    Abstract: OBJECTIVES: In colorectal cancer, to describe the cytophotometric expression of the CD-34 and Factor VIII; to evaluate the degree of correlation between them; and to compare the CD-34 and Factor VIII expressions in relationship to label index and optical density. METHODS: Seventeen cases of colorectal adenocarcinoma recovered from paraffin-embedded archival tissue and confirmed by hematoxilin-eosin staining, were submitted to streptavidin-biotin-peroxidase immunohistochemical method. In this process was used the tumor markers CD-34 and Factor VIII. The obtained slides were analysed using the SAMBA 4000® system with Immuno® software. The results were evaluated into two parameters: label index and optical density, and expressed by averages, medians, minimum values, maximum values, and standard deviation values. The normality condition of the quantitative variables was investigated by using the Shapiro-Wilks test. In order to evaluate the degree of association between the expressions of the markers, Pearson's Correlation Coefficient or Spearman's Correlation Coefficient were applied. To evaluate the comparison degree of the markers expression, Student's t test or Wilcoxon's no parametric test were used. RESULTS: For the CD-34 there was no data normality for the label index and there was normality in the optical density. For the Factor VIII, there was data normality for the label index and for the optical density. CONCLUSION: When the expressions of CD-34 and Factor VIII markers were correlated, there was no difference between them in relationship to the average label index and average optical density. When the expressions of the CD-34 and Factor VIII were compared, there was no correlation between the two variables.

    Abstract: OBJCTIVE: 1. To evaluate the percentage of caspase-3 and CD-34 expression on adenocarcinoma; 2. to quantify caspase-3 and CD-34 in tumor cells; 3. to verify the relationship between biomarkers and its malignancy; 4. to correlate biomarkers themselves. METHODS: Thirty-eight human malignant prostate specimens, Gleason's score, were immunohistochemically stained for caspase-3 and CD-34 protein. Quantification was done under Samba 4000 Immuno System reading, yielding two variables: label index and optical density. Statistical analyses were based on cross-methods involving univariate and bivariate as well as correlation factors among independent variables. RESULTS: Immunostaining was revealed in 25 plates for caspase-3 and 34 for CD-34. Caspase-3 expression for label index was over 50 in 76%, while for optical density was below 50 in 96%. CD-34 expression demonstrated label index over 50 in 59% and optical density below 50 in 56%. Correlation among expression and severity did not demonstrate to be statistically significant. There was no correlation between protein expression and Gleason's score. CONCLUSION: 1. Caspase-3 and CD-34 were present respectively in 73.5% and 100% of samples; 2. caspase-3 and CD-34 showed high expression regarding label index and low expression in optical density; 3. there was no statistical significance among expressions and tumor severity according to Gleason's score; 4. no significant correlation could be set between the biomarkers themselves.

    Abstract: OBJECTIVE: To describe quantitative cytophotometric expression of the marker caspase-3 in colloid goiter and in papillary carcinoma of the thyroid, comparing the immunoexpression between them. METHODS: An immunohistochemistry study has been made on the protein caspase-3 in 17 of paraffin blocks of papillary carcinoma of the thyroid and 20 cases of colloid goiter, using SAMBA 4000 ® (System of Microscopic Analysis of Automatic Search), aiming analysis of the two variables: marker index and optical density. RESULTS: Statistic analysis indicated a significant difference for marker index between colloid goiter and papillary carcinoma, being this difference larger in the carcinoma. It hasn't been found significant difference in optical density. For colloid goiter, the estimated correlation coefficient between the marker index and optical density was 0,72, indicating the rejection of the null hypothesis (p 〈 0,001), affirming that positive and significant association exists between them. For the papillary carcinoma of the thyroid, the same correlation was 0,34 and so, it is not possible to affirm that there's an association between them. CONCLUSION: For colloid goiter there is a positive and significant association between the two variables - marker index and optical density -, while for the papillary carcinoma, this is not proved. The quantitative analysis for caspase-3 demonstrates that apoptosis is larger in the papillary carcinoma of the thyroid than in colloid goiter.

    Abstract: OBJETIVO: Investigar os resultados da cicatrização de feridas cirúrgicas em dorso de ratos, utilizando curativos de prata nanocristalina e iônica. MÉTODOS: Sessenta ratos Wistar foram submetidos à feridas cirúrgicas com punch de 8mm de diâmetro. Foram confeccionadas duas lesões diametralmente opostas nos animais dos grupos prata nanocristalina (PN) e controle água destilada (AD). Na lesão do lado direito foi utilizado curativo com prata nanocristalina e na do lado esquerdo curativo com água destilada. No outro grupo de 30 ratos, apenas foi realizada uma lesão com o punch no lado esquerdo e curativo com prata iônica. Os grupos foram divididos em subgrupos conforme o dia da morte (7º, 14º e 21º dias), o que caracterizou a existência de três subgrupos, nos quais foram tomadas as medidas do centro das lesões para estudar macroscopicamente a contração da ferida Microscopicamente foi utilizada a coloração H & E, através da qual foi observado o processo inflamatório e a neovascularização. Com a coloração Tricômio de Masson, foi estudada a fibrose. RESULTADOS: Macroscopicamente apenas o subgrupo 21 dias apresentou significância estatística entre os grupos AD e prata iônica (PI), porém quando comparados os dias de avaliação, dois a dois, dentro de cada tratamento, todos os subgrupos apresentaram significância estatística (p 〈 0,05). A variável intensidade da inflamação apresentou-se de forma acentuada no subgrupo sete dias em 90% dos casos, entre os grupos AD e PI. Na variável neovascularização houve significância estatística entre os grupos PN e AD no subgrupo sete dias. A variável fibrose não apresentou significância estatística nos subgrupos estudados. CONCLUSÕES: Em relação à contração da ferida, os grupos PN e PI apresentaram resultados superiores ao grupo AD. Com relação à análise histológica, na coloração H & E observou-se que houve presença de processo inflamatório nos grupos estudados sendo que, ao final, o grupo controle (AD) no 7º dia mostrou-se superior aos grupos PN e PI. Com relação à variável fibrose, não houve diferenças entre os grupos.

    Abstract: RESUMO Racional: Tumores neuroendócrinos gastrointestinais são raros geralmente apresentados como tumores subepiteliais ou polipoides. O diagnóstico preciso e a indicação do tipo de ressecção ainda são desafiadores. Objetivo: Determinar a eficácia da ecoendoscopia em determinar a profundidade das lesões (T) identificadas pela endoscopia com objetivo de avaliar indicação cirúrgica e/ou endoscópica, e avaliar os resultados da remoção endoscópica em seguimento em médio prazo. Métodos: Foram incluídos 27 pacientes todos submetidos à ecoendoscopia para estadiamento tumoral TN e à avaliação de possível ressecção endoscópica. Os parâmetros estudados foram: tamanho da lesão, camada de origem, profundidade do acometimento e adenopatias perilesionais identificadas. Os critérios de inclusão para ressecção endoscópica foram: 1) risco cirúrgico elevado; 2) aqueles com TNE 〈 2 cm; 3) ausência de comprometimento da muscular própria; e 4) ausência de adenopatias perilesionais na ecoendoscopia e em outros sem metástases à distância. Os critérios de exclusão foram TNE 〉 2 cm; os com infiltração da muscular própria; com adenopatias perilesionais e metástases à distância. As técnicas utilizadas foram: ressecção com alça de polipectomia; mucosectomia com injeção de solução salina; e mucosectomia após a ligadura com banda elástica. O estudo anatomopatológico dos espécimes incluiu avaliação das margens e imunoistoquímica (cromogranina, sinaptofisina e Ki 67) para caracterizar o tumor. O seguimento foi feito com 1, 6 e 12 meses. Resultados: Ressecções com alça de polipectomia foram realizadas em 15 pacientes; mucosectomia em cinco; mucosectomia e ligadura com banda elástica em três e os quatro restantes foram encaminhados para cirurgia. O anatomopatológico dos espécimes e as análises imunoistoquímicas mostraram cromogranina e sinaptofisina positivas, enquanto que o Ki 67 foi menor que 5% dentre todos os casos. O seguimento em médio prazo revelou três recidivas. A média de tamanho dos tumores no estômago foi de 7,6 mm e no duodeno 7,2 mm. As lesões bem demarcadas, hipoecóicas, homogêneas ocorreram em 75%; da camada mucosa em 80%; e da mucosa profunda e submucosa em 70%. Conclusões: A ecoendoscopia mostrou ser bom método para o estudo de lesões subepiteliais podendo identificar a camada acometida pela neoplasia, grau de invasão, ecogeneicidade, heterogeneidade, tamanho da lesão e acometimento linfonodal perilesional e melhor indicar a opção de tratamento.

    Abstract: BACKGROUND: The esophageal squamous cell carcinoma treatment strategy is still based on the tumor staging, where tumor histopathologic charac-teristics are the major determinants. In parallel, studies have been developed in order to better understand the tumor biology using immunohistochemical meth-ods with manual quantification evaluating the proliferative and apoptotic activi-ties of the cells. The disadvantages related to the manual method rose the de-velopment of computerized ways to do the image analysis. OBJETIVES: To verify the expressions of the markers Ki-67 (proliferative) and Caspase-3 (apoptotic) and to correlate them with the clinic and pathologic characteristics of the tumor. METHODS: Twenty-nine paraffin embedded blocks were studied, each one con-taining tissue samples from patients with esophageal squamous cell carcinoma submitted to esophagectomies. The clinic and pathological data were obtained from histopathologic informations and from medical records. The slides were prepared following the routine immunohistochemical method until the point to utilize the specific antibodies (MIB-1 and CPP32). Positive quantification of the immunoreactivity to the proteins Ki-67 and Caspase-3 was performed by the software for computerized image analysis SAMBA (Systeme d' Analyse Micro-photometrique a Balayage Automatique). Statistical analysis was done having P 〈 0.05 considered significant. RESULTS: There was predominance of males (82.7%); age over 50 years; moderately differentiated tumors (68.98%); tumor stage III (72.42%); diameter of lesions 〉 3cm; and lesions located in the lower third of the organ. The mean score indexes found were 62.05% for Ki-67 and 86.06% for Caspase-3 and there was no correlation with the clinic or pathologi-cal characteristics as gender, age and tumor staging. There was significant dif-ference of Ki-67 expression among the histological grades (P=0.047) and corre-lation between the evaluated indexes (r=0.41 and P=0.032). CONCLUSION: The protein expressions were high and the Caspase-3 protein activity was higher than the Ki-67, without correlation with clinic or pathological characteristics as gender, age, tumor staging, grade of lesion depth and lymph node invasion.

    Abstract: BACKGROUND: Colorectal cancer is one of the most common causes of death for malignant neoplasm in the Occidental World, corresponding colorectal adenocarcinoma to represent more than 90% of malignant tumors in large bowel. Caspase-3 cysteine protease, one of the most important enzymes related in the apoptosis process, may be used as a tumor marker. AIM: To identify and quantify Caspase-3 expression in colorectal adenocarcinoma samples and to correlate the marker expression using an image computerized analysis system. METHOD: Caspase-3 expressions were studied by immunohistochemistry in 55 colorectal adenocarcinoma sampled-tissues, prepared in paraffin blocks. SAMBA 4000 System was used for the immunostained tissues analysis. Microscopic evaluation was performed by the IMUNNO 4.00 software, which analyzed 8 to 10 fields in order to determine two parameters: Labeling Index (LI), indicating the percentage of cells specifically stained by immunohistochemistry, and Mean Optical Density (MOD), representing enhance intensity, measured by positive pixels. For the statistical analysis, t-Student test and Pearson coefficient verified the possible correlation concerning LI and MOD. RESULTS: All samples presented positivity for Caspase-3 expression (LI 85,24% ± 8,71% positive cells). Caspase-3 expression revealed statistically significance correlation between LI and MOD (P = 0,013). CONCLUSIONS: The intensity of Caspase-3 expression and the number of positive cells expressing such a marker showed a positive and significant correlation.

    Abstract: OBJETIVO: Estudar a lesão provocada pela secção e pela clipagem no nervo simpático cervical de coelhos, avaliando-se a macroscopia, microscopia e quantificando-se o colágeno nas lesões. MÉTODOS: Foram utilizados 20 coelhos, distribuídos em dois grupos de dez, sendo nos animais do grupo 1 (secção) realizada secção do nervo simpático cervical direito, enquanto nos do grupo 2 (clipagem) realizada a clipagem desse nervo. Todos os coelhos foram induzidos à morte no sétimo dia de pós-operatório. As variáveis macroscópicas avaliadas foram: presença de lesão de descontinuidade do nervo, aspecto do clipe, presença de infecção e de aderências ao redor do nervo. A microscopia com hematoxilina-eosina foi feita para avaliar as fases, o grau do processo inflamatório e a presença de necrose; a coloração de Picrosirius red F3BA quantificou o colágeno. Utilizou-se o teste de Mann-Whitney nas comparações dos colágenos tipo I e tipo III entre os grupos. As variáveis macroscópicas, o grau do processo inflamatório e presença de necrose foram analisadas pelo teste de Fisher. RESULTADOS: Não houve lesão de descontinuidade do nervo no grupo clipagem, assim como o clipe encontrou-se fechado em todos os animais desse grupo. A presença de aderências intensas foi significativamente maior no grupo clipagem (p 〈 0,05). Não houve diferença significativa quanto às demais variáveis analisadas macroscopicamente. Não houve diferença significativa entre os grupos quanto ao tipo de processo inflamatório e sua intensidade, assim como quanto à presença de necrose e ao depósito de colágeno nos nervos. CONCLUSÕES: Na avaliação macroscópica, a secção causou lesão de descontinuidade, o que não ocorreu na clipagem; não houve desenvolvimento de infecção local; a clipagem do nervo simpático cervical foi associada à presença de maior quantidade de aderências em relação à secção. Na avaliação microscópica reconheceu-se não haver diferença no tipo e na intensidade do processo inflamatório entre os grupos; ocorreu predomínio de processo inflamatório crônico e acentuado; as necroses ocorreram igualmente em ambos os grupos; houve predomínio de depósito de colágeno tipo I em relação ao tipo III em ambos os grupos.

    Abstract: INTRODUCTION: The suture of tissues and their healing process is one of the basic fundamentals of surgery, and the research on substances which could improve the process is an ongoing challenge. The use of plants has been tested but till nowadays without scientific demonstration. PURPOSE: To compare the macroscopic and histological alterations done by the use of intraperitoneal Jatropha gossypiifolia L. gross extract, in the healing process of sutures performed on the bladder of rats. METHODS: Forty adult, male, Wister rats were distributed into two animal groups. One centimeter longitudinal incision on the bladder ventral wall, and single plane synthesis with separated polyglactine 910 5-0 (Ethicon) stitches was done in all the animals. In the control group an intraperitoneal cavity instillation of distilled water at a ratio of 1ml/kg/weight, and a 1ml/kg/weight of Jatropha gossypiifolia L. extract for the Jatropha group was done representing 200 mg of the substance. Each group was subdivided into two with 10 animals in each, being submitted to euthanasia on 3th and 7th post-operative days. Comparative histological, macroscopic and statistical analysis were undertaken between the subgroups. RESULTS: Statistical significant difference was observed in the acute inflammation changes, vascular neoformation and bonding on day 3; the first one was greater in the control group, and the latter two greater in the Jatropha group; acute inflammation variables and fibroblastic proliferation presented to be more intense on day 7, with statistical significance favoring the control group. CONCLUSION: No favorable healing effect was observed with the administration of single intraperitoneal dose of Jatropha gossypiifolia L. gross extract on the sutures of bladder in rats.

    Abstract: OBJECTIVE: Describe, correlate and compare the expression of the tumor markers CD 34 (angiogenesis) and caspase-3 (apoptosis) in invasive breast adenocarcinoma, through image cytometry with the system SAMBA4000®. METHODS: Twenty-two cases of invasive breast adenocarcinoma from paraffin-embedded archival tissue, and after specific prepare, fifteen cases presented a satisfactory lecture with SAMBA4000® and could, finally, be evaluated by the software IMMUNO® (n = 15). The parameters analysed were the label index - in percentage, indicating the marked surface - and the optical density, in pixels - indicator of the marker intensity. The results were tabulated and expressed in averages, mediums, minimum and maximum values. The statistic analysis was realized by the Shapiro-Wilkins, Student test, Pearson's and Spearman's correlation, with statistic significance for values from p 〈 0,05. RESULTS: There was no data normality for the label index CD34 (p= 0,019), there was normality in the analysis of the optical density of both markers and label index of the marker Caspase-3. There was no difference relating to the average of the index marker and the optical density when they were compared. CONCLUSIONS: There was a tendency to correlate the label index and the optical density of the tumor marker caspase-3, the same did not occur with the tumor marker CD 34. Other analysis did not show any correlation between the two studied markers. Other studies involving theses two cell processes are needed to extend the knowledge of the cancer biomolecular mechanic and to permit new diagnostic and therapeutic strategies.
