    Abstract: Aim: To conduct the clinical, genetic, and molecular characterization of 494 Han Chinese subjects with tic disorders (TD). Methods: In the present study, we performed the mutational analysis of 22 mitochondrial tRNA genes in a large cohort of 494 Han Chinese subjects with TD via Sanger sequencing. These variants were then assessed for their pathogenic potential via phylogenetic, functional, and structural analyses. Results: A total of 73 tRNA gene variants (49 known and 24 novel) on 22 tRNA genes were identified. Among these, 18 tRNA variants that were absent or present in & lt;1% of 485 Chinese control patient samples were localized to highly conserved nucleotides, or changed the modified nucleotides, and had the potential structural to alter tRNA structure and function. These variants were thus considered to be TD-associated mutations. In total, 25 subjects carried one of these 18 putative TD-associated tRNA variants with the total prevalence of 4.96%. Limitations: The phenotypic variability and incomplete penetrance of tic disorders in pedigrees carrying these tRNA mutations suggested the involvement of modifier factors, such as nuclear encoded genes associated mitochondrion, mitochondrial haplotypes, epigenetic, and environmental factors. Conclusion: Our data provide the evidence that mitochondrial tRNA mutations are the important causes of tic disorders among Chinese population. These findings also advance current understanding regarding the clinical relevance of tRNA mutations, and will guide future studies aimed at elucidating the pathophysiology of maternal tic disorders.

    Abstract: Low‐dimensional perovskites have gained increasing attention recently, and engineering their material phases, structural patterning and interfacial properties is crucial for future perovskite‐based applications. Here a phase and heterostructure engineering on ultrathin perovskites, through the reversible cation exchange of hybrid perovskites and efficient surface functionalization of low‐dimensional materials, is demonstrated. Using PbI 2 as precursor and template, perovskite nanosheets of varying thickness and hexagonal shape on diverse substrates is obtained. Multiple phases, such as PbI 2 , MAPbI 3 and FAPbI 3 , can be flexibly designed and transformed as a single nanosheet. A perovskite nanosheet can be patterned using masks made of 2D materials, fabricating lateral heterostructures of perovskite and PbI 2 . Perovskite‐based vertical heterostructures show strong interfacial coupling with 2D materials. As a demonstration, monolayer MoS 2 /MAPbI 3 stacks give a type‐II heterojunction. The ability to combine the optically efficient perovskites with versatile 2D materials creates possibilities for new designs and functionalities.

    Abstract: Histone chaperones facilitate DNA replication and repair by promoting chromatin assembly, disassembly and histone exchange. Following histones synthesis and nucleosome assembly, the histones undergo posttranslational modification by different enzymes and are deposited onto chromatins by various histone chaperones. In Tetrahymena thermophila, histones from macronucleus (MAC) and micronucleus (MIC) have been comprehensively investigated, but the function of histone chaperones remains unclear. Histone chaperone Nrp1 in Tetrahymena contains four conserved tetratricopepeptide repeat (TPR) domains and one C-terminal nuclear localization signal. TPR2 is typically interrupted by a large acidic motif. Immunofluorescence staining showed that Nrp1 is located in the MAC and MICs, but disappeared in the apoptotic parental MAC and the degraded MICs during the conjugation stage. Nrp1 was also colocalized with α-tubulin around the spindle structure. NRP1 knockdown inhibited cellular proliferation and led to the loss of chromosome, abnormal macronuclear amitosis, and disorganized micronuclear mitosis during the vegetative growth stage. During sexual developmental stage, the gametic nuclei failed to be selected and abnormally degraded in NRP1 knockdown mutants. Affinity purification combined with mass spectrometry analysis indicated that Nrp1 is co-purified with core histones, heat shock proteins, histone chaperones, and DNA damage repair proteins. The physical direct interaction of Nrp1 and Asf1 was also confirmed by pull-down analysis in vitro. The results show that histone chaperone Nrp1 is involved in micronuclear mitosis and macronuclear amitosis in the vegetative growth stage and maintains gametic nuclei formation during the sexual developmental stage. Nrp1 is required for chromatin stability and nuclear division in Tetrahymena thermophila .

    Abstract: Inorganic oxides, alumina and silica, were successively modified on titanate whiskers surface. Enrolled into phenolic resin, the phenolic/whisker composites were prepared by melting molding, with intentions for wearing property. The alumina‐coated whisker surface fulfilled wear resistance, while the outer silica layer was less stiff for compatible with polymer matrix. The dual surface modifications improved the whiskers dispersion in the matrix, enhanced interfacial adhesion, and significantly triggered wear resistance of the composites. With 7.5 wt% multiply modified whiskers involved, the phenolic composite showed specific wear rate of 0.49 × 10 −5 mm 3 /Nm, remarkably lower than that of phenolic resin (4.79 × 10 −5 mm 3 /Nm). The worn surface of the composites detected by a scanning electron microscope showed abrasive wear with slight fatigue wear. Optical microscope images indicated that the whisker‐filled composite had a shallow scratch on the worn surface, which positively favored wear resistance.

    Abstract: Clinical target volumes (CTVs) and organs at risk (OARs) could be autocontoured to save workload. This study aimed to assess a convolutional neural network for automatic and accurate CTV and OARs in prostate cancer, while comparing possible treatment plans based on autocontouring CTV to clinical treatment plans. Methods Computer tomography (CT) scans of 217 patients with locally advanced prostate cancer treated at our hospital were retrospectively collected and analyzed from January 2013 to January 2019. A deep learning-based method, CUNet, was used to delineate CTV and OARs. A training set of 195 CT scans and a test set of 28 CT scans were randomly chosen from the dataset. The mean Dice similarity coefficient (DSC), 95th percentile Hausdorff distance (95HD), and subjective evaluation were used to evaluate the performance of this strategy. Predetermined evaluation criteria were used to grade treatment plans, and percentage errors for clinical doses to the planned target volume (PTV) and OARs were calculated. Results The mean DSC and 95HD values of the defined CTVs were (0.84 ± 0.05) and (5.04 ± 2.15) mm, respectively. The average delineation time was  〈  15 s for each patient's CT scan. The overall positive rates for clinicians A and B were 53.15% versus 46.85%, and 54.05% versus 45.95%, respectively ( P  〉  .05) when CTV outlines from CUNet were blindly chosen and compared with the ground truth (GT). Furthermore, 8 test patients were randomly chosen to design the predicted plan based on the autocontouring CTVs and OARs, demonstrating acceptable agreement with the clinical plan: average absolute dose differences in mean value of D2, D50, D98, Dmax, and Dmean for PTV were within 0.74%, and average absolute volume differences in mean value of V45 and V50 for OARs were within 3.4%. Conclusion Our results revealed that the CTVs and OARs for prostate cancer defined by CUNet were close to the GT. CUNet could halve the time spent by radiation oncologists in contouring, demonstrating the potential of the novel autocontouring method.

    Abstract: Anthocyanins, a group of flavonoids, are widely present in plants and determine the colors of the peels of stems, fruits, and flowers. In this study, we used UHPLC-ESI-MS to identify anthocyanins in the herbal plant Dendrobium officinale , which has been used for centuries in China. The results indicated that the total anthocyanin content in samples from Guangxi was the highest. Seven anthocyanins were identified, and the fragmentation pathways were proposed from D. officinale . Most of the identified anthocyanins were composed of cyanidin and sinapoyl groups. We also carried out that the sinapoyl group had active sites on breast cancer receptors by using Schrödinger. The relative levels of the 7 anthocyanins in the samples from the three locations were determined. Transcriptomic analysis was used to analyze the sinapoyl anthocyanin synthesis-related genes in plants, such as genes encoding UGTs and serine carboxypeptidase. We speculated that sinapoyl anthocyanin biosynthesis was associated with the activities of certain enzymes, including chalcone flavonone isomerase-like, hydroxycinnamoyltransferase 1, UGT-83A1, UGT-88B1 isoform X1, serine carboxypeptidase-like 18 isoform X3, and serine carboxypeptidase-like 18.

    Abstract: At present, the surface quality of Yuba skin is determined by sensory methods. In order to realize the intelligent classification detection of Yuba skin quality, this study designed a system that automatically determines the quality of Yuba skin surfaces based on image processing and support vector machine (SVM) approaches. Specifically, the system uses image preprocessing to extract the grayscale eigenvalues, gray level co-occurrence matrix (GLCM) eigenvalues, and gray level run length matrix (GLRLM) eigenvalues of the sample image and uses them as input values for a quality grading system. Through model evaluation of three classification models, the SVM classification model was selected according to the evaluation results, and different kernel functions were used in the model for sample training. Based on Matlab, the quality grading software of Yuba skin was developed and designed. Intelligent detection and grading were realized through the radial basis kernel function support vector machine (RBF-SVM) grading model. The best penalty factor (c = 3.50) and kernel parameter value (g = 0.98) were obtained through cross-validation. The accuracy of the model was 95.31% and 94.16% for the training and test sets, respectively. The grading accuracy of the RBF-SVM grading system was 93.56%, and the error was less than 5% compared with the traditional sensory method of grading; thus, the quality classification method based on the SVM classification system for Yuba skin is feasible and can be used for quality detection.

    Abstract: To examine the potential benefits and adverse events of neoadjuvant Chemoradiotherapy (CRT) versus upfront surgery in pancreatic cancer (PC) patients. Extensive librarian-led literature searches were conducted on PubMed, Web-of-Science, Scopus, Google Scholar, the Cochrane Central Library and Embase. The primary outcomes were resectability, adverse events, pathological and survival outcomes. Five studies, including 437 participants, were analyzed. Upfront surgery had a significantly higher resectability among PC patients than neoadjuvant CRT group (Odds ratio = -0.11, 95% CI = -0.19–0.02, P = 0.01). The neoadjuvant CRT group had a comparatively higher Ro resection rate (OR = 3.38, 95% CI = 2.03–5.62, P & lt; 0.01), fewer severe adverse events(OR = 0.56, 95% CI = 0.34–0.92, P = 0.02), lower positive LN rate(OR = 0.18, 95% CI = 0.11-0.31, P & lt; 0.01) and higher 2-year OS(OR = 1.60, 95% CI = 1.02-2.52, P = 0.04) among PC patients than control group. There was no significant difference between neoadjuvant CRT and upfront surgery among PC patients on postoperative complications(OR = 1.49, 95% CI = 0.86-2.57, P = 0.16), metastasis rate(OR = 1.32, 95% CI = 0.42-4.18, P = 0.64) and 1-year OS(OR = 1.30, 95% CI = 0.85-1.98, P = 0.22). This systematic review confirmed the status of neoadjuvant CRT in the PC treatment. The neoadjuvant CRT could increase the R0 resection rate, which was important to the survival and life quality of patients. The specific choice of various neoadjuvant CRT therapy needs to be further studied. Individualized neoadjuvant therapy should be suitable for each patient, and patients with PC are best managed by a multidisciplinary team.
