    Abstract: The purpose of this study was to determine the phytochemical compositions and to evaluate the biological activity of the stem bark of Dialium cochinchinense. The phytochemical analysis showed the presence of steroids, terpenoids, anthraquinones, saponins, and phenolics in the stem bark of D. cochinchinense. The optimal extraction conditions were 70% ethanol at 30°C for 40min with a material/solvent ratio of 1/20. Under the optimal conditions, the corresponding predicted response value for the total phenolic content was 100.80 ± 0.40 mg GAE/g DW. The evaluation of antioxidant activity indicated that the polyphenol-rich extract was a good source of antioxidants as measured by the DPPH assay with the IC50 of 3.81 ± 0.58 μg mL-1. The extract also demonstrated a strong a-glucosidase inhibitory activity with the IC50 value of 2.14 ± 0.05 μg/mL. Therefore, D. cochinchinense could be useful as a potential preventive intervention for free radicals in mediated diseases as well as an antioxidant drug and a potential source in treating diabetes mellitus in the pharmaceutical industry.

    Abstract: From marine sponge‐associated fungus Hamigera avellanea , thirteen secondary metabolites including a pair of undescribed alkaloid enantiomers (+)‐hamiavemin A (4 S ) (+)‐ 1 and (−)‐hamiavemin A (4 R ) (−)‐ 1 . Compound 1 was enantiomers resolved by the Chiralpak AS‐3 column, using a hexane/isopropanol mobile phase. Their structures were determined based on extensive analyses of HR‐ESI‐MS, 1D and 2D NMR spectra. The absolute configuration of (+)‐ 1 and (−)‐ 1 were assigned tentatively by ECD calculations. Among the isolates, compound 6 showed strongest antibacterial activity against Enterococcus faecalis , Staphylococcus aureus , Bacillus cereus , Escherichia coli , Salmonella enterica , and Candida albicans with the MIC values of 2, 2, 16, 32, 64, and 16 μg/mL, respectively, which were stronger than that of the positive control compound, kanamycin (MIC values ranging from 4 to 128 μg/mL). In addition, compounds 1 , 2 , and 9 showed moderate cytotoxic activity against three cancer cell lines, HepG2, A549, and MCF‐7 with the IC50 values ranging from 55.35±1.70 to 83.02±2.85 μg/mL.

    Abstract: BACKGROUND: Spirulina platensis contains a large amount of chlorophylls, chlorophyll a, that are starting materials to synthesize functionalized chlorins. Chlorin e6 (Ce6) as well as its derivatives are second generation sensitizers using in photodynamic therapy (PDT) of various cancers. In this study, we transfer chlorophyll a of S. platensis to Ce6derivatives and determine their several applications. AIM: to evaluate the effects of Ce6 derivatives to treat cancer cells. METHODS: Ce6 trimethylester was created from methyl pheophorbide a2 in S. platensis provided by the Hidumi Company, Nghe An province, Viet Nam. Hela cells were incubated with Ce6 trimethylester and the irradiated with the diode laser dose of 1.2 J/cm2/min through the system of filters £ 650 nm. MTT assay and clonogenic assay were used to determine survival rate and cloning efficiency of cells. Antimicrobial effect of Ce6 trimethylester with halogen light were studied with Propionibacterium acnes VTCC 0218 and Staphylococcus aureus VTCC 0173. RESULTS: From dry biomass (700 g) of S. platensis, after extracting chlorophyll a and methanolysis, 4.2 g of methyl pheophorbide a was obtained. The reaction to give Ce6 trimethylester with 82% yield was performed with potassium hydroxide (KOH) in MeOH/THF/CHCl3. After irradiation with a 650 nm laser at 1.2 J, the cell viability in all samples decreased with Ce6 trimethylester treatment, the survival declining trend of Hela cells treated with Ce6 trimethylester were proportional when concentration of Ce6 trimethylester increased. The rate of colony formation was declined as the concentration of Ce6 trimethylester treated was increased. The growth of both S. aureus and P. acnes can be inactivated by Ce6 trimethylester PDT. The MIC99 value against P. acnes VTCC 0218 and S. aureus VTCC 0173 of Ce6 trimethylester with halogen light was 1.25 μg/ml. CONCLUSION: The Ce6 trimethylester from S. platensis cultivated in Viet Nam could be used as a potential photosentizer for photodynamic therapy for treatment of cancer and acne.

    Abstract: The marine environmental conditions are particularly different from terrestrial ones; recent studies have showed that marine actinomycetes might produce many novel compounds with good biological activity. The objective of this study is to isolate and screen actinomycetes strains from the marine environment with activity against pathogenic microorganisms. Fifty strains of actinomycetes were isolated from 40 samples including: sediments, sponges, soft corals, echinoderms... collected from Van Phong Bay area of  Khanh Hoa province, Vietnam. The strains fermented in A+ medium and fermentation broths were extracted 5 times with ethyl acetate. The extracts were evaporated under reduced  pressure to yield crude extracts. Quantitative assay was used to determine MIC (minimum inhibitory concentration) of extract against 7 test strains, including three Gram-negative bacteria (Escherichia coli ATCC25922, Pseudomonas aeruginosa ATCC27853, Salmonella enterica ATCC13076), three Gram-positive bacteria (Enterococcus faecalis ATCC29212, Staphylococcus aureus ATCC25923, Bacillus cereus ATCC 14579), and the yeast Candida albicans ATCC10231. The result of screening showed that: 18/50 strains have shown the antibacterial activities against at least 3 strains of tested microorganisms. In which, strains G650, G654, G657, G666, G675 inhibited three Gram-positive test microorganisms at MIC values from 8 to 64 µg/mL and C. albicans ATCC10231 with MIC values from 2 to 16 µg/mL. In addition, the two strains G657 and G666 had inhibitory effect on  P. aeruginosa ATCC27853 và S. enterica ATCC13076 with MIC value 128 µg/mL. Morphological and phylogenetic investigations based on 16S rRNA gene sequences of the selected five strains showed that: strain G650 belonged to species Streptomyces ardesiacus and strain G666 high similarity to the genus Streptomyces; strain G654 showed the highest similarity to the Salinispora arenicola species; strain G657 highest similarity to the Micromonospora aurantiaca; whereas G675 belonged to the Nocardiopsis flavescens species.

    Abstract: Marine environment is rich in natural product resources, including marine microorganisms, especially fungi which are not only seen as a potential source of highly applicable bioactive substances but also can provide for science new chemical structures. The objective of this study is to isolate and screen fungal strains with antibacterial activity from the marine environment. Twenty five strains of fungi were isolated from marine sediments of Thanh Lan, Co To island and assessed on antibiotic activity against 7 tested microbial strains, including three Gram-negative bacteria (Escherichia coli ATCC25922, Pseudomonas aeruginosa ATCC27853, Salmonella enterica ATCC13076), three Gram-positive bacteria (Enterococcus faecalis ATCC29212, Stapphylococus aureus ATCC25923, Bacillus cereus ATCC 13245), and the yeast Candida albicans ATCC10231. The minimum inhibitory concentration (MIC) against the tested microorganisms was determined for the crude extracts obtained from the culture broths after ethyl acetate extraction and vacuum rotary evaporation. Three strains with the highest antimicrobial activity M26, M30 and M45 were capable of inhibiting 4 - 5 of the 7 tested microorganisms with MIC values from 64 to 256 μg/ml, depending on each tested strain. Morphological and phylogenetic investigations based on 18S rRNA gene sequences of the three selected strains showed that strains M26 and M30 belonged to the genus Penicillium, whereas strain M45 belonged to the genus Neurospora. The sequences of 18S rRNA gene of three strains M26, M30 and M45 were registered on GenBank database with accession numbers: MH673730, MH673731, MH673732, respectively. Research results showed that marine environment has a great potential in isolation of fungal strains for the search for antibacterial substances as well as other biologically active compounds.

    Abstract: Actinomycetes has been extensively studied due to its ability to produce secondary compounds with high application value. Especially their antibiotic ability, more than 40% of antibiotics are derived from actinomycetes of which genus Streptomyces predominates. Moreover, marine actinomycetes have been being of great interest in recent years due to their ability to produce bioactive substances capable of providing diverse and novel chemical structures. In this study, from 8 samples of marine sediments collected in Vung Ang bay in Ha Tinh provinces, we have isolated 20 strains of actinomycetes. The strains were fermented in A1+ medium, the fermentation fluid was extracted 5 times with ethyl acetate, recovered sediment and determined antibacterial and cytotoxic activity. From the screening results, two strains with the highest antibacterial activity and highest cytotoxicity were selected ie., HT03 and HT06. Both strains had antagonistic activity of Enterococcus faecalis ATCC29212 with MICHT03= 32 μg/mL, MICHT06= 16μg/mL, with Stapphylococus aureus ATCC25923 with MICHT03= 64 μg/mL, MICHT06= 32 μg/mL and with Bacillus cereus ATCC 13245 has the same MIC = 16 μg / mL. In addition, the two strains HT03 and HT06 were able to strongly inhibit the yeast Candida albicans ATCC10231 with MICHT03= 16 μg/mL, MICHT06= 8 μg/mL. Especially, two strains, HT03 and HT06, exhibited very good toxicity on all 5 cancer cell lines (MCF-7 breast cancer cell; MDA-MB-231 breast cancer cell; lung cancer cell). NCI-H1975; HeLa cervical cancer cell; AGS gastric cancer cell) at both test concentrations of 30 µg/mL and 100µg/mL. By the analysis of 16S rRNA sequence, the results showed that the HT03 strain had the highest similarity (99.93%) to that of Streptomyces fradiaes and Streptomyces fradiae ATCC. The HT06 strain was defined to belong to Nocardiopsis synnemataformans with the 16S sequence identity of 99.89% to the Japanese standard Nocardiopsis synnemataformans DSM 44143 strain NBRC-102581.

    Abstract: Phản ứng multiplex-PCR đang được ứng dụng rộng rãi trong nghiên cứu phân tử bởi tính ưu việt của nó. Phản ứng multiplex-PCR có thể xác định đồng thời nhiều gen mục tiêu của các mồi trong cùng một phản ứng, giúp tiết kiệm thời gian, hóa chất, công sức và đặc biệt có hiệu quả cao. Trong nghiên cứu này, chúng tôi ứng dụng phản ứng multiplex-PCR để phát hiện kiểu huyết thanh và gen độc lực của vi khuẩn Streptococcus agalactiae gây bệnh xuất huyết trên cá rô phi vằn (Oreochromis niloticus). Phản ứng multiplex-PCR được tối ưu dựa trên kit My Taq™ HS Mix 2× và sử dụng chu trình nhiệt 2 bước. Kết quả đã xác định được vi khuẩn S. agalactiae gây bệnh trên cá rô phi vằn ở Việt Nam mang kiểu huyết thanh Ia và III, chứa 11-13 gen độc lực thuộc 3 nhóm yếu độc lực chính giúp phát động quá trình gây bệnh của vi khuẩn (yếu tố bám dính, yếu tố xâm nhập và yếu tố kháng miễn dịch). Điều này chứng tỏ các chủng nghiên cứu đều mang độc lực. Kết quả này cũng sẽ được ứng dụng để xác định độc lực của vi khuẩn S. agalactiae phục vụ phát triển chỉ thị phân tử (microsatellite và SNPs) trong chọn giống cá rô phi vằn kháng bệnh xuất huyết.
