    Kurzfassung: The primary mode of transmission of Helicobacter pylori , a human pathogen carried by more than half the population worldwide, is still unresolved. Some epidemiological data suggest water as a possible transmission route. H. pylori in the environment transforms into a nonculturable, coccoid form, which frequently results in the failure to detect this bacterium in environmental samples by conventional culture techniques. To overcome limitations associated with culturing, molecular approaches based on DNA amplification by PCR have been developed and used for the detection of H. pylori in clinical and environmental samples. Our results showed the glmM gene as the most promising target for detection of H. pylori by PCR amplification. Under optimal amplification conditions, glmM -specific primers generated PCR-amplified products that were specific for H. pylori and some other Helicobacter species. Genome sequence analysis revealed the existence of a conserved region linked to a hypervariable region upstream of the 16S rRNA gene of H. pylori . Selective PCR primer sets targeting this sequence were evaluated for the specific detection of H. pylori . One primer set, Cluster2 and B1J99, were shown to be highly specific for H. pylori strains and did not produce any PCR products when other Helicobacter species and other bacterial species were analyzed. In tests with 32 strains of H. pylori , 6 strains of other Helicobacter species, 8 strains of Campylobacter jejuni , and 21 strains belonging to different genera, the primers for glmM were selective for the Helicobacter genus and the primers containing the region flanking the 16S rRNA gene were selective for H. pylori species only. The combination of two sensitive PCR-based methods, one targeting the glmM gene and the other targeting a hypervariable flanking region upstream of the 16S rRNA gene, are complementary to each other. Whereas the glmM -specific primers provide a rapid, sensitive presumptive assay for the presence of H. pylori and closely related Helicobacter spp., the primers for sequences flanking the 16S rRNA gene can confirm the presence of H. pylori and locate the potential source of this bacterium.

    Kurzfassung: The immunoreactivity of a commercial preparation of human chorionic gonadotrophin (HCG) was determined in a homologous double antibody radioimmunoassay for HCG using antisera to the β-subunit of the hormone. The immunoreactivity of the commercial HCG was found to be 2·2 ± 0·3 (mean ± 2 s.d. ) times the biological potency. Exclusion chromatography of the commercial HCG and then curve resolution of the elution profile derived from the radioimmunoassay revealed that on a molar basis, 21% of the immunoreactivity was attributable to β-HCG. The rate of clearance of this preparation of HCG from the plasma after intravenous administration was determined as a function of the dose administered to ten normal men (age 36–64 years). The doses ranged from 10 000 to 300 000 i.u. immunological potency. The rate of clearance decreased significantly ( r = 0·574, P 〈 0·05) with increasing doses of HCG from a mean of 786 ml/h at the lowest dose to a mean of 298 ml/h at the highest dose. The renal clearance of administered HCG also decreased with increasing doses; the mean renal clearance of the 10 000 i.u. dose was 3·6 times the mean renal clearance after administration of 200 000 i.u. When the accumulated urinary HCG was expressed as a percentage of the dose administered, 14·1% of the 10 000 i.u. dose and 9·8% of the higher doses accumulated in the urine, suggesting that non-renal clearance increased with increasing dose.

    Kurzfassung: The air occluded in ice sheets and glaciers has, in general, a younger age (defined as the time after its isolation from the atmosphere) than the surrounding ice matrix because snow is first transformed into open porous firn, in which the air can exchange with the atmosphere. Only at a certain depth (firn‐ice transition) the pores are pinched off and the air is definitely isolated from the atmosphere. The firn‐ice transition depth is at around 70 m under present climatic conditions at Summit, central Greenland. The air at this depth is roughly 10 years old due to diffusive mixing, whereas the ice is about 220 years old. This results in an age difference between the air and the ice of 210 years. This difference depends on temperature and accumulation rate and did thus not remain constant during the past. We used a dynamic firn densification model to calculate the firn‐ice transition depth and the age of the ice at this depth and an air diffusion model to determine the age of the air at the transition. Past temperatures and accumulation rates have been deduced from the δ 18 O record using time independent functions. We present the results of model calculations of two paleotemperature scenarios yielding a record of the age difference between the air and the ice for the Greenland Ice Core Project (GRIP) and the Greenland Ice Sheet Project Two (GISP2) ice cores for the last 100,000 years. During the Holocene, the age difference stayed rather stable around 200 years, while it reached values up to 1400 years during the last glaciation for the colder scenario. The model results are compared with age differences obtained independently by matching corresponding climate events in the methane and δ 18 O records assuming a very small phase lag between variations in the Greenland surface temperature and the atmospheric methane. The past firn‐ice transition depths are compared with diffusive column heights obtained from δ 15 N of N 2 measurements. The results of this study corroborate the large temperature change of 20 to 25 K from the coldest glacial to Holocene climate found by evaluating borehole temperature profiles.

    Kurzfassung: The 1992 Mw 7.3 Landers earthquake rupture propagated from the Johnson Valley fault to the Homestead Valley fault across Homestead Valley. Rupture was concentrated along the Kickapoo fault (a.k.a, Landers fault), a previously unrecognized north-south-striking, 5-km-long fault in Homestead Valley, and along small secondary fault traces east of the Kickapoo fault. To better understand the role of the Kickapoo fault in accommodating slip between the Johnson Valley fault and the Homestead Valley fault, we performed detailed aerial photograph interpretation and field mapping of alluvial deposits, geomorphology, and co-seismic surface rupture; evaluated existing water-well data; and conducted a gravity survey of the Homestead Valley area. These data show that the Kickapoo fault is a pre-1992 structure best interpreted as the northward continuation of the southern Johnson Valley fault. The Kickapoo fault merges northward with the northwest-striking Homestead Valley fault. Thus, we interpret that the Johnson Valley, Kickapoo, and Homestead Valley faults form an interlinked system. Large ruptures similar to the 1992 Landers rupture may have repeatedly propagated between the Johnson Valley and Homestead Valley faults along the Kickapoo fault. The Kickapoo-Johnson Valley fault intersection defines a 1-km-wide, left-restraining bend. Uplifted alluvial deposits and pressure ridges are common within the bend, indicating the bend is a long-lived contractional feature. Homestead Valley itself is underlain by relatively thin alluvial deposits over crystalline bedrock, showing that no significant subsidence has occurred during the late Quaternary. These observations are consistent with the accommodation of north-south right shear on the southern Johnson Valley and Kickapoo faults in Homestead Valley.

    Kurzfassung: The hormonal response of the anterior pituitary was studied in 10 normal males undergoing treadmill exercise testing, in 5 male patients undergoing diagnostic gastroscopy, and in 8 male patients undergoing elective surgery under general anaesthesia. Serum TSH was depressed below the baseline value at 2 and 3 h post-treadmill exercise, at 1, 2 and 3 h post-gastroscopy and from 10 min through 2 h post-surgery. Serum triiodothyronine was depressed below the baseline value at 10 min through 2 h post-surgery. Serum prolactin, growth hormone and cortisol were elevated by all three stressful procedures. Both gastroscopy and surgery resulted in an elevation of serum luteinizing hormone levels. There was no significant change in serum FSH levels in any of the three procedures. The post-stress depression in TSH levels could result from the suppressive effect at the hypothalamic-pituitary level of high serum levels of cortisol generated by the stress of the procedures.
