    Abstract: Petal senescence is the final stage of flower development. Transcriptional regulation plays key roles in this process. However, whether and how post‐transcriptional regulation involved is still largely unknown. Here, we identified an ethylene‐induced NAC family transcription factor DcNAP in carnation ( Dianthus caryophyllus L.). One allele, DcNAP‐dTdic1 , has an insertion of a dTdic1 transposon in its second exon. The dTdic1 transposon disrupts the structure of DcNAP and causes alternative splicing, which transcribes multiple domain‐deleted variants ( DcNAP2 and others). Conversely, the wild type allele DcNAP transcribes DcNAP1 encoding an intact NAC domain. Silencing DcNAP1 delays and overexpressing DcNAP1 accelerates petal senescence in carnation, while silencing and overexpressing DcNAP2 have the opposite effects, respectively. Further, DcNAP2 could interact with DcNAP1 and interfere the binding and activation activity of DcNAP1 to the promoters of its downstream target ethylene biosynthesis genes DcACS1 and DcACO1 . Lastly, ethylene signalling core transcriptional factor DcEIL3‐1 can activate the expression of DcNAP1 and DcNAP2 in the same way by binding their promoters. In summary, we discovered a novel mechanism by which DcNAP regulates carnation petal senescence at the post‐transcriptional level. It may also provide a useful strategy to manipulate the NAC domains of NAC transcription factors for crop genetic improvement.

    Abstract: To define the impact of hyperlipidemia as a coexisting factor on the prognosis of Idiopathic Sudden Sensorineural Hearing Loss (ISSHL), we prospectively analysed the Lipid‐lowering therapy Group (LLTG) data compared with Control Group (CG) data to determine the effects of Lipid intervention on the prognosis of sudden hearing loss. Design A prospective, non‐randomized study. Setting Shandong Provincial Hospital Affiliated to Shandong University, Shandong, China. Participants A total of 653 in‐patient SSHL patients were enrolled between January 2014 to August 2018. Main outcome measures From January 2014 to August 2018, 653 patients with ISSHL who also had hyperlipidemia as coexisting factor were identified. Patients enrolled in LLTG (n = 200) or CG (n = 453) were compared by a propensity score matching analysis (PSM, caliper = 0.01, n = 2) to balance pre‐existing clinical characteristics. After matching, the effective rate of different hyperlipidemia types and different types of audiogram in both groups were performed by Cochran‐Mantel‐Haenszel test (CMH). Results After PSM, 440 patients were studied (146 in LLTG, 294 in CG), and the influence of interference factors was balanced, meanwhile, the final hearing level was better in LLTG than CG ( P  = .043), and hearing gain was higher in LLTG than CG ( P  = .006). Cure rate (32.9%), significant improvement rate (22.6%) and the total effective rate (76.0%) in LLTG were better than that in CG group (26.5%, 15.6% and 63.6%) after the Pearson chi‐square test ( P   〈  .05). Analysis with the Cochran‐Mantel‐Haenszel test showed that the total effective rate was better in LLTG than CG respectively ( P  = .009) in each different hyperlipidemia types, and there were statistically significant differences in TG higher group (TG Group; P  = .018). Moreover, the total effective rate was better in LLTG than CG ( P  = .006) for all patterns of audiogram, and there were statistically significant differences in flat audiogram ( P  = .043). Conclusions Lipid‐lowering therapy can improve the curative effect of sudden hearing loss patients combined with hyperlipidemia. There was a significant difference in the total effective rate of TG Group after lipid intervention, suggesting that there might be causal relationship between TG and sudden hearing loss. There was a significant difference in the total effective rate between flat audiogram, which may suggest flat hearing loss was more likely caused by vascular dysfunction.

    Abstract: What are the dynamic expression features of plasma microRNAs (miRNAs) during the peri-implantation period in women with successful pregnancy via single frozen-thawed blastocyst transfer? SUMMARY ANSWER There is a significant change in the plasma miRNA expression profile before and after blastocyst transfer, during the window of implantation. WHAT IS KNOWN ALREADY The expression of miRNAs in peripheral blood has indicative functions during the peri-implantation period. Nevertheless, the dynamic expression profile of circulating miRNAs during the peri-implantation stage in women with a successful pregnancy has not been studied. STUDY DESIGN, SIZE, DURATION Seventy-six women treated for infertility with a single frozen-thawed blastocyst transfer in a natural cycle were included in this study. Among them, 57 women had implantation success and a live birth, while 19 patients experienced implantation failure. Peripheral blood samples were collected at five different time points throughout the peri-implantation period, including D0 (ovulation day), D3, D5, D7, and D9 in this cycle of embryo transfer. The plasma miRNAs in women with blastocyst transfer were isolated, sequenced, and analyzed. PARTICIPANTS/MATERIALS, SETTING, METHODS Peripheral blood samples were collected in EDTA tubes and stored at −80°C until further use. miRNAs were isolated from blood, cDNA libraries were constructed, and the resulting sequences were mapped to the human genome. The plasma miRNAs were initially analyzed in a screening cohort (n = 34) with successful pregnancy. Trajectory analysis, including a global test and pairwise comparisons, was performed to detect dynamic differentially expressed (DE) miRNAs. Fuzzy c-means clustering was conducted for all dynamic DE miRNAs. The correlation between DE miRNAs and clinical characteristics of patients was investigated using a linear mixed model. Target genes of the miRNAs were predicted, and functional annotation analysis was performed. The expression of DE miRNAs was also identified in a validation set consisting of women with successful (n = 23) and unsuccessful (n = 19) pregnancies. MAIN RESULTS AND THE ROLE OF CHANCE Following small RNA sequencing, a total of 2656 miRNAs were determined as valid read values. After trajectory analysis, 26 DE miRNAs (false discovery rate & lt; 0.05) were identified by the global test, while pairwise comparisons in addition identified 20 DE miRNAs. A total of seven distinct clusters representing different temporal patterns of miRNA expression were discovered. Nineteen DE miRNAs were further identified to be associated with at least one clinical trait. Endometrium thickness and progesterone level showed a correlation with multiple DE miRNAs (including two of the same miRNAs, hsa-miR-1-3p and hsa-miR-6741-3p). Moreover, the 19 DE miRNAs were predicted to have 403 gene targets, and there were 51 (12.7%) predicted genes likely involved in both decidualization and embryo implantation. Functional annotation for predicted targets of those clinically related DE miRNAs suggested the involvement of vascular endothelial growth factor and Wnt signaling pathways, as well as responses to hormones, immune responses, and cell adhesion-related signaling pathways during the peri-implantation stage. LIMITATIONS, REASONS FOR CAUTION Although the RNA sequencing results revealed the global dynamic changes of miRNA expression, further experiments examining the clinical significance of the identified DE miRNAs in embryo implantation outcome and the relevant regulatory mechanisms involved are warranted. WIDER IMPLICATIONS OF THE FINDINGS Understanding the dynamic landscape of the miRNA transcriptome could shed light on the physiological mechanisms involved from ovulation to the post-implantation stage, as well as identifying biomarkers that characterize stage-related biological process.     Abstract: Lots of research efforts have been devoted to increase the transmission capacity in optical communications using orbital angular momentum (OAM) multiplexing. To enable long-haul OAM mode transmission, an in-line OAM fiber amplifier is desired. A ring-core fiber (RCF) is considered to be a preferable design for stable OAM mode propagation in the fiber. Here, we demonstrate an OAM fiber amplifier based on a fabricated ring-core erbium-doped fiber (RC-EDF). We characterize the performance of the RC-EDF-assisted OAM fiber amplifier and demonstrate its use in OAM multiplexing communications with OAM modes carrying quadrature phase-shift keying (QPSK) and quadrature amplitude modulation (QAM) signals. The amplification of two OAM modes over four wavelengths is demonstrated in a data-carrying OAM-division multiplexing and wavelength-division multiplexing system. The obtained results show favorable performance of the RC-EDF-assisted OAM fiber amplifier. These demonstrations may open up new perspectives for long-haul transmission in capacity scaling fiber-optic communications employing OAM modes.

    Abstract: Objectives: Pulmonary air leakage is a common complication following lung resection. We have designed a new method combining oxidized regenerated cellulose and fibrin glue to cover the intersegmental plane in clinical lung segmentectomy to prevent postoperative air leakage. In this study, an excised porcine lung segmentectomy model was created to validate its adhesive strength and effect on reducing air leakage. Methods: In the pre-experiment, six different larger lung segments were separated using electrocautery on the fresh isolated porcine lungs ( n = 5 in each group). The air leakage degree and operation time of the lung segments were comprehensively evaluated to select the most suitable target segment for establishing the ex vivo porcine lung segmentectomy models. In the experiment, according to the different materials covered on the intersegmental plane, these models were randomly divided into four groups: group A used fibrin glue and oxidized regenerated cellulose (ORC) mesh ( n = 20); group B used fibrin glue and polyglycolic acid (PGA) sheet (n = 20); group C used fibrin glue ( n = 20); group D was the blank control group ( n = 20). The minimum air leakage pressure (MALP) of the selected target segment in each group was measured using a stepwise increase of airway pressure, and histological assessment was performed on the sealed area samples from the four groups. Results: The operation time of the a segment of the right cranial lobe (R1a) was shorter than that of other segments ( p & lt; 0.05), and there was no significant difference in the air leakage pressures between the six isolated segments ( p = 0.76); thus, R1a was chosen for segmentectomy. In addition, the MALP was significantly higher in group A (41.8 ± 4.5 cmH 2 O) than in groups C (28.1 ± 2.3 cmH 2 O) and D (17.3 ± 1.2 cmH 2 O) (both p & lt; 0.001). The MALP of group B (69.5 ± 5.2 cmH 2 O) was significantly higher than that of group A ( p & lt; 0.001), whereas that of group C was significantly higher than that of group D ( p & lt; 0.001). Histological examination confirmed that the combined use of fibrin glue and ORC or PGA patch adhered more firmly to the intersegmental plane than that of fibrin glue alone, although some gaps could be seen between the fibrin glue and the surface of the lung segments in group C. Conclusion: The application of ORC combined with fibrin glue on the intersegmental plane has a good sealing performance in the ex vivo porcine lung segmentectomy model, suggesting that ORC may be an effective alternative material to replace PGA sheet to combine with fibrin glue for preventing air leakage after segmentectomy.

    Abstract: Chiral mirrors can produce spin selective absorption for left-handed circularly polarized (LCP) or right-handed circularly polarized (RCP) waves. However, the previously proposed chiral mirror only absorbs the designated circularly polarized (CP) wave in the microwave frequency band, lacking versatility in practical applications. Here, we propose a switchable chiral mirror based on a pair of PIN diodes. The switchable chiral mirror has four working states, switching from the handedness-preserving mirror to the LCP mirror, RCP mirror, and perfect absorber. The basis of these advances is to change the chirality of two-dimensional (2D) chiral metamaterials and the circular conversion dichroism related to it, which is the first report in the microwave frequency band. Surface current distributions shed light on how switchable chiral mirrors work by handedness-selective excitation of reflective and absorbing electric dipole modes. Energy loss distributions verify the working mechanism. The thickness of the switchable chiral mirror is one-tenth of the working wavelength, which is suitable for integrated manufacturing. The measurement results are in good agreement with the simulation results.
