    Abstract: A majority of fishermen in the coastal areas of Bumi Seribu Nyiur Melambai, East Kalimantan, Indonesia, are migrants from other islands of Indonesia. This paper presents results from a research of the fisherman community. Most of the fisherman came from lower-middle-class families. Both lower-and lower-middle-class fisherman have access to the development program, aids and others resources delivered by government and international corporate. This paper describes how most fisherman see and believe that Bumi Seribu Nyiur Melambai is an area of battle for many actors to upscale their economic and social mobility through accumulation of wealth and social status. Many fishermen express the ambiguity, uncertainty, and marginality of their social-economic status. The marginal status is caused by (i) Long Story of Unequal Social Structure; (ii) elite capture of programs and aids (iii) uncertainty and the loss of fishing area due to the presence of the company; (iv) Their technology are not able to resist the change of seasons. Data in this research are collected by qualitative and quantitative method, such as a participatory observation, in-depth interview, FGD, and survey.

    Abstract: A Community service program (PkM) carried out by the Department of Anthropology, Faculty of Cultural Sciences, Gadjah Mada University in 2018 took place in Pucung Hamlet, Wukirsari Village, Bantul, which aims to develop Pedukuhan Pucung as a tourism village through participatory methods. In the implementation of this PkM, the participatory method that is introduced is the PRA (participatory rural appraisal) that researchers could act as facilitators and could directly facilitate citizens to recognize and utilize participatory methods to develop a CAP (Community Action Plan) which would later become a tourist village development plan. At the end of the implementation of this program, the CAP has successfully compiled the development of a Pucung tourism village based on local culture and it was also agreed to create a tourism village development organization and to select several local facilitators. Further, the PkM team will still provide assistance to the sustainability and implementation of the CAP of the Pucung Tourism Village.--------------------------------------------------Kegiatan program pengabdian kepada masyarakat (PkM) yang dilakukan oleh Departemen Antropologi, Fakultas Ilmu Budaya, Universitas Gadjah Mada pada tahun 2018 ini mengambil lokasi di Pedukuhan Pucung, Desa Wukirsari, Bantul, yang terkait dengan pengembangan Pucung sebagai desa wisata secara partisipatoris. Dalam pelaksanaan PkM ini, metode partisipatoris yang dikenalkan adalah metode PRA sehingga peneliti dapat bertindak sebagai fasilitator dan langsung dapat memfasilitasi warga dalam mengenal dan memanfaatkan metode partisipatoris untuk menyusun CAP (Community Action Plan), yang nantinya akan menjadi program pengembangan desa wisata. Pada akhir pelaksanaan program ini telah berhasil disusun CAP pengembangan Desa Wisata Pucung yang berbasis budaya lokal dan juga disepakati untuk membentuk kepengurusan organisasi pengembangan desa wisata dan pemilihan beberapa fasilitator lokal. Dalam pengembangan selanjutnya, tim PkM masih akan melakukan pendampingan terhadap keberlanjutan dan pelaksanaan CAP pengembangan Desa Wisata Pucung.

    Abstract: The Indonesian Ministry of Environment and Forestry recently issued a 35-year permit-based social forestry, called Izin Pemanfaatan Hutan Perhutanan Sosial (IPHPS), which was implemented in forestlands managed by the State Forest Corporation (SFC). IPHPS is a unique scheme because social forestry permits were previously granted on forestland unencumbered with rights. It provides more secure tenure rights (long-term permits), greater decision-making authority, and improved profit-sharing arrangements compared with the SFC’s co-management model. However, IPHPS has not attracted widespread interest from local communities. This paper aims to identify and to analyse factors that explain local communities’ low interest in the policy. Results show that local communities have not been attracted by the scheme because it requires them to undertake substantial investments in reforestation and make several payments to the government beyond their means. This paper highlights the specific challenges related to access mechanisms and benefits to local communities from the granted rights. Lastly, local communities were prone to manipulative persuasion by the SFC to continue the co-management model.

    Abstract: Synedrella nodiflora (L.) Gaertn has taxonomically been the only species of genus Synedrella, which spreads over many tropical countries. In spite of its wide range of distribution, the genus remains monospecific. This leads to assumption of the very low genetic diversity among S. nodiflora populations worldwide. It may also be the case in Java Island, though rapid changes in ecosystem condition occurs. Here we report our study on S. nodiflora population genetics in Java Island using intergenic spacer (IGS) atpB –  rbcL as a molecular marker, since it has been well known as one of the most variable chloroplast genome regions in a wide range of plant species so far. As many as 58 individuals were collected randomly from ten different locations in the island. Based on IGS atpB – rbcL sequences of 860 bp length, only two haplotypes were observed. Both show only one polymorphic site (0.12%) and one transversion, where T is substituted by G at position 790, indicating that high connectivity among populations of S. nodiflora in Java Island is observed. This results in a low genetic differences among the populations, which at the same time provides a fact of nearly no variation among the IGS atpB – rbcL sequences.

    Abstract: Previous study on Synedrella nodiflora (L.) Gaertn populations in Java Island showed both very low haplotype and nucleotide diversity, and at the same time revealed high connectivity among the populations. Sumatra Island, which is like Java Island located in Sunda Shelf, has been subjected to relatively increasing human population and overexploitation of natural resources in a few last decades. This condition put the island of being vulnerable to terrestrial ecosystem changes that potentially influence the existing populations of S. nodiflora . Hence, this study aimed to assess genetic differences among S. nodiflora populations in Sumatra Island using intergenic spacer (IGS) atp B – rbc L. This molecular marker has been used in the population genetic study of some plant species. In this study we collected randomly 20 individuals from four different locations in Sumatra. The results showed, based on IGS atp B – rbc L sequences of 860 bp length, that only two haplotypes were found. One of them was the same haplotype mostly found in Java Island, and the other showed some base substitutions. Low genetic differences indicating high connectivity among populations of S. nodiflora in Sumatra Island is observed.

    Abstract: Penelitian ini bertujuan mengetahui kemampuan perkembangan awal embrio sapi in vitro menggunakan media tunggal untuk maturasi, fertilisasi, dan kultur berbahan dasar tissue culture medium (TCM) 199. Oosit sapi dikumpulkan dari rumah potong hewan dengan teknik aspirasi dan diklasifikasikan berdasarkan kekompakan sel kumulus dan sitoplasma yang homogen. Oosit dimaturasi pada medium TCM 199 yang disuplementasi dengan 10 IU/ml pregnant mare’s serum gonadotropin (PMSG), 10 IU/ml human chorionic gonadotropin (hCG), dan 10% fetal bovine serum (FBS), dilakukan selama 24 jam pada inkubator 5% CO2, 39 C. Fertilisasi dilakukan pada dua media yang berbeda yaitu media rutin fertilisasi dan media berbahan dasar TCM 199 dengan suplemen bovine serum albumin (BSA) dan heparin. Setelah fertilisasi, kumulus sel dihilangkan (denudasi), kemudian dikultur pada media TCM 199 yang disuplementasi dengan asam amino esensial dan non-esensial serta 10% FBS selama 3 hari. Hasil penelitian menunjukkan tingkat maturasi oosit pada sistem yang digunakan mampu mendukung 81,5% oosit mencapai tahap metafase II (M-II). Tingkat pembelahan embrio lebih tinggi pada media rutin dibandingkan dengan media TCM 199 yakni masing-masing 44,4 dan 23,2%. Jumlah embrio tahap 4-8 sel pada kedua perlakuan tidak berbeda nyata. Dapat disimpulkan media tunggal berbasis TCM dapat digunakan untuk produksi embrio in vitro.

    Abstract: Penelitian ini bertujuan isolasi dan purifikasi sel Leydig dengan menggunakan gradien Nycodenz untuk meningkatkan jumlah perolehan sel Leydig yang hidup setelah purifikasi dan kultur. Isolasi dan purifikasi sel Leydig dilakukan dengan perlakuan gradien Nycodenz 3 kolom (5, 10, dan 15% sebagai Nycodenz I) dan 5 kolom (4, 8, 10, 12, dan 15% sebagai Nycodenz II) serta gradien Percoll 5 kolom (21, 26, 34, 40 dan 60%) sebagai kontrol. Parameter yang diamati adalah konsentrasi, kemurnian, dan viabilitas sel Leydig setelah isolasi dan purifikasi serta kultur in vitro. Hasil yang diperoleh menunjukkan bahwa isolasi dan purifikasi sel Leydig dengan gradien Nycodenz (I dan II) secara nyata (P0,01) menghasilkan konsentrasi sel yang lebih rendah dibandingkan dengan gradien Percoll. Namun penggunaan gradien Nycodenz II c enderung menghasilkan kemurnian sel yang lebih tinggi (91,40%) dibandingkan dengan Nycodenz I (85,53%). Hasil tersebut tidak berbeda nyata dibandingkan dengan gradien Percoll (92,20%). Viabilitas sel Leydig pada semua perlakuan hampir sama yaitu 98%. Namun demikia n, setelah dikultur selama 3 hari, konsentrasi sel Leydig pada perlakuan Percoll (2,44x106 sel/ml) dan Nycodenz I (3,21x106 sel/ml) secara statistik (P0,05) lebih rendah dan dibandingkan dengan Nycodenz II (3,88x10 6 sel/ml), sedangkan viabilitas sel Leydig setelah dikultur pada gradien Nycodenz I (90,00%) dan Nycodenz II (91,17%) secara sangat nyata (P0,01) menghasilkan viabilitas yang lebih tinggi dibandingkan dengan gradien Percoll (82,30%). Berdasarkan hasil tersebut dapat disimpulkan bahwa penggunaan gradien Nycodenz II efektif untuk mempurifikasi sel Leydig dan setelah dikultur menghasilkan konsentrasi dan viabilitas sel yang lebih tinggi dibandingkan dengan gradien Percoll.

    Abstract: This study to do examine the effectiveness of using heparin and hypotaurine to improve the in vitro fertilization rate. In first stage, oocytes were matured in the maturation medium supplemented with 0.3 mg/mL L-carnitine to improve nuclear maturation. In second stage, matured oocytes were fertilized with semen incubated in fertilization medium supplemented with 0.1 IU/mL heparin and 10 µM hypotaurine. The final concentration of sperm used was 5x106 spermatozoa/mL, and its interaction with oocytes was carried out for 12 hours in 5% CO2 incubator at 38.5° C. The results showed that percentage of oocytes that reached metaphase II was significantly higher (P0.05) in media supplemented with 0.3 mg/mL L-carnitine compared to control (88.2±4.9% vs 79.8±5.1%). Furthermore, fertilization rate was characterized by formation of 2 PN was significantly higher (P0.05) in media supplemented with heparin and hypotaurine than control (82.6±2.0% vs 73.3±2.4%). In conclusion, L-carnitine supplementation at dose of 0.3 mg/mL on maturation media was effective in increasing percentage of matured oocytes. The combination of 0.1 IU/mL heparin and 10 µM hypotaurine in fertilization medium was also successful in elevating the percentage of fertilized oocytes.

    Abstract: Introduction: Sapphire bracket is the best aesthetic bracket with metal bracket-like mechanical properties. In conditions causing attachment lost between sapphire bracket and enamel, a rebonding procedure will needed. The shear bond strength of sapphire bracket decrease after the rebonding procedure. Sandblasting, hydrofluoric acid etching, and silanization as surface treatment are widely used to improve the bracket bond strength. The purpose of this study was to evaluate the effect of zirconia as sandblasting material towards the shear bond strength of reconditioned sapphire bracket. Methods: This study was an experimental laboratory with as much as 27 samples of sapphire bracket divided into 3 groups. Group I was rebonded with sandblasting zirconia and silane, group II with hydrofluoric acid etching and silane, and group III with new bracket bonding. The attachment strength measurement indicator was the shear bond strength test and the SEM analysis. Results: The shear bond strength test showed the average value for each group were 12.97 MPa, 9.26 MPa, and 10.58 MPa, consecutively. The test results were then analysed using a one way ANOVA and LSD (p 〈 0.05), which showed that significant difference only found in the comparison between group I with group II, but not in the comparison between group I with group III. The SEM analysis result showed that sandblasted surface has homogenous microporosities in high quantities. Conclusion: Zirconia as sandblasting material was effective in increasing the shear bond strength of reconditioned sapphire bracket.
