    Abstract: Combinatorial antibody libraries not only effectively reduce antibody discovery to a numbers game, but enable documentation of the history of antibody responses in an individual. The severe acute respiratory syndrome coronavirus 2 (SARS‐CoV‐2) pandemic has prompted a wider application of this technology to meet the public health challenge of pandemic threats in the modern era. Herein, a combinatorial human antibody library constructed 20 years before the coronavirus disease 2019 (COVID‐19) pandemic is used to discover three highly potent antibodies that selectively bind SARS‐CoV‐2 spike protein and neutralize authentic SARS‐CoV‐2 virus. Compared to neutralizing antibodies from COVID‐19 patients with generally low somatic hypermutation (SHM), these three antibodies contain over 13–22 SHMs, many of which are involved in specific interactions in their crystal structures with SARS‐CoV‐2 spike receptor binding domain. The identification of these somatically mutated antibodies in a pre‐pandemic library raises intriguing questions about the origin and evolution of these antibodies with respect to their reactivity with SARS‐CoV‐2.

    Abstract: α-Linolenic acid (ALA) is a plant-derived omega-3 unsaturated fatty acid that is rich in flaxseed oil (FO). The effect of FO on bone health is controversial. This study aims to evaluate the effect of FO on bone damage induced by a high-fat diet (HFD) and to explore the possible mechanism. Methods Male Sprague-Dawley rats were fed a normal control diet (NC, 10% fat), FO diet (NY, 10% fat), HFD (60% fat), or HFD containing 10% FO (HY, 60% fat) for 22 weeks. Micro CT and three-point bending tests were conducted to evaluate bone microstructure and biomechanics. Serum was collected for the detection of ALP, P1NP, and CTX-1. Rat primary osteoblasts (OBs) were treated with different concentrations of ALA with or without palmitic acid (PA) treatment. The ALP activity, osteogenic-related gene and protein expression were measured. Results Rats in the HFD group displayed decreased biomechanical properties, such as maximum load, maximum fracture load, ultimate tensile strength, stiffness, energy absorption, and elastic modulus, compared with the NC group ( p   〈  0.05). However, HY attenuated the HFD-induced decreases in bone biomechanical properties, including maximum load, maximum fracture load, and ultimate tensile strength ( p   〈  0.05). Trabecular bone markers such as trabecular volume bone mineral density (Tb. vBMD ), trabecular bone volume/total volume (Tb. BV/TV), trabecular number (Tb. N), trabecular thickness (Tb. Th) were decreased, trabecular separation (Tb. Sp) and the structure model index (SMI) were increased in the HFD group compared with the NC group, and all parameters were remarkably improved in the HY group compared to the HFD group (p  〈  0.05). However, cortical bone markers such as cortical volume bone mineral density (Ct. vBMD), cortical bone volume/total volume (Ct. BV/TV) and cortical bone thickness (Ct. Th) were not significantly different among all groups. Moreover, the serum bone formation markers ALP and P1NP were higher and the bone resorption marker CTX-1 was lower in the HY group compared with levels in the HFD group. Compared with the NC group, the NY group had no difference in the above indicators. In rat primary OBs, PA treatment significantly decreased ALP activity and osteogenic gene and protein (β-catenin, RUNX2, and osterix) expression, and ALA dose-dependently restored the inhibition induced by PA. Conclusions FO might be a potential therapeutic agent for HFD-induced bone loss, most likely by promoting osteogenesis.

    Abstract: Visceral adipose tissue-derived serine protease inhibitor (vaspin), an adipose-derived hormone, exhibits various biological functions. Recently, studies showed that vaspin is closely related to bone metabolism. However, how vaspin influences bone formation and its underlying mechanisms in high fat-induced obese rats and rat primary osteoblasts (OBs) are not fully understood. In this study, the effects of vaspin on bone mechanical parameters and microarchitecture were evaluated. Methods A total of 40 male Sprague-Dawley (SD) rats at 5-week old were fed with high fat diet (HFD) and normal diet (ND) for 12 weeks followed by treatment of vaspin for 10 weeks. Micro CT and three-point bending tests were conducted to evaluate bone microstructure and biomechanics. The alkaline phosphatase (ALP) activity, expression of Runt-related transcription factor 2 (Runx2), Osterix (Osx), Collegen alpha1 (Colla1) procollagen I N-terminal peptide (PINP), C-telopeptide of type I collagen (CTX), Smad2/3 and p-Smad2/3 was detected by different methods. Results Our data indicated that, compared with ND rats, HFD rats exhibited high body weight, decreased bone strength and deteriorative bone quality. In contrast, vaspin reduced the body weight, improved the whole body metabolic status, enhanced bone strength, trabecular bone mass, and expression of Runx2, Osx, PINP, and decreased the expression level of plasma CTX. In vitro studies showed that vaspin promoted osteogenic differentiation and ALP activity in rat primary OBs in a dose dependent manner. Vaspin also upregulated mRNA expression of osteogenesis-related genes Runx2, Osx and Colla1 and protein expression of Runx2, Smad2/3 and p-Smad2/3. Conclusions Our results indicated that vaspin protects against HFD-induced bone loss, and promotes osteogenic differentiation by activating the Smad2/3-Runx2 signaling pathway .

    Abstract: Due to their tense mining succession relationship, gob-side roadways may undergo significant deformation under multi-mining pressure. In this article, many methods, such as on-site research, a theoretical analysis, a numerical simulation and an industrial experiment, are used to research the mechanism of asymmetric floor heave in a gob-side coal roadway affected by mining pressure during the mining of extra-thick coal seams. Our main research is as follows: (1) By monitoring the floor deformation in the roadway on site, it is concluded that the roadway floor shows asymmetry, indicating that the floor displacement near the coal pillar side is relatively large. (2) Based on a lateral overburden structure model of the roadway, the calculation formulas of the horizontal vertical stress caused by the roadway excavation and the excavation of the upper working face are derived separately, and the vertical stress coupling curves on both sides of the roadway during the mining of the upper working face are obtained through a numerical simulation. It is concluded that the cause of the asymmetric floor heave in the roadway is an uneven distribution of vertical stress. (3) The numerical simulation shows a symmetrical distribution of the floor displacement curve during the roadway excavation with a max. displacement of 49.5 mm. The floor displacement curve during the mining of the upper working face is asymmetric with a max. displacement of 873 mm at a distance of 1 m from the central axis near the coal pillar side. The range of the plastic zone in the roadway gradually expands with the mining of the upper working face, and the maximum depth of floor failure is 5.5 m. (4) According to the cooperative control principle of “roof + two sides + floor”, an asymmetric floor heave joint control scheme of “floor leveling + anchor cable support + concrete hardening” is proposed. The floor deformation monitoring results indicate that the max. floor heave at the measurement point near the coal pillar in the roadway is 167 mm, and the floor heave is effectively controlled.

    Abstract: Under the condition of fully mechanized top coal caving in close-distance coal seams, the surrounding rock of the stopping space easily loses stability during the withdrawal of mining equipment in the working face because the lower coal seam working face is located under the goaf and the overburden rock has a large range of complex interaction. Field investigation, theoretical analysis, laboratory experiment, similar simulation experiment, numerical simulation, and field industrial tests are used to carry out the research on the stability and control of the surrounding rock in the large section stopping space under the goaf in this paper. The research conclusions are as follows. (1) It is determined that the lower coal seam working face can only stop mining under the goaf, and the reasonable stopping position under the goaf should ensure that the key block fracture line of the main roof is behind the support. (2) The interaction law between the main roof’s key blocks of the upper and lower coal seams is analyzed, and the catastrophic conditions for sliding instability and rotary instability of the main roof’s key blocks of the upper and lower coal seams are obtained. (3) “Anchorage with push and pull equipment-Embedded anchorages and trays” integral anchoring technology is developed. The dimensions of the push and pull equipment are determined. (4) Through numerical simulation of the distribution characteristics of the anchor cable pre-stress field, the asymmetric control scheme of “Partition long and short anchor cables + Integral polyurethane mesh + Embedded anchorages and trays for roof protection” is determined. The rock pressure observation shows that the withdrawal of the working face equipment is implemented safely.

    Abstract: Multi-coal-seam mining creates surrounding rock control difficulties, because the mining of a coal face in one seam can affect coal faces in another. We examine the effects of multi-coal-seam mining on the evolution of the deviatoric stress distribution and plastic zone in the roadway surrounding rock. In particular, we use numerical simulation, theoretical calculation, drilling detection, and mine pressure observation to study the distribution and evolution characteristics of deviatoric stress on Tailgate 8709 in No. 11 coal seam in Jinhuagong mine when the N8707 and N8709 coal faces in No. 7-4 coal seam and the N8707 and N8709 coal faces in No. 11 coal seam are mined. The evolution laws of deviatoric stress and the plastic zone of roadway surrounding rock in the advance and behind sections of the coal face are studied, and a corresponding control technology is proposed. The results show that the peak value of deviatoric stress increases with the advance of the coal face, and the positions of the peak value of deviatoric stress and the plastic zone become deeper. The deflection angle of the peak stress after mining at each coal face and the characteristics of the peak zone of deviatoric stress and the plastic zone of the roadway surrounding rock under the disturbance of multi-coal-seam mining are determined. In conclusion, the damage range in the roadway roof in the solid-coal side and coal pillar is large and must be controlled. A combined support technology based on high-strength and high pretension anchor cables and truss anchor cables is proposed; long anchor cables are used to strengthen the support of the roadway roof in the solid-coal side and coal pillar. The accuracy of the calculated plastic zone range and the reliability of the combined support technology are verified through drilling detection and mine pressure observation on site. This research can provide a point of reference for roadway surrounding rock control under similar conditions.

    Abstract: Background . This study explored the therapeutic efficacy of standard triple therapy combined with sucralfate suspension gel as well as the mechanisms of action in mouse models of H . pylori infection. Materials and Methods . C57BL/6J mice were randomly divided into 5 groups: NC (natural control), HP ( H . pylori infection), RAC (rabeprazole, amoxicillin, and clarithromycin), RACS (RAC and sucralfate suspension gel), and RACB (RAC and bismuth potassium citrate). HE staining and electron microscopy were performed to estimate histological and ultrastructural damages. The IL-8, IL-10, and TNF- α of gastric antrum tissues were measured by immunohistochemistry and qRT-PCR. ZO-1 and Occludin were also detected with immunohistochemistry. The genomes of gastric and fecal microbiota were sequenced. Results . The eradication rate of H . pylori in the RACS group was higher than the RAC group. RACS therapy had protective effects on H . pylori -induced histological and ultrastructural damages, which were superior to the RAC group. RACS therapy reduced the protein and mRNA levels of IL-8 compared with the RAC group. The expression of Occludin in the RACS group was significantly higher than that of the RAC group. The composition of gastric and fecal microbiota for RACS was similar to the RACB group according to PCA. Conclusions . The RACS regimen eradicated H . pylori infection effectively and showed RACS had protective effects against H . pylori -induced histological and ultrastructural damage. The mechanisms of RACS effects included decreasing IL-8, enhancing Occludin, and transforming gastric microbiota. Moreover, RACS and RACB have a similar effect on gastrointestinal flora.
