Abstract: The overuse of antibiotics is losing its effectiveness due to increased antibiotic resistance in bacteria. Over the last two decades, the evaluation of antibiotic residues in the environment has greatly attracted the attention of researchers around the world. The more new groups of antibiotics were validated to use in medical treatment, the more antibiotic residues were discharged into the environment. In this work, a combined SPE-UPLC-MS/MS method was developed and validated for simultaneously analyzing 9 antibiotic compounds belonging 3 different groups which are tetracyclines (tetracycline – TET, chlortetracycline – CHL, oxytetracycline – OXY), fluoroquinolones (ciprofloxacin – CIP, norfloxacin – NOR, ofloxacin – OFL) and macrolides (azithromycin – AZI, erythromycin – ERY, clarithromycin – CLA) in surface water. All target analytes were separated on a reversed-phase column (Water BEH C18 column, 1.7 µm particle size, 100 x 2.1 mm) coupled with a mass in tandem employing positive electrospray ionization (+ESI). All target analytes were well separated with an overall run time of 16 minutes. The limit of detection was from 0.2 to 10 µg L-1. The extraction process has been successfully optimized using the HLB column (Oasis, 6mL, 200mg, Waters) with recoveries from 71 to 125%. The optimized method was finally applied to analyze ten surface water samples (lake and river). OFL, CLA, and ERY were most found in lake samples at concentrations from 35 – 570.3 ng.L-1 while only CIP was found in all river samples. The other compounds were also detected in both types of samples but the concentrations were lower than the quantification limit.

Abstract: An ultrahigh-performance liquid chromatography in combination with high-resolution mass spectrometry Thermo Q-Extractive Focus Orbitrap MS has been introduced for analysis of multiclass pesticides in vegetable samples collected in Hanoi, Vietnam. Multiclass pesticides were separated on the Thermo Hypersil Gold PFP column utilizing a gradient of the mobile phase consisting of 5 mM ammonium formate, 0.1% formic acid in deionized water, and methanol. The target analytes were detected in the full-scan mode on Thermo Scientific Q-Exactive Focus Orbitrap MS for quantitation at the optimum operating conditions. These conditions included, but not limit to, the resolution of 70000 at the full width at half maximum in both positive and negative mode, mass range from 80 to 1000  m/z , and optimized parameters for the heated electrospray ionization source. The identification of the analytes in real samples was based on retention times, mass to charge ratios, mass accuracies, and MS/MS spectra at the confirmation mode with the inclusion list of target analytes. The mass accuracies of target analytes were from −4.14 ppm (dinotefuran) to 1.42 ppm (cinosulfuron) in the neat solvent and from −3.91 ppm (spinosad D) to 1.29 ppm (cinosulfuron) in the matrix-matched solution. Target analytes in the vegetable-based matrix were extracted by the QuEChERS method. Some critical parameters of the analytical method such as linearity, repeatability, limit of detection, and limit of quantitation have been evaluated and implemented. Excellent LOD and LOQ of the developed method were achieved at the range of 0.04–0.85 and 0.13–2.9  μ g·kg −1 , respectively. Intraday and interday repeatability of the analytical signal (peak area, n = 6 ) of the developed method were below 3% and 10%, correspondingly. The matrix effect, extraction recovery, and overall recovery were fully investigated by spiking experiments. Experimental results demonstrated that the ionization suppression or enhancement was the main contribution on the overall recoveries of target analytes. Finally, the in-house validated method was applied to pesticides screening in vegetables samples in local villages in Hanoi, Vietnam. The concentrations of all target analytes were below limit of quantitation and lower than US-FDA or EU maximum residue levels.

Abstract: The study is aimed to develop a relevant synbiotic to promote growth performance of the whiteleg shrimp, Litopenaeus vannamei. For this, four common natural fiber extracts from Arcera banana, Siamese banana, yellow sweet potato, and white sweet potato were screened for supporting the growth of Lactobacillus sp. which was isolated from whiteleg shrimp intestines with probiotic activity, prebiotic score, and ability to induce bacterial enzyme activities of protease, leu-aminopeptidase, and a-amylase. Results showed that Lactobacillus sp. was able to utilize all extracts from banana and sweet potato as the sole carbon sources. At 24 hours of culture, the growth of Lactobacillus sp. was highest after adding the extract from white sweet potato as the sole carbon source. Considering pathogenic bacteria, including Vibrio parahaemolyticus, white sweet potato extract had the highest prebiotic score with a mean of 0.25 as compared with those of V. harveyi with a mean of 0.16. White sweet potato extract induced the highest activities of protease. These results indicated that white sweet potato extract was more suitable for combining with Lactobacillus sp. as a synbiotic for shrimp culture.

Abstract: The aim of this study is to characterize the sulfur oxidizing bacteria (SOB) isolates from the sediments of extensive shrimp ponds for recommending the use of this group for water quality management in aquaculture. Sediment samples were collected from 12 extensive shrimp ponds located in Tra Vinh, Soc Trang, Bac Lieu, and Ca Mau provinces. To screen the potential sulfur oxidizing bacteria, medium was amended with sodium thiosulfate, and the sulfate ion production ability and sulfur oxidase enzyme activity of the isolates were measured spectrophotometrically. Results showed that 30 isolates grew on the thiosulfate agar medium. Among these, only five isolates reduced the pH of the growth medium and showed high sulfur oxidase activity and production of sulfate ion when isolates were inoculated with thiosulfate as a substrate. Physiological and biochemical tests indicated that five selected isolates were Gram negative, short rod, non-motile, non-spore forming, negative for oxidase reaction, and positive for catalase reaction. The isolates SOBTB1.1 and SOBTB6.2 showed the significantly higher sulfur oxidase activity and production of sulfate ion compared to other isolates. SOBTB6.2 isolate produced sulfate ion and exhibited higher sulfur oxidase activity at pH4-5, followed by pH6-7. It is, therefore, suggested that the SOBTB 1.1 and SOBTB6.2 could be promising sulfur oxidizers for further research and uses in aquaculture.

Abstract: Current research on the photocatalytic activity of TiO2 mainly focuses on its nano- or micro-particle forms, which are difficult to recycle and apply in real engineering applications. This study aims to apply a small pilot of TiO2 in the ceramic form to remove carbamazepine (CBZ) from an aqueous solution under simulated sunlight. A high removal efficiency up to 〉 99% was shown in a 5 mg L−1 CBZ solution after 6 h of irradiation with a total energy of 150.92 kJ. The kinetic degradation was not affected in an alkaline solution (at pH 7, pH 10, and pH 13) but was faster under acidic conditions (pH 2) in which CBZ existed in the protonated form. The presence of NO3− (10–50 mg L−1) slightly affected the photodegradation of CBZ while humic acid significantly reduced the photocatalytic activity. In addition, the presence of major ions in water also had a negative effect at concentrations between 10 and 50 mg L−1. The MS/MS was used to identify the transformation products of CBZ, and a possible degradation mechanism was proposed. The toxicity of CBZ and the by-products was primarily evaluated. The results showed that TiO2 ceramics show high reusability and stability with a photocatalytic performance of 〉 95% and a mass loss of 〈 5% after 90 degradation cycles.

Abstract: Antimicrobial stewardship (AMS) programmes have been implemented around the world to guide rational use of antibiotics but implementation is challenging, particularly in low- and middle-income countries, including Vietnam. Understanding factors influencing doctors’ prescribing choices for empirical treatment can help design AMS interventions in these settings. Objectives To understand doctors’ choices of antibiotics for empirical treatment of common bacterial infections and the factors influencing decision-making. Methods We conducted a cross-sectional survey among medical professionals applying for a postgraduate programme at Hanoi Medical University, Vietnam. We used a published survey developed for internal medicine doctors in Canada. The survey was self-administered and included four clinical scenarios: (i) severe undifferentiated sepsis; (ii) mild undifferentiated sepsis; (iii) severe genitourinary infection; and (iv) mild genitourinary infection. Results A total of 1011/1280 (79%), 683/1188 (57.5%), 718/1157 (62.1%) and 542/1062 (51.0%) of the participants selected combination therapy for empirical treatment in scenarios 1, 2, 3 and 4, respectively. Undifferentiated sepsis (OR 1.82, 95% CI 1.46–2.27 and 2.18, 1.51–3.16 compared with genitourinary) and severe infection (1.33, 1.24–1.43 and 1.38, 1.21–1.58 compared with mild) increased the likelihood of choosing a combination therapy and a carbapenem regimen, respectively. Participants with higher acceptable minimum threshold for treatment coverage and young age were also more likely to prescribe carbapenems. Conclusions Decision-making in antibiotic prescribing among doctors in Vietnam is influenced by both disease-related characteristics and individual factors, including acceptable minimum treatment coverage. These findings are useful for tailoring AMS implementation in Vietnam and other, similar settings.
