    Abstract: Background: Seed recalcitrance is a major problem associated with many tropical plants, limiting their natural regeneration. Vateria indica L. is a vulnerable and endemic tree species in South-Western Ghats of India, which is also recalcitrant. ABA and gibberellins are the most important plant hormones required for seed germination. It is the balance between ABA and GA which is responsible for desiccation tolerance in orthodox seeds. Exogenous hormones pretreatment has been also reported to influence seed germination. But such studies had been sparsely done in the case of recalcitrant seeds. This study aims to find out whether GA/ABA antagonism in recalcitrant plants is operating in the same way, like that in the orthodox seeds.Methods: The effect of the exogenous pre-soaking application of phytohormones viz. GA3 and ABA individually as well as their combinations on seed germination and growth of Vateria indica L. were carried out in the present work. The seeds were collected from April to July 2018 and the experiment was designed at Post Graduate and Research Department of Botany, Government College for Women, Thiruvananthapuram. When different concentrations of each phytohormone were externally given to the seeds, ABA reduced the germination and growth in almost all the concentrations. But GA3 gave better results. When combinations of GA3 and ABA were used, germination was poor in the sample where ABA was more than GA3, But in samples with same concentrations of both the hormones and with more GA3 gave better results. Result: This study clearly showed that GA3 when given externally along with ABA, might have affected the endogenous ABA in this recalcitrant seed and suppressed its retarding effect. Thus ABA/GA antagonism is working out, here, in the same way as in orthodox seeds. Since the germination of recalcitrant seeds is a less investigated area, the present study will form a basis and a lot more for further such studies.

    Abstract: Thattekad bird sanctuary, located in the Western Ghats of Kerala, India, which hosts an unexplored microbial community, is selected for the present investigation. Microbes play a major role in mineral recycling and nutrient absorption by the flora and fauna in the habitat. Various bacterial extracellular enzymes facilitate all these activities. The increasing demand for microbial enzymes in favor of green technology encouraged us to focus on exoenzyme profiling of bacterial isolates from forest soil samples. The present study is aimed at the screening and identification of exoenzyme producing soil bacterial strains isolated from evergreen forests and moist deciduous forests of Thattekad bird sanctuary. In this study, only multienzyme producing bacteria were selected for detailed analysis because such bacteria are highly relevant in multi-enzyme dependent processes such as biowaste degradation. We screened for nine hydrolytic exoenzymes namely, amylase, cellulase, ligninase, pectinase, xylanase, caseinase, gelatinase, esterase and lipase, and identified 79 multienzyme-producing bacterial strains, mostly belonging to phylum Firmicutes and Proteobacteria. Firmicutes from evergreen forests and moist deciduous forests produced a greater number of enzymes compared to Proteobacteria. Also, bacterial strains isolated from evergreen forest soil produced more enzymes compared to moist deciduous forest. Bacillus amyloliquefaciens strain TBS040 isolated from moist deciduous forest soil was found to produce all the nine enzymes screened. Enzymatic hydrolysis of biowaste using cell free crude enzyme extract from Bacillus velezensis strain TBS064 resulted in enhanced bioethanol production. These findings highlight the importance of screening unexplored habitats for the identification of novel strains, which can contribute to the future of green technology.

    Abstract: Introduction: Postural alignment is commonly assessed, and the abnormal postures are associated with musculoskeletal disorders. There are different methods for screening spinal column anomalies. The flexible ruler provides a safe, portable, inexpensive and non-invasive way to measure the spinal curvature in clinical and larger population. Aim: To determine the intra-rater and inter-rater reliability of flexible ruler in measuring lumbar lordosis among children. Materials and Methods: In this cross-sectional study 81 school children that fulfilled the criteria were recruited with age ranging from 9 to 15 years. Youdas method in standing position was adopted. By using flexible ruler, measurement was taken and without altering the shape of the lumbar curve it was traced in a graph sheet. By using straight ruler, line L and H were drawn by following deep point method. Lumbar lordosis angle was calculated by using the formula, θ=4Arctan 2H/L. Intra-rater reliability: Four measurements were taken by evaluator 1 with a gap of one minute between first two measurements and 3 minutes rest then other two measurements were taken with a gap of one minute between each. The average of first two and the last two measurements were considered as measurements 1 and 2. Inter rater reliability: Two measurements were taken by evaluator 2 by adapting the same procedure. The averages of two measurements were considered as measurement for evaluator 2. This value was correlated with the value of measurement 1 of evaluator 1. The data was analysed using Intra Class Correlation coefficient (ICC) test. Results: The results showed high intra-rater reliability (ICC-0.991; p 〈 0.001) for lumbar lordosis angle by flexible ruler and poor inter-rater reliability (ICC-0.241; p=0.11) for lumbar lordosis angle by flexible ruler. Conclusion: The flexible ruler has shown only intra-rater reliability and additional studies are required to prove inter-rater reliability in measuring lumbar lordosis among children.
