    Abstract: To solve the problem of soil degradation in coastal saline–alkali land, three different types of biochar (rice straw biochar, magnetic biochar, and humic acid–magnetic biochar) were prepared to remedy the saline–alkali soil under different mixing ratios. The effects of biochar on the growth of crops in saline–alkali soil were explored through a pot experiment on Chinese cabbage. The experimental results showed that the soil leaching treatment combined with humic acid–magnetic biochar could effectively repair the coastal saline–alkali soil. After adding 5% humic acid–magnetic biochar, the content of soil organic matter was 33.95 g/kg, the water content was 13.85%, and the contents of available phosphorus and available potassium were 9.43 mg/kg and 29.51 mg/kg. After adding 5% humic acid–magnetic biochar, the plant height of Chinese cabbage was 9.16 ± 0.19 cm, and the plant germination rate reached 83.33 ± 5.54%. The incorporation of biochar could effectively increase the chlorophyll content and soluble protein content of pakchoi and reduce the soluble sugar content of pakchoi. The study analyzed the effect of different modified biochar on saline–alkali land restoration and crop growth and explored the action rule of hydrochloric acid magnetic biochar on saline–alkali land restoration, which has important practical value for improving coastal saline–alkali land.

    Abstract: Fibro-adipogenic progenitor cells (FAPs) are a population of stem cells in skeletal muscle that play multiple roles in muscle repair and regeneration through their complex secretome; however, it is not well understood how the FAP secretome is altered with muscle disuse atrophy. Previous work suggests that the inflammatory cytokine IL-1β is increased in FAPs with disuse and denervation. Inflammasome activation and IL-1β secretion are also known to stimulate the release of extracellular vesicles (EVs). Here, we examined the microRNA (miRNA) cargo of FAP-derived, platelet-derived growth factor receptor A (PDGFRα + ) EVs from hindlimb muscles of wild-type and IL-1β KO mice after 14 days of single-hindlimb immobilization. Hindlimb muscles were isolated from mice following the immobilization period, and PDGFRα + extracellular vesicles were isolated using size-exclusion chromatography and immunoprecipitation. Microarrays were performed to detect changes in miRNAs with unloading and IL-1β deficiency. Results indicate that the PDGFRα + , FAP-derived EVs show a significant increase in miRNAs, such as miR-let-7c, miR-let-7b, miR-181a, and miR-124. These miRNAs have previously been demonstrated to play important roles in cellular senescence and muscle atrophy. Furthermore, the expression of these same miRNAs was not significantly altered in FAP-derived EVs isolated from the immobilized IL-1β KO. These data suggest that disuse-related activation of IL-1β can mediate the miRNA cargo of FAP-derived EVs, contributing directly to the release of senescence- and atrophy-related miRNAs. Therapies targeting FAPs in settings associated with muscle disuse atrophy may therefore have the potential to preserve muscle function and enhance muscle recovery.

    Abstract: Soybean seeds contain substantial triacylglycerols and fatty acids that are prone to oxidation during storage, contributing to the dramatic deterioration of seed vigor. This study reports an ultrasonic waves treatment (UWT), which is a physical method capable of promoting the germination ability of the aged soybean seeds by regulating the antioxidant defense and gluconeogenesis. Germination test revealed that UWT significantly increased the germination rate and seedlings’ establishment of the soybean seeds stored for 12 months, although insignificantly impacting the vigor of fresh (stored for 1 month) and short-term stored (for 6 months) seeds. Further biochemical analysis revealed that UWT decreased the hydrogen peroxide (H 2 O 2 ), O 2 ⋅ – , and malondialdehyde contents in the aged soybean seeds during early germination. Consistently, UWT prominently elevated the activities of superoxide dismutase, catalase, and acetaldehyde dehydrogenase, and also the corresponding gene expressions. Besides, the soluble sugar content of UWT was significantly higher than that of the untreated aged seeds. Analysis of enzyme activity showed UWT significantly upregulated the activities of several key enzymes in gluconeogenesis and the transcription levels of corresponding genes. Moreover, UWT enhanced the invertase activity within aged seeds, which was responsible for catalyzing sucrose hydrolysis for forming glucose and fructose. In summary, UWT improved germination and seedlings establishment of aged soybean seeds by regulating antioxidant defense and gluconeogenesis. This study expands the application of ultrasonication in agricultural production and further clarifies the physiological and molecular mechanisms of the aged seed germination, aiming to provide theoretical and practical guidance for seed quality and safety.

    Abstract: This paper presents an overlapping decentralized guaranteed cost hybrid control method for adjacent buildings with uncertain parameters, by combining the guaranteed cost control algorithm with the overlapping decentralized control strategy. The passive dampers are used as link members between the two parallel buildings, and the active control devices are installed between two consecutive floors in two adjacent buildings. The passive coupling dampers modulate the relative responses between the two buildings, and the active control devices modulate the interstory responses of each building. Based on the inclusion principle, a large-scale structure is divided into a set of paired substructures with common parts first. Then, the controller of each pair of substructures is designed by using the guaranteed cost algorithm. After that, the controller of the original system is formed by using the contraction principle. Consequently, the proposed approach is used to prevent pounding damage and achieve the best results in earthquake response reduction of uncertain adjacent buildings when compared with the calculation results obtained by the centralized control strategy. Furthermore, the stability and reliability of the control system are promoted by adopting the overlapping decentralized control strategy.

    Abstract: Since raw data recorded by patch-clamp systems are always stored in binary format, electrophysiologists may experience difficulties with patch clamp data preprocessing especially when they want to analyze by custom-designed algorithms. In this study, we present desktop software, called PCDReader, which could be an effective and convenient solution for patch clamp data preprocessing for daily laboratory use. We designed a novel class module, called clsPulseData, to directly read the raw data along with the parameters recorded from HEKA instruments without any other program support. By a graphical user interface, raw binary data files can be converted into several kinds of ASCII text files for further analysis, with several preprocessing options. And the parameters can also be viewed, modified and exported into ASCII files by a user-friendly Explorer style window. The real-time data loading technique and optimized memory management programming makes PCDReader a fast and efficient tool. The compiled software along with the source code of the clsPulseData class module is freely available to academic and nonprofit users.

    Abstract: Acidic tea (Camellia sinensis) plantation soil usually suffers from magnesium (Mg) deficiency, and as such, application of fertilizer containing Mg can substantially increase tea quality by enhancing the accumulation of nitrogen (N)-containing chemicals such as amino acids in young tea shoots. However, the molecular mechanisms underlying the promoting effects of Mg on N assimilation in tea plants remain unclear. Here, both hydroponic and field experiments were conducted to analyze N, Mg, metabolite contents, and gene expression patterns in tea plants. We found that N and amino acids accumulated in tea plant roots under Mg deficiency, while metabolism of N was enhanced by Mg supplementation, especially under a low N fertilizer regime. 15N tracing experiments demonstrated that assimilation of N was induced in tea roots following Mg application. Furthermore, weighted gene correlation network analysis (WGCNA) analysis of RNA-seq data suggested that genes encoding glutamine synthetase isozymes (CsGSs), key enzymes regulating N assimilation, were markedly regulated by Mg treatment. Overexpression of CsGS1.1 in Arabidopsis (Arabidopsis thaliana) resulted in a more tolerant phenotype under Mg deficiency and increased N assimilation. These results validate our suggestion that Mg transcriptionally regulates CsGS1.1 during the enhanced assimilation of N in tea plant. Moreover, results of a field experiment demonstrated that high Mg and low N had positive effects on tea quality. This study deepens our understanding of the molecular mechanisms underlying the interactive effects of Mg and N in tea plants while also providing both genetic and agronomic tools for future improvement of tea production.

    Abstract: The protooncogene MYC encodes the c-Myc transcription factor that regulates cell growth, cell proliferation, cell cycle, and apoptosis. Although deregulation of MYC contributes to tumorigenesis, it is still unclear what direct Myc-induced transcriptomes promote cell transformation. Here we provide a snapshot of genome-wide, unbiased characterization of direct Myc binding targets in a model of human B lymphoid tumor using ChIP coupled with pair-end ditag sequencing analysis (ChIP-PET). Myc potentially occupies 〉 4,000 genomic loci with the majority near proximal promoter regions associated frequently with CpG islands. Using gene expression profiles with ChIP-PET, we identified 668 direct Myc-regulated gene targets, including 48 transcription factors, indicating that Myc is a central transcriptional hub in growth and proliferation control. This first global genomic view of Myc binding sites yields insights of transcriptional circuitries and cis regulatory modules involving Myc and provides a substantial framework for our understanding of mechanisms of Myc-induced tumorigenesis.
