    Abstract: The site of callus initiation in sugarcane (Saccharum sp.) rolled young leaf and stem tip explants was histologically traced. It was found that the callus initials in the former were derived from the parenchyma adjacent to the primary phloem, whereas those in the latter arose from the primary phloem itself. The initial callus cells proliferated periclinally and consequently a nodulelike callus mass was formed after 40 days in culture. This finding is discussed in relation to its significance for in vitro mutation induction.
