Abstract: Ascorbic acid (AsA) and glutathione (GSH) are considered important factors to protect plants against abiotic stress. To investigate whether altered endogenous GSH and AsA affect seed germination, plant performance and the abiotic stress tolerance, GSH deficient mutant cad2-1 and AsA-deficient mutants (vtc2-4 and vtc5-2) were phenotypically characterized for their seed germination, shoot growth, photosynthetic activity and root architecture under abiotic stresses. The cad2-1, vtc2-4 and vtc5-2 mutants showed a decrease in osmotic and salt stress tolerance, in sensitivity to ABA during seed germination, and in plant performance under severe abiotic stresses. GSH deficiency in the cad2-1 plants affected plant growth and root development in plants exposed to strong drought, oxidative and heavy metal stress conditions. Plants with lower GSH did not show an increased sensitivity to strong salt stress (100 mM NaCl). In contrast, the mutants with lower AsA enhanced salt stress tolerance in the long-term exposures to strong salt stress since they showed larger leaf areas, longer primary roots and more lateral root numbers. Limitations on AsA or GSH synthesis had no effect on photosynthesis in plants exposed to long-term strong salt or drought stresses, whereas they effected on photosynthesis of mutants exposed to CdCl2. Taken together, the current study suggests that AsA and GSH are important for seed germination, root architecture, shoot growth and plant performance in response to different abiotic stresses, and their functions are dependent on the stress-inducing agents and the stress levels.

Abstract: Japanese Encephalitis vaccine has contributed to repel Japanese Encephalitis in young children in the national vaccination campaign. To qualitatively evaluate the protective efficiency of Japanese Encephalitis vaccine, a standard antiserum used as a positive control was performed in parallel with testing samples. A study was organized at National Institute for Control of Vaccines and Biologicals (NICVB) as following. Anti-sera were produced by 2-dose-injection of national standard vaccine to 28-day-old white mice, weighing 11- 13g. After blood collection and serum separation, the PRNT50 neutralizing antibody potency was evaluated in parallel with international standard antisera from NIBSC and antisera from Vabiotech manufacturer. Antiserum lot 0118 was produced with a neutralizing antibody potency PRNT50 of 1.74 to ensure criteria of a positive control standard for performing Japanese Encephalitis vaccine potency test.

Abstract: Abutilon indicum L. belonging to Malvaceae is used as traditional medicinal herbs to treat syphilis, hypoglycemia, hepatic disorders, and malaria... Because of the medicinal value of A. indicum, the aims of this study are the evaluation of bioactivities of A. indicum and production of transformed hairy root for pharmaceutical production. In this study, the reducing power of ethanol root extract and stem extract are higher than that of leave. The root extract exhibited potent brine shrimp (Artemia salina) lethality with LC50 37.04 μg/mL. The α-glucosidase and acetylcholinesterase inhibitory activities of the root extract are the highest. These results showed that roots are higher bioactivities than stems and leaves. This study induced successfully hairy roots of A. indicum L. via Agrobacterium rhizogenes ATCC 15834 in the plant cells. The frequency of hairy root and number of hairy root induction from the wounded sites of leaves are the highest (88.66 % and 8.66 roots). The stable introduction of the rolB and rolC genes of A. rhizogenes strain 15834 into A. indicum plants was confirmed by PCR analysis. Besides, the absence of the virD gene confirmed hairy roots as a bacteria-free. Subsequently, these results demonstrated that A. indicum, particularly the roots, have great potential as pharmacological values and hairy root production maybe used for pharmaceutical sources.

Abstract: Premna flavescens Wall. ex C. B. Clarke, a medicinal plant in the family Lamiaceae, was used for the treatment of arthritis in folk medicine. From the essential oil extracted from the leaves of this plant we have identified 51 components, with β-caryophyllene as the major component (26.3%). The essential oil showed a strong anti-inflammatory effect in vitro with an IC 50 of 5.88 µg/mL. DNA barcoding is an useful tool for species identification based on the standardized genomic DNA fragments, but the molecular database of P flavescens is still lacking from Genebank. A molecular species identification tool for P flavescens was developed for the first time using DNA barcoding. The sequences of rbcL, trnH–psbA, internal transcribed spacer, and 18S barcodes of P flavescens were submitted to the Genebank with the accession numbers MW553265, MW553266, MT935698.1, and MW485128.1, respectively. This will be highly useful for the molecular authentication of the marker samples.

Abstract: Aeroponics is considered as a potential method for the culture of herbal plants due to the high growth rate, quantity and quality enhancement of secondary metabolites, and substantial environmental progress associated with this method. The aim of this study was to develop a sufficient protocol for successful Urena lobata hairy root induction by Agrobacterium rhizogenes ATCC 15834, using a precursor and elicitor to enhance α-glucosidase inhibitory activity (GIA) of aeroponic hairy roots (AHRs) in greenhouse conditions. In this study, we found that the optimized procedure (10 min, Woody plant medium (WPM), 1/25 salt strength) had an outstanding effect with a reduction in the rooting time (RT), promotion of the rooting rate (RR), and increase in the fresh weight (FW) and dry weight (DW) compared with the original procedure (30 min, Murashige and Skoog (MS) medium, 1/25 salt strength) after 30 days of culture. The highest DW, GIA, flavonoid (FLA) and phenolic (PHEL) contents were observed for individual addition of 10 mM phenylalanine (PA) or 50 mM chitosan (CS) in the late exponential phase (eighth week) with 15 days of elicitation compared to the control AHRs. However, individual treatment was less effective than the combination of the two. Positive correlations among the GIA, FLA and PHEL indicate that AHRs accumulated phenolic compounds, leading to an increase in the GIA by a synergistic effect. In conclusion, the culture of Urena lobata AHRs with PA and CS is an efficient procedure to produce GIA material in greenhouse conditions.

Abstract: In this study, we aimed at evaluating in vitro and in vivo anti-inflammatory activity of various extracts of the rhizomes of Globba pendula Roxb. Three extracts ( n-hexane, ethyl acetate, and water) were screened for their inhibitory effect on NO production by lipopolysaccharide-stimulated RAW 264.7 macrophages. The ethyl acetate extract of G. pendula rhizomes (EGP) showed a potential effect with an IC 50 value of 32.45 µg/mL. For in vivo study, the ethyl acetate extract was further investigated for its anti-inflammatory effect using collagen antibody-induced arthritic mice (CAIA). The level of arthritis in experimental mice significantly reduced ( P  〈  .05) after treatment with EGP at a dose of 500 mg/kg body weight (b.w.). This study also revealed that EGP is orally non-toxic. Ethyl p-methoxy cinamate was identified as the main constituent of EGP, which may result in its anti-inflammatory effect.
