Abstract:
Pour savoir si la fixation de gastrine et de sécrétine est spécifique à certains organes (le rein surtout) ou s'il s'agit d'un phénomène non spécifique survenant dans tous les lits capillaires, nous avons mesuré la fixation de gastrine et de sécrétine, endogènes et exogènes, chez 32 chiens anesthésiés. Chez 6 chiens, l'irrigation antrale avec une solution d'acéthylcholine à 0.5% provoque une brusque augmentation de la gastrinémie mesurée dans le sang aortique. Les concentrations dans le sang des veines rénale, jugulaire et fémorale sont toutes significativement plus faibles et la fixation globale de gastrine a été de 48% pour le rein, de 43% pour le membre inférieur et de 36% pour la tête (ces différences ne sont pas statistiquement significatives). La perfusion de gastrine humaine synthétique chez 6 chiens, à deux doses (0.4 et 0.8 μ g/kg/hr) entraîne une élévation des taux de gastrine dans le sang aortique, avec des valeurs significativement plus faibles dans le sang des veines rénale, fémorale et jugulaire. Si l'on calcule les fixations globales, le rein fixe 42%, le membre inférieur 35%, et la tête 30% de la gastrine à la dose de 0.8 μ g/kg/hr. Pour des doses plus faibles, les pourcentages d'extraction sont similaires. Chez 5 chiens, une libération endogène de secrétine a été produite par irrigation du duodénum avec de l'HCl 0.1 N. Dans le sang aortique, la concentration de secrétine passe d'une valeur basale d'environ 60 pg/ml à 217+24 pg/ml á 45 minutes. Les concentrations de secrétine sont nettement plus basses dans le sang des veines rénale, fémorale et jugulaire: le rein fixe 39%, le membre inférieur 29% et la tête 32% de la quantité totale de secrétine endogène (ces différences ne sont pas significatives). Pendant une perfusion de secrétine exogène à la dose de 0.4 U/kg/hr, le taux moyen dans le sang aortique varie entre 340 et 420 pg/ml, alors que, dans le sang des veines rénale, fémorale et jugulaire, il est entre 220 et 295 pg/ml. Lorsqu'on calcule les quantités totales de secrétine, on trouve que le rein fixe 42%, le membre inférieur 43% et la tête 43%. Pour des doses de secrétine de 1 U/kg/hr, la fixation est de 46% pour le rein, de 31% pour le membre inférieur et de 26% pour la tête. L'extraction de gastrine et de secrétine se fait donc dans tous les lits capillaires et semble bien être un processus non spécifique.
