stations in low-noise environments.

    Abstract: The selection and weighting of ground-motion models (GMMs) introduces a significant source of uncertainty in U.S. Geological Survey (USGS) National Seismic Hazard Modeling Project (NSHMP) forecasts. In this study, we evaluate 18 candidate GMMs using instrumental ground-motion observations of horizontal peak ground acceleration (PGA) and 5%-damped pseudospectral acceleration (0.02–10 s) for tectonic earthquakes and volcanic eruptions, to inform logic-tree weights for the update of the USGS seismic hazard model for Hawaii. GMMs are evaluated using two methods. The first is a total residual visualization approach that compares the probability density function (PDF), mean and standard deviations σ, of the observed and predicted ground motion. The second GMM evaluation method we use is the common total residual probabilistic scoring method (log likelihood [LLH]). The LLH method provides a single score that can be used to weight GMMs in the Hawaii seismic hazard model logic trees. The total residual PDF approach provides additional information by preserving GMM over- and underprediction across a broad spectrum of periods that is not available from a single value LLH score. We apply these GMM evaluation methods to two different data sets: (1) a database of instrumental ground motions from historic earthquakes in Hawaii from 1973 to 2007 (Mw 4–7.3) and (2) available ground motions from recent earthquakes (Mw 4–6.9) associated with 2018 Kilauea eruptions. The 2018 Kilauea sequence contains both volcanic eruptions and tectonic earthquakes allowing for statistically significant GMM comparisons of the two event classes. The Kilauea ground observations provide an independent data set allowing us to evaluate the predictive power of GMMs implemented in the new USGS nshmp-haz software system. We evaluate GMM performance as a function of earthquake depth and we demonstrate that short-period volcanic eruption ground motions are not well predicted by any candidate GMMs. Nine of the initial 18 candidate GMMs fit the observed ground motions and meet established criteria for inclusion in the update of the Hawaii seismic hazard model. A weighted mean of four top performing GMMs in this study (NGAsubslab, NGAsubinter, ASK14, A10) is 50% lower for PGA than for GMMS used in the previous USGS seismic hazard model for Hawaii.

    Abstract: The International Physical Activity Questionnaire-Short Form (IPAQ-S) has been evaluated against accelerometer-determined physical activity measures in small homogenous samples of adults in the United States. There is limited information about the validity of the IPAQ-S in diverse US samples. Methods: 142 Blacks residing in low-income housing completed the IPAQ-S and wore an accelerometer for up to 6 days. Both 1- and 10-minute accelerometer bouts were used to define time spent in light, moderate, and vigorous physical activity. Results: We found fair agreement between the IPAQ-S and accelerometer-determined physical activity ( r = .26 for 10-minute bout, r = .36 for 1-minute bout). Correlations were higher among men than women. When we classified participants as meeting physical activity recommendations, agreement was low (kappa = .04, 10-minute; kappa = .21, 1-minute); only 25% of individuals were classified the same by both instruments (10-minute bout). Conclusions: In one of the few studies to assess the validity of a self-reported physical activity measure among Blacks, we found moderate correlations with accelerometer data, though correlations were weaker for women. Correlations were smaller when IPAQ-S data were compared using a 10- versus a 1-minute bout definition. There was limited evidence for agreement between the instruments when classifying participants as meeting physical activity recommendations.

    Abstract: Early diagnosis of ankylosing spondylitis (AS) remains challenging because of the high prevalence of chronic back pain in patients initially treated by nonrheumatology health care providers (HCPs). We assessed the patient pathway to rheumatology referral, including HCP recognition of inflammatory back pain (IBP) and other features suggestive of AS, diagnostic workup, treatment, and referral to a specialist with the goal of identifying barriers to patient referral to a rheumatologist. Methods US HCPs from 10 specialties were invited to participate in a cross‐sectional web‐based survey on clinical characteristics and diagnostic measures leading to IBP suspicion and the subsequent referral process. Eligible HCPs were actively practicing and had referred a patient with suspected IBP or ocular findings (ophthalmology only) within 12 months. Data were analyzed descriptively. Results Of 1690 HCPs, 61% identified morning stiffness lasting more than 30 minutes, 29% sleep disturbance due to back pain, and 28% pain that improves with activity as features suggestive of IBP. Nearly two‐thirds of primary care HCPs reported that they were the first HCPs consulted by patients with suspected IBP. Among HCPs ordering diagnostic blood work, approximately 90% selected antinuclear antibody and rheumatoid factor, whereas 76% selected human leukocyte antigen B27. Almost 40% would treat patients with suspected IBP themselves. HCPs cited lack of adequate specialists nearby (35.1%), insurance restrictions (47.1%), and long wait time (77.0%) as barriers to early referral. Conclusion Most HCPs had difficulty identifying features suggestive of IBP and indicated insurance restrictions and long wait times as barriers to early referral of patients with potential AS.
