    Abstract: 〈 b 〉 〈 i 〉 Background/Aims: 〈 /i 〉 〈 /b 〉 The kinases SPAK (SPS1-related proline/alanine-rich kinase) and OSR1 (oxidative stress-responsive kinase 1) participate in the regulation of the NaCl cotransporter NCC and the Na 〈 sup 〉 + 〈 /sup 〉 ,K 〈 sup 〉 + 〈 /sup 〉 ,2Cl 〈 sup 〉 - 〈 /sup 〉 cotransporter NKCC2. The kinases are regulated by WNK (with-no-K[Lys]) kinases. Mutations of genes encoding WNK kinases underly Gordon's syndrome, a monogenic disease leading to hypertension and hyperkalemia. WNK kinases further regulate the renal outer medullary K 〈 sup 〉 + 〈 /sup 〉 channel ROMK1. The present study explored, whether SPAK and/or OSR1 have similarly the potential to modify the activity of ROMK1. 〈 b 〉 〈 i 〉 Methods: 〈 /i 〉 〈 /b 〉 ROMK1 was expressed in 〈 i 〉 Xenopus 〈 /i 〉 oocytes with or without additional expression of wild-type SPAK, constitutively active 〈 sup 〉 T233E 〈 /sup 〉 SPAK, catalytically inactive 〈 sup 〉 D212A 〈 /sup 〉 SPAK, wild-type OSR1, constitutively active 〈 sup 〉 T185E 〈 /sup 〉 OSR1 and catalytically inactive 〈 sup 〉 D164A 〈 /sup 〉 OSR1. Channel activity was determined utilizing dual electrode voltage clamp and ROMK1 protein abundance in the cell membrane utilizing chemiluminescence of ROMK1 containing an extracellular hemagglutinin epitope (ROMK1-HA). 〈 b 〉 〈 i 〉 Results: 〈 /i 〉 〈 /b 〉 ROMK1 activity and ROMK1-HA protein abundance were significantly down-regulated by wild-type SPAK and 〈 sup 〉 T233E 〈 /sup 〉 SPAK, but not by 〈 sup 〉 D212A 〈 /sup 〉 SPAK. Similarly, ROMK1 activity and ROMK1-HA protein abundance were significantly down-regulated by wild-type OSR1 and 〈 sup 〉 T185E 〈 /sup 〉 OSR1, but not by 〈 sup 〉 D164A 〈 /sup 〉 OSR1. 〈 b 〉 〈 i 〉 Conclusion: 〈 /i 〉 〈 /b 〉 ROMK1 protein abundance and activity are down-regulated by SPAK and OSR1.

    Abstract: Background/Aims: Janus kinase 3 (JAK3), a tyrosine kinase contributing to the regulation of cell proliferation and apoptosis of lymphocytes and tumour cells, has been shown to modify the expression and function of several ion channels and transport proteins. Channels involved in the regulation of cell proliferation include the large conductance voltage- and Ca2+-activated K+ channel BK. The present study explored whether JAK3 modifies BK channel protein abundance and current. Methods: cRNA encoding Ca2+-insensitive BK channel (BKM513I+Δ899-903) was injected into Xenopus oocytes with or without additional injection of cRNA encoding wild-type JAK3, constitutively active A568VJAK3, or inactive K851AJAK3. Voltage gated K+ channel activity was measured utilizing dual electrode voltage clamp. Moreover, BK channel protein abundance was determined utilizing flow cytometry in CD19+ B lymphocyte cell membranes from mice lacking functional JAK3 (jak3-/-) and corresponding wild-type mice (jak3+/+). Results: BK activity in BKM513I+Δ899-903 expressing oocytes was slightly but significantly decreased by coexpression of wild-type JAK3 and of A568VJAK3, but not by coexpression of K851AJAK3. The BK channel protein abundance in the cell membrane was significantly higher in jak3-/- than in jak3+/+ B lymphocytes. The decline of conductance in BK and JAK3 coexpressing oocytes following inhibition of channel protein insertion by brefeldin A (5 µM) was similar in oocytes expressing BK with JAK3 and oocytes expressing BK alone, indicating that JAK3 might slow channel protein insertion into rather than accelerating channel protein retrieval from the cell membrane. Conclusion: JAK3 is a weak negative regulator of membrane BK protein abundance and activity.

    Abstract: Background/Aims: The oxidative stress-responsive kinase 1 (OSR1) and the serine/threonine kinases SPAK (SPS1-related proline/alanine-rich kinase) are under the control of WNK (with-no-K [Lys]) kinases. OSR1 and SPAK participate in diverse functions including cell volume regulation and neuronal excitability. Cell volume and neuronal excitation are further modified by the large conductance Ca2+-activated K+ channels (maxi K+ channel or BK channels). An influence of OSR1 and/or SPAK on BK channel activity has, however, never been shown. The present study thus explored whether OSR1 and/or SPAK modify the activity of BK channels. Methods: cRNA encoding the Ca2+ insensitive BK channel mutant BKM513I+Δ899-903 was injected into Xenopus laevis oocytes without or with additional injection of cRNA encoding wild-type OSR1 or wild-type SPAK, constitutively active T185EOSR1, catalytically inactive D164AOSR1, constitutively active T233ESPAK or catalytically inactive D212ASPAK. K+ channel activity was measured utilizing dual electrode voltage clamp. Results: BK channel activity in BKM513I+Δ899-903 expressing oocytes was significantly decreased by co-expression of OSR1 or SPAK. The effect of wild-type OSR1/SPAK was mimicked by T185EOSR1 and T233ESPAK, but not by D164AOSR1 or D212ASPAK. Conclusions: OSR1 and SPAK suppress BK channels, an effect possibly contributing to cell volume regulation and neuroexcitability.

The present study explored whether SPAK and/or OSR1 influence the excitatory amino acid transporter EAAT3, which accomplishes glutamate and aspartate transport in kidney, intestine and brain. 〈 b 〉 〈 i 〉 Methods: 〈 /i 〉 〈 /b 〉 cRNA encoding EAAT3 was injected into 〈 i 〉 Xenopus laevis 〈 /i 〉 oocytes with or without additional injection of cRNA encoding wild-type SPAK, constitutively active 〈 sup 〉 T233E 〈 /sup 〉 SPAK, WNK insensitive 〈 sup 〉 T233A 〈 /sup 〉 SPAK, catalytically inactive 〈 sup 〉 D212A 〈 /sup 〉 SPAK, wild-type OSR1, constitutively active 〈 sup 〉 T185E 〈 /sup 〉 OSR1, WNK insensitive 〈 sup 〉 T185A 〈 /sup 〉 OSR1 and catalytically inactive 〈 sup 〉 D164A 〈 /sup 〉 OSR1. Glutamate-induced current was taken as measure of electrogenic glutamate transport and was quantified utilizing dual electrode voltage clamp. Furthermore, Ussing chamber was employed to determine glutamate transport in the intestine from gene-targeted mice carrying WNK insensitive SPAK 〈 i 〉 (spak 〈 /i 〉 〈 sup 〉 〈 i 〉 tg/tg 〈 /i 〉 〈 /sup 〉 〈 i 〉 ) 〈 /i 〉 and from corresponding wild-type mice 〈 i 〉 (spak 〈 /i 〉 〈 sup 〉 +/+ 〈 /sup 〉 〈 i 〉 ) 〈 /i 〉 . 〈 b 〉 〈 i 〉 Results: 〈 /i 〉 〈 /b 〉 EAAT3 activity was significantly decreased by wild-type SPAK and 〈 sup 〉 T233E 〈 /sup 〉 SPAK, but not by 〈 sup 〉 T233A 〈 /sup 〉 SPAK and 〈 sup 〉 D212A 〈 /sup 〉 SPAK. SPAK decreased maximal transport rate without affecting significantly affinity of the carrier. Similarly, EAAT3 activity was significantly downregulated by wild-type OSR1 and 〈 sup 〉 T185E 〈 /sup 〉 OSR1, but not by 〈 sup 〉 T185A 〈 /sup 〉 OSR1 and 〈 sup 〉 D164A 〈 /sup 〉 OSR1. Again OSR1 decreased maximal transport rate without affecting significantly affinity of the carrier. Intestinal electrogenic glutamate transport was significantly lower in 〈 i 〉 spak 〈 /i 〉 〈 sup 〉 +/+ 〈 /sup 〉 than in 〈 i 〉 spak 〈 /i 〉 〈 sup 〉 〈 i 〉 tg/tg 〈 /i 〉 〈 /sup 〉 mice. 〈 b 〉 〈 i 〉 Conclusion: 〈 /i 〉 〈 /b 〉 Both, SPAK and OSR1 are negative regulators of EAAT3 activity.

    Abstract: Introduction: Pregnancy is a rollercoaster ride for a woman. With joy and excitement, there are also times of stress, anxiety, and depression. This research aims to investigate the effect of anxiety, depression and stress on blood glucose levels in pregnant women. Methodology: In this cross-sectional study, 123 pregnant women were included using random sampling. DASS-21 questionnaire, a validated tool, was used to determine stress, anxiety and depression. Random Blood glucose levels of the study participants were measured. The Chi-square test was used, and p values were determined. Results: Among 123 females, 56.91% (n=70) were stressed, 78.86% (n=97) were depressed, and 84.55% (n=104) were anxious. Blood glucose level was significantly higher in stressed and depressed pregnant females (X2  = 12.77, P-value = 0.0004 and X2  =5.75  0.016, P-value: 0.016 respectively). Conclusion: Stressed and depressed pregnant women were found to have higher blood glucose levels. Anxiety didn’t significantly affect blood glucose levels in pregnant females.

    Abstract: Objective: To evaluate the factors leading to the early onset of menopause ( 〈 40 years) in the women in the interior of Sindh. Materials and Methods: A cross-sectional study was conducted on 218 individuals among them 109 were postmenopausal women (with premature menopause)  and 109 were normal menstruating females during the period of six months from November 2014 to April  2015, data was obtained from Interior Sindh, Larkana, Hyderabad, and Benazirabad.  Factors considered were age, number of children, history of the obstetric process (ovarian and uterine), diseases, and premature menopause history in first-cousin marriages. The questionnaire data and blood samples were collected for hormonal assays such as LH,  prolactin, and FSH. The hormone levels were analyzed by ELISA method, SPSS version 17 was used for data analysis. Results:  Out of 109 subjects the percentage of early menopause due to: anorexia nervosa was 17%, brain tumor 7%, ovarian cancer 6%, hormonal disorders 23%, hysterectomy 15%, oophorectomy 5%, Pituitary gland dysfunction 4%, Sheehan Syndrome 24%, Polycystic Ovarian Syndrome (PCOS) 8%. Conclusion: Early menopause was found related to pathological and psychological factors including brain tumor, ovarian cancer, family history, Anorexia nervosa, and certain surgical interventions.

    Abstract: Objective: To Compare and Analyze the impact of different methods of contraception on liver Profile of females of Sindh province. Study setting: This is a comparative cross-sectional study that was done at Liaquat university of medical and health sciences Jamshoro, at center 'A' of reproductive health services. Methodology: 300 participants from Hyderabad Sindh were selected. Females aged 20 to 40 years and those on an individual method of contraception were selected.300 females were selected for this study. Among them, 138 ( 46.0%) were residents of rural Sindh whereas162(54.0%) belonged tour ban Sindh. Consent from participants both verbal and written was taken. A structured questionnaire was filled out for demographic characteristics following detailed history. Blood Sampling was done for liver function test. Results: The results showed that present users of contraceptives generally had significantly greater liver parameters biliru bin, gamma GT, SGPT, and Alkaline phosphates compared to non-users ( p-value= 〈 0.05). Bilirubin level was considerably greater among women who were using the contraceptive methods DMPA injection (0.72±0.24), Jadelle (0.54±0.26), IUCD (0.44±0.18) than the corresponding values for nonusers (0.43±0.194) [control 0.43±0.194 vs DMPA injection(0.72±0.24)vsJadelle(0.54±0.26)vsIUCD(0.44±0.18),P-value= 〈 0.05)]. Gama GT level was considerably greater among women who were using the contraceptive methods Jadelle (20.36±11.97), Oral (14.00±3.2) than the corresponding values for non users (15.42±7.36)[control 15.42±7.36 Jadelle (20.36±11.97) vs Oral (14.00±3.2) Oral (14.00±3.2) P-value = 〈 0.05)]. SGPT level was considerably greater among women who were using the contraceptive methods DMPA injection (32.68±18.37), Jadelle (31.34±14.13), IUCD (29.66±14.62), Tubal ligation (18.00±0.00) than the corresponding values for nonusers (25.39±14.59) [control25.39±14.59 vs injection (32.68±18.37) vs Jadelle (31.34±14.13) vs IUCD (29.66±14.62), Tubal ligation (18.00±0.00) P-value = 〈 0.05)]. Alkaline Phosphatase level was considerably greater among women who were using the contraceptive methods DMPA injection (114.65±48.19),Jadelle (99.28±38.076),IUCD(101.74±42.23)Oral(78.89±21.70)TubalLigation(62.50±2.12)Condom (94.62±25.67) than the corresponding values for nonusers (131.51±59.04) [control131.51±59.04 vs DMPA injection (114.65±48.19) vs Jadelle (99.28±38.076) vs IUCD(101.74±42.23),Oral(78.89±21.70)Tuballigation (62.50±2.12)Condom(94.62±25.67)P-value= 〈 0.05)]. Conclusion: The impact on the liver of different contraceptive methods was compared and analyzed in the population of Sindh.
